Venous thrombosis and prothrombotic factors in inflammatory bowel disease

Patients with inflammatory bowel disease(IBD)may have an increased risk of venous thrombosis(VTE).PubMed,ISI Web of Knowledge and Scopus were searched to identify studies investigating the risk of VTE and the prevalence of acquired and genetic VTE risk factors and prothrombotic abnormalities in ***,IBD patients have a two-to fourfold increased risk of VTE compared with healthy controls,with an overall incidence rate of 1%-8%.The majority of studies did not show significant differences in the risk of VTE between Crohn’s disease and ulcerative *** acquired factors are responsible for the increased risk of VTEin IBD:inflammatory activity,hospitalisation,surgery,pregnancy,disease phenotype(e.g.,fistulising disease,colonic involvement and extensive involvement)and drug therapy(mainly steroids).There is also convincing evidence from basic science and from clinical and epidemiological studies that IBD is associated with several prothrombotic abnormalities,including initiation of the coagulation system,downregulation of natural anticoagulant mechanisms,impairment of fibrinolysis,increased platelet count and reactivity and dysfunction of the *** genetic alterations are not generally found more often in IBD patients than in nonIBD patients,suggesting that genetics does not explain the greater risk of VTE in these *** VTE may have clinical specificities,namely an earlier first episode of VTE in life,high recurrence rate,decreased efficacy of some drugs in preventing further episodes and poor *** should be aware of these risks,and adequate prophylactic actions should be taken in patients who have disease activity,are hospitalised,are submitted to surgery or are undergoing treatment.

Immune response to inactivated bacterial vector carrying the recombinant K39 antigen of Leishmania infantum in mice

objective:To evaluate the immunological response elicited by an inactivated bacterial vector carrying the K39 antigen of Leishmania infantum,and a purified ***:Mice were subjected to the following treatments:(1)Purified recombinant K39(rK39)protein at a 20μg dose with complete Freund’s adjuvant;(2)Inactivated Escherichia coli(BL21 DE3)carrying the K39 protein at an equivalent total protein content of 200μg;(3)Inactivated bacteria lacking the K39 protein;(4)Non-immunized control *** monitoring was *** groups were challenged by intraperitoneal injection of 10^(7) Leishmania infantum *** euthanasia,the liver and spleen were collected to analyze the levels of TNF,IFN-γ,IL-12,IL-4,and ***:Mice immunized with purified rK39 or the inactivated bacterial vector carrying the K39 antigen of Leishmania infantum showed a long-lasting immune response with high levels of polyclonal antibodies specifically recognizing the recombinant *** IgG1 subclass was the predominant immunoglobulin;however,the induction of IgG2a and the profile of cytokines produced were indicative of the induction of a mixed-type ***:The inactivated bacterial vector carrying the K39 antigen,as well as the purified antigen can induce a long-lasting immune response in immunized mice,predominantly favouring a Th2 profile response.

Introduction: 68Ga-PSMA-11 is considered the gold standard in detection of micro and oligometastases in advanced prostate cancer, being used for therapeutic planning, as well as, potentially, for evaluating response to treatment. The development of ready-to-use lyophilized kit of PSMA-11 adds quality and safety to the routine use of this radiopharmaceutical and represents a pharmacotechnical challenge as it must preserve the integrity and specificity of the ligand. Methods: PSMA-11 kit formulation was proposed, considering radiolabeling parameters and the preservation of the peptide during the lyophilization process, using mannitol as an excipient. Critical temperature characterization studies were carried out using DSC equipment and the freeze-drying process was developed. The direct radiolabeling conditions were evaluated and standardized using 68Ge/68Ga generator eluate from two different manufacturers (ITG and Eckert & Ziegler). The radiochemical purity was evaluated by TLC and HPLC. Biological evaluation was carried out with lyophilized PSMA-11 to demonstrate the integrity of the peptide and preservation of biological activity after the lyophilization process. Results: Based on critical temperature characterization studies, the freeze-drying cycle was designed to reach a freezing temperature of around −40˚C and primary drying at 2˚C. Using 20 mg of mannitol, an intact and elegant lyophilized cake was obtained. PSMA-11 lyophilized kit was directly labeled with 68Ga eluate from 68Ge/68Ga GMP generators (ITG and Eckert & Ziegler) resulting in % RP > 95% at pH 4.0 to 4.5. The results obtained from in vitro and in vivo biological competition studies confirmed the preservation of PSMA-11 affinity for the receptor after lyophilization. Conclusion: A lyophilized formulation (Kit) of PSMA-11 was successfully obtained, which preserved the integrity and biological activity of the peptide and guaranteed radiolabeling efficiency.

