Anti-metastatic potential of OR3 pigment from Streptomyces coelicolor JUAcT03 on MDA-MB-231 breast cancer in vitro and in vivo

Objective:To investigate the mechanism of anticancer activity of a pigment OR3 from Streptomyces coelicolor in in vitro and in vivo metastatic breast cancer models and to characterize the ***:The anticancer mechanism was analyzed in MDA-MB-231 cells using MTT,lactate dehydrogenase,caspase,DNA fragmentation,clonogenic,flow cytometry,Western blot,and scratch *** effects of OR3 on xenograft mouse models were evaluated by tumor volume measurement,hematological analysis,and histopathological *** characterization of OR3 was also performed using gas chromatograohy-mass spectrometry and nuclear magnetic resonance ***:OR3 exhibited potent cytotoxicity against MDA-MB-231 cells,with no observed effects on HEk-293 ***-9 activation was detected in OR3-treated MDA-MB-231 *** cytometry showed a dose-dependent induction of apoptosis and cell cycle arrest at the sub-G_(1) and S ***,OR3 completely inhibited MDA-MB-231 cell migration and demonstrated anti-proliferative effects by downregulating the protein expression of kPNA2,XPO1,RAB5B,and p38 *** in vivo studies,OR3 was non-toxic to mice,inhibited tumor xenograft growth,and maintained normal hematological parameters and tissue *** magnetic resonance spectroscopy demonstrated the presence of a prodigiosin-like compound,while gas chromatography-mass spectrometry analysis identified additional compounds in ***:Our findings validate OR3 pigment as a promising compound for the treatment of metastatic breast cancer,warranting further studies.

Pharmacogenomics of cisplatin Sensitivity in Non-small cell Lung cancer

cisplatin, a platinum-based chemotherapeutic drug, has been used for over 30 years in a wide variety of cancers with varying degrees of success. In particular, cisplatin has been used to treat late stage non-small cell lung cancer （NScLc） as the standard of care. However, therapeutic outcomes vary from patient to patient. considerable efforts have been invested to identify biomark- ers that can be used to predict cisplatin sensitivity in NScLc. Here we reviewed current evidence for cisplatin sensitivity biomarkers in NScLc. We focused on several key pathways, including nucleotide excision repair, drug transport and metabolism. Both expression and germline DNA variation were evaluated in these key pathways. current evidence suggests that cisplatin-based treatment could be improved by the use of these biomarkers.

Artemisia annua, commonly known as sweet wormwood or Qinghao, is a shrub native to china and has long been used for medicinal purposes. A. annua is now cultivated globally as the only natural source of a potent anti-malarial compound, artemisinin. Here, we report a high-quality draft assembly of the 1.74-gigabase genome of A. annua, which is highly heterozygous, rich in repetitive sequences, and contains 63 226 protein-coding genes, one of the largest numbers among the sequenced plant species. We found that, as one of a few sequenced genomes in the Asteraceae, the A. annua genome contains a large number of genes specific to this large angiosperm clade. Notably, the expansion and functional diversification of genes encoding enzymes involved in terpene biosynthesis are consistent with the evolution of the artemi- sinin biosynthetic pathway. We further revealed by transcriptome profiling that A. annua has evolved the sophisticated transcriptional regulatory networks underlying artemisinin biosynthesis. Based on compre- hensive genomic and transcriptomic analyses we generated transgenic A. annua lines producing high levels of artemisinin, which are now ready for large-scale production and thereby will help meet the chal- lenge of increasing global demand of artemisinin.

Information Security, Ethics, and Integrity in LLM Agent Interaction

This study addresses security and ethical challenges in LLM-based Multi-Agent Systems, as exemplified in a blockchain fraud detection case study. Leveraging blockchain’s secure architecture, the framework involves specialized LLM Agents—contractMining, Investigative, Ethics, and PerformanceMonitor, coordinated by a ManagerAgent. Baseline LLM models achieved 30% accuracy with a threshold method and 94% accuracy with a random-forest method. The claude 3.5-powered LLM system reached an accuracy of 92%. Ethical evaluations revealed biases, highlighting the need for fairness-focused refinements. Our approach aims to develop trustworthy and reliable networks of agents capable of functioning even in adversarial environments. To our knowledge, no existing systems employ ethical LLM agents specifically designed to detect fraud, making this a novel contribution. Future work will focus on refining ethical frameworks, scaling the system, and benchmarking it against traditional methods to establish a robust, adaptable, and ethically grounded solution for blockchain fraud detection.

