Adeno-associated virus(AAV)is an essential.instrument in the neuroscientist’s tool.it,which al.ows del.very of DNA to provide l.bel.ng with fl.orescent proteins or genetic instructions to regul.te gene *** the fiel. of neural.regeneration,the transduction of neurons enabl.s the observation and regul.tion of axon growth and regeneration,and in the future wil. l.kel. be a mechanism for del.vering mol.cul.r therapies to promote sprouting and regeneration after central.nervous system *** formul.tions of AAV preparations permit efficient viral.transduction under physiol.gic conditions,but an improved understanding of the mechanistic l.mitations of AAV transduction may facil.tate production of more resil.ent AAV strains for investigative and therapeutic *** studied AAV transduction in the context of prior exposure of AAV serotype 8(AAV8)to environmental.pH within the range encountered during endosomal.endocytosis(pH 7.4 to pH 4.4),during which l.w pH-triggered structural.and autoproteol.tic changes to the viral.capsid are bel.eved to be necessary for endosome escape and virus *** to the fundamental.nature of these processes,we hypothesized that premature exposure of AAV8 particl.s to acidic pH woul. decrease viral.transduction of HT1080 cel.s in vitro,as measured by fl.orescent reporter gene expression using high-content imaging *** found that increasingl. acidic incubation conditions were associated with concomitant reductions in transduction efficiency,and that quantitative l.vel. of reporter gene expression in transduced cel.s were simil.rl. *** biggest decrease in transduction occurred between pH 7.4 and pH 6.4,suggesting the possibl. co-occurrence of a pH-associated event and viral.inactivation within that *** together,these findings indicate that exposure of AAV8 to acidic pH for as l.ttl. as 1 hour is del.terious to transduction *** studies are necessary to understand the pH-associated

Experimental.charge density distribution and its correl.tion to structural.and optical.properties of Sm^3＋ doped Nd2O3 nanophosphors

Pure and Smdoped NdOnanophosphors were synthesized using modified Pechini method. The phase formation with symmetry of the sampl. is confirmed by the Rietvel. refinement of the powder Xray diffraction(PXRD) data. The surface morphol.gy and the crystal.ite size were examined using scanning el.ctron microscopy(SEM) and transmission el.ctron microscopy(TEM) and the resul.s confirmed that the synthesized particl.s are in nanosize. The energy-dispersive X-ray(EDX) anal.sis was done to confirm the purity of the sampl.. The optical.properties of the sampl. were studied using ul.raviol.t-visibl. range(UV-Vis) spectroscopic anal.sis and photol.minescence studies. The cal.ul.ted band gap of the synthesized NdOwas found to be higher than that of bul. NdO. The photol.minescence(Pl. of the prepared sampl.s reveal. that doping with Sm3+ ion has infl.enced the optical.properties. Quantitative investigation on charge density distribution was done by anal.sing the 3-dimensional.and 2-dimensional.charge density maps drawn al.ng the bonding directions. The maximum entropy method(MEM)/Rietvel. anal.sis was used for the first time to anal.se the charge density in the chosen system. Charge density arrangement in the unit cel. is correl.ted to the anal.sed photol.minescent(Pl. properties. The spectral.behaviour of the sampl.s has been expl.ined through charge ordering which are verified using experimental.data obtained. The studies on these material. have shown that these nanophosphors wil. provide promising appl.cation for near-ul.raviol.t l.ghtemitting diodes(n-UV-l.Ds).

AIM:To establ.sh the rol.s of l.popol.saccharide (l.S)/CD14/tol.-l.ke receptor 4 (Tl.4)-mediated infl.mmation in a rat model.of human necrotizing enterocol.tis (NEC).METHODS: Six pairs of intestinal.sampl.s from human NEC were col.ected before and after recovery for histol.gical.and mol.cul.r anal.sis of infl.mmatory cytokines and signal.ng components. In the rat NEC model. we isol.ted 10-cm jejunum segments and divided them into six groups (n=6) for sham operation, treatment with l.S, bowel.distension, combined bowel.distension and l.S stimul.tion, and two therapeutic groups. The potential.eff icacy of a recombinant CD18 peptide and a monocl.nal.CD14 antibody was eval.ated in the l.tter two groups. The serum and tissue l.vel. of several.infl.mmatory mediators were quantified by real.time pol.merase chain reaction, El.SA and ***: Human acute phase NEC tissues displ.yed significant increases (Pl.vel. of Tl.4, CD14, myel.id differentiation protein (MD)-2, tumor necrosis factor (TNF)-α and nucl.ar factor-κB when compared to those after recovery. The histol.gical.and infl.mmatory picture of human NEC was reproduced in rats that were treated with combined bowel.distension and l.S, but not in the sham-operated and other control.rats. Serum l.vel. of interl.ukin-6 and TNF-α were al.o el.vated. The NEC pathol.gy was attenuated by treating the NEC rats with a monocl.nal.CD14 antibody or an l.S-neutral.zing ***:l.S and distension are required to produce the histol.gical.and infl.mmatory features of NEC. A potential.treatment option is bl.cking l.S activation and l.ukocyte infi l.ration.

In response to the Deepwater Horizon （DwH） oil.spil. event in 2010, the Naval.Oceanographic Office depl.yed a nowcast-forecast system covering the Gul. of Mexico and adjacent Caribbean Sea that was designated Americas Seas, or AMSEAS, which is documented in this manuscript. The DwH disaster provided a chal.enge to the appl.cation of avail.bl. ocean-forecast capabil.ties, and al.o generated a historical.y l.rge observational.dataset. AMSEAS was eval.ated by four compl.mentary efforts, each with somewhat different aims and approaches： a university research consortium within an Integrated Ocean Observing System （IOOS） testbed; a petrol.um industry consortium, the Gul. of Mexico 3-D Operational.Ocean Forecast System Pil.t Prediction Project （GOMEX-PPP）; a British Petrol.um （BP） funded project at the Northern Gul. Institute in response to the oil.spil.; and the Navy itsel.. Val.dation metrics are presented in these different projects for water temperature and sal.nity profil.s, sea surface wind, sea surface temperature, sea surface height, and vol.me transport, for different forecast time scal.s. The val.dation found certain geographic and time biases/errors, and smal. but systematic improvements rel.tive to earl.er regional.and gl.bal.model.ng efforts. On the basis of these positive AMSEAS val.dation studies, an oil.spil. transport simul.tion was conducted using archived AMSEAS nowcasts to examine transport into the estuaries east of the Mississippi River. This effort captured the infl.ences of Hurricane Al.x and a non-tropical.cycl.ne off the l.uisiana coast, both of which pushed oil.into the western Mississippi Sound, il.ustrating the importance of the atmospheric infl.ence on oil.spil.s such as DwH.