The broke of a tailings dam from Vale S.A. destroyed a gigantic area killing 270 persons in Brumadinho/Brazil. organizing activities after a mass disaster is a complex process that requires the involvement of many people and resources in the laboratory. To DNA identification of the victims, a daunting work had to be made by DNA laboratory staff in a collaborative effort with many partners. Efforts to implement good practice guidelines in Disaster Victim Identification (DVI) have revealed several important aspects that need to change in the forensic DNA laboratory. This article highlights the challenges of implementing DVI best practice guidelines in resource-poor settings, but with professionals from different sectors engaged in the same goal of briefly identifying victims and helping families and society.

High Frequency of Antiviral Resistance Mutations in HBV Genotypes A2 and H: Multidrug Resistance Strains in Mexico

Background and Aims:Lamivudine(3TC),telbivudine(LdT),entecavir(ETV),adefovir(ADF),and tenofovir(TFV)are drugs used to treat hepatitis B virus(HBV)infection,but specific mutations allow some viruses to become resistant to antiviral drugs or to acquire immune escape *** mutations have not been thoroughly investigated in *** study aimed to estimate the prevalence of HBV antiviral resistance and escape ***:This cross-sectional study analyzed 158 *** DNA was extracted,amplified,and sequenced in serum samples using the spin column method,PCR assay,and Sanger’s sequencing,*** genotypes were determined,and HBV mutations were tested using the Geno2pheno ***:overall,68.4%(108/158)of HBV patients were infected with genotype H,followed by G(11.4%,18/158),A2(10.8%,17/158),F1b(6.9.0%,11/158),D(1.9%,3/158),and E(0.6%,1/158),and 5.1%(8/158)had evidence of *** prevalence of resistance mutations was 8.2%(13/158)and the most common combined mutation was rt180M+***,we found the combinations rt180M+rt204V+rt173L(n=2)and rt180M+rt204V+rt202G(n=1)that confer multidrug resistance to 3TC,LdT,and *** mutations were found in genotypes A2(11.8%,2/17),and H(10.2%,11/108),and escape mutations were detected in HBV genotypes A2(11.8%,2/17),H(10.2%,11/108),F1b(9.1%,1/11)and G(5.6%,1/18).Conclusions:The highest prevalence of antiviral resistance mutations or escape mutations was detected in HBV genotypes A2 and *** earliest cases of HBV multidrug resistance were detected in Mexico.

Mammalian cell surfaces consist of the plasma membrane supported by an underneath cortical ***,these structures can control not only the shape of cells but also a series of cellular functions ranging from migration and division to exocytosis,endocytosis and ***,the cell surface is capable of exerting and reacting to mechanical *** viscoelastic properties,especially membrane tension and bending modulus,are fundamental parameters involved in these *** viewpoint summarizes our current knowledge on how to measure the viscoelastic properties of cell surfaces employing optical tweezers-based tether assays,paving the way for a better understanding of how cells react to external mechanical forces,with a glance on their remodeling dynamics and possible consequences on downstream cellular processes.