This study was conducted to investigate the properties of Pr3+-Dy3+doped Zn2Y-type hexaferrite for potential technological applications.X-ray diffraction（XRD）,Fourier transform infrared spectroscopy（FTIR） and field-emission scanning electron microscopy（FESEM） were used to analyze structural *** crystallite size in the synthesized samples varies between 57.54 and 68.57 *** vibrational bands at 400 and 600 cm-1,common to all hexaferrites,were confirmed through *** FESEM analysis reveals an agglomeration of magnetic grains and a decrease in the average grain size from 1.24 to1.06 μ*** M-H loops show that,with x values of 0.0,0.1,and 0.2,the saturation magnetization is determined to be 34.76,34.23,and 32.52 emu/g,*** corresponding coercivity values are21.24,30.39,and 33.99 ***-visible spectroscopy using Tauc theory reveals an increase in the optical band gap from 2.32 to 2.50 eV,indicating a tunable energy band structure by incorporating Pr3+-Dy3+*** dielectric constant increases,whereas Ac conductivity decreases with increased Pr3+-Dy3+*** obtained results suggest the potential suitability of these materials for various technological applications.

Late Pleistocene calcretes from central Anatolia(Lakes Eymir and Mogan, G?lba?? Basin): comparison to Quaternary calcretes from Turkey

This study is the first to report the radiocarbon ages, mineralogical, morphological, geochemical and stable isotope compositions of calcretes from Neogene sediments around lakes Eymir and Mogan in the G？lba？？ Basin of central Anatolia. Morphologically different forms of calcretes in the G？lba？？ Basin include powdery, nodular, fracture infill, laminar and hardpan types. calcite is the dominant mineral of calcrete compositions; the diagnostic features of dessication cracks, random fractures, Mn O linings and dense sparitic infillings are observed. chemical analyses show arid conditions with mean annual precipitation of 〈50 mm. δ13c compositions of the calcretes range from-6.77‰ to-9.32‰ PDB, typical for most pedogenic calcretes, reflecting the development under seasonally arid climates and c3-dominated vegetation cover. δ18O values are between-5.57‰ and-7.80‰ PDB, indicating the formation from meteoric water in a vadose zone environment. The results suggest that the Middle Pleistocene was arider and warmer, favouring the formation of palygorskite in association with the different forms of calcrete occurrences, whereas the Late Pleistocene was dryer and cooler, supporting the development of calcretes.

Adenoid cystic carcinoma(Acc) of the breast is a rare special subtype of breast cancer characterized by the presence of a dual cell population of luminal and basaloid cells arranged in specific growth patterns. Most breast cancers with triple-negative, basal-like breast features(i.e., tumors that are devoid of estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2 expression, and express basal cell markers) are generally high-grade tumors with an aggressive clinical course. conversely, while Accs also display a triple-negative, basal-like phenotype, they are usually low-grade and exhibit an indolent clinical behavior. Many discoveries regarding the molecular and genetic features of the Acc, including a specific chromosomal translocation t(6;9) that results in a MYB-NFIB fusion gene, have been made in recent years. This comprehensive review provides our experience with Acc of the breast, as well as an overview of clinical, histopathological, and molecular genetic features.

Developing a chloramine decay index to understand nitrification:A case study of two chloraminated drinking water distribution systems

The management of chloramine decay and the prevention of nitrification are some of the critical issues faced by water utilities that use chloramine as a *** this study,potential association between high performance size exclusion chromatography（HPSEc）data obtained with multiple wavelength Ultraviolet（UV） detection from two drinking water distribution systems in Australia and nitrification occurrence was *** increase in the absorbance signal of HPSEc profiles with UV detection at λ = 230 nm between apparent molecular weights of 200 to 1000 Da was observed at sampling sites that experienced rapid chloramine decay and nitrification while its absorbance signal at λ =254 nm decreased.A chloramine decay index（c.D.I） defined as the ratio of area beneath the HPSEc spectra at two different wavelengths of 230 and 254 nm,was used in assessing chloramine decay *** *** of waters at locations that experienced nitrification were consistently higher than locations not experiencing nitrification.A simulated laboratory study showed that the formation of nitrite/nitrate and/or soluble microbial products and/or the release of extracellular polymeric substances（EPS） during nitrification may contribute to the *** findings suggest that c.D.I derived from HPSEc with multiple wavelength UV detection could be an informative index to track the occurrence of rapid chloramine decay and nitrification.

THE NONLINEAR INTERAcTION OF VORTEX RINGS WITH A FREE SURFAcE

Nonlinear interactions of vortex rings with a free surface are considered in an incompressible, ideal fluid using the vortex contour dynamics technique and the boundary integral equation method. The flow is axisymmetric and the vorticity is linearly distributed in the vortex. Effects of the gravity and the surface tension as well as the initial geometric parameter of the vortex on the interaction process are investigated in considerable detail. The interaction process may be divided into three major stages: the vortex free-traveling stage, the collision stage, and the vortex stretching and rebounding stage. Time evolutions of both the vortex and free surface under various conditions are provided and analyzed. Two kinds of waves exist on the free surface during interaction. In a special case where the gravity and surface tension are very weak or the vortex is very strong, an electric-bulb-like 'cavity' is formed an the free surface and the vortex is trapped in the 'cavity' for quite a. long time, resulting in a large amount, of fluid above the mean fluid surface.