Modelling the role of ground-true riparian vegetation for providing regulating services in a Mediterranean watershed

Intensive agricultural and industrial activities are often considered major sources of water ***,riparian vegetation(RV)is increasingly being promoted as a solution to balance the potentially adverse effects that agriculture may have on water ***,existing RV is often overlook in recent modelling efforts,failing to capture the current amount of ecosystem services ***,we used the Soil and Water Assessment Tool ecohydrological model to simulate the influence of ground-true RV on i)nutrient(nitrate and total phosphorus)and sediment exports from agricultural areas and ii)its effect for in-stream *** results are further compared against a set of hypothetical scenarios of different RV widths and different *** results point to a great rele-vance of existing RV in controlling in-stream concentration of sediments and nutrients where pressure from agriculture is highest,preventing them to surpass limits set in the EU Water Framework *** the other hand,in areas with industry discharges,the role of RV is limited and model results suggest that restoring RV would have limited *** illustrate how existing RV may already provide strong but not acknowledged water quality regulation services,how these services can differ substantially between nearby streams,and that effective strategies to improve water quality using RV must acknowledge existing patterns of vegetation,land use and contamination sources.

Automatic detection and evaluation of sugarcane planting rows in aerial images

Sugarcane planting is an important and growing activity in ***,several techniques have been developed over the years to maximize crop productivity and profit,amongst them,processing of sugarcane field *** this sense,this research aims to identify and analyze crop rows and measure their gaps from aerial images of sugarcane *** this,a small Remotely Piloted Aircraft captured the images,generating orthomosaics of the areas for ***,each orthomosaic is classified with the K-Nearest Neighbor algorithm to segment regions of *** row orientation is estimated using the RGB gradient *** operations and computational geometry models are then used to detect and map rows and gaps along the planting row *** evaluate the results,crop rows are mapped and compared to manually taken *** technique obtained an error smaller than 2%when compared to gap length in crop rows from an orthomosaic with the area of 8.05 ha(ha).The proposed approach can map the positioning of the automatically generated row segments appropriately onto manually created ***,our method also achieved similar results when confronted with a manual technique for differing growth stages(40 and 80 days after harvest)of the sugarcane *** proposed method presents a great potential to be adopted in sugarcane planting monitoring。

Recent advances in magnetic electrospun nanofibers for cancer theranostics application

Cancer theranostics is a recent concept that aims to combine in the same device diagnostic and therapeutic features. Magnetic nanoparticles(m NPs) are commonly used as a critical part of these systems due to their ability to respond to an external magnetic field. Consequently, m NPs can generate heat when an alternating magnetic field is applied and enhance image contrast in magnetic resonance. However, direct administration of m NPs intravenously or directly in the tumor can lead to undesired side effects because of m NP elimination by macrophages or leakage to healthy tissues. Therefore, m NPs can be retained in a polymeric nanofibrous mesh, thus preventing misplacing or loss of m NPs. Furthermore, these magnetic nanofibers can be directly implanted in the tumor site, thus ensuring high m NPs loading and higher magnetic response. In addition, polymeric nanofibers produced by electrospinning are frequently used to maintain a sustained drug release in the tumor site. Therefore,a magnetic polymeric nanofiber produced by electrospinning is an ideal nanosystem for cancer theranostics application. This review summarizes the most recent developments of magnetic nanofibers produced by electrospinning for cancer theranostics applications.

Competition among rare earth elements on sorption onto six seaweeds

Sorption has been proposed as a promising approach towards a sustainable water decontamination and recently to the recycling of rare earth elements(REE).Although living seaweed have been shown to be capable of removing REE from contaminated solutions,no studies have yet compared the effects of REE competition on sorption onto different groups of *** effects were analysed in the present study by exposing six living seaweeds(Ulva lactuca,Ulva intestinalis,Fucus spiralis,Fucus vesiculosus,Gracilaria *** osmundea pinnatifida)(3 g/L,fresh weight) to mono-element and multi-element solutions of Y,La,Ce,Pr,Nd,Eu,Gd,Tb and Dy(1 μmol/L).Results show a preference towards light REE in mono-element solutions,which is reduced when in competition with other *** competitive effect is less pronounced in heavy REE indicating that these are still able to bind to the macroalgae despite the presence of competing *** to water content(%),seaweed specific surface area(cm^(2)/g) shows to be an important factor in the sorption of REE,since larger surface area is associated with higher removal and larger competitive effect.
