Proteome of human colon cancer stem cells:A comparative analysis

AIM: To isolate and identify the biological characteristics of human colon cancer stem cells (SW1116 cells) and further study their proteome. METHODS: SW1116 cells were isolated and cultured with a serum-free medium (SFM). Sphere formation was assayed to observe the formation of colon cancer stem cell spheres. SW1116 cells were inoculated into a serum-containing medium for observing their differentiation characteristics. Proliferation curve and cross-resistance of SW1116 cells to different drugs were detected by MTT. Percentage of SP cells in SW1116 cells was detected with Hoechst33342 staining. Telomerase activity in SW1116cells was checked by polymerase chain reaction (PCR)-enzyme linked immunosorbent assay. Expressions of stem cell relevant genes and proteins were detected by reverse transcription-PCR and Western blot, respectively. Total protein was isolated from SW1116 cells by two-dimensional gel electrophoresis (2-DE) and differentially expressed proteins were identified by tandem mass spectrometry (MALDI-TOF/TOF). RESULTS: The isolated SW1116 cells presented as spheroid and suspension growths in SFM with a strong self-renewal, proliferation, differentiation and drug-resistance ability. The percentage of SP cells in SW1116 cells was 38.9%. The SW1116 cells co-expressed the CD133 and CD29 proteins. The telomerase activity in SW1116 cells was increased. The expressions of different stem cell relevant genes and proteins were detected. The proteomic analysis showed that the 26 protein spots were differently expressed in SW1116 cells and 10 protein spots were identified as ubiquitin fusiondegradation 1-like protein, nuclear chloride channel protein, tubulin β, Raichu404X, stratifi n, F-actin capping protein α-1 subunit, eukaryotic translation elongation factor 1 delta isoform 2, hypothetical protein, glyceraldehyde-3-phosphate dehydrogenase and guanine nucleotide binding protein β polypeptide 2-like 1, respectively. miR-93 suppresses proliferation and colony formation of human colon cancer stem cells

AIM:To identify differentially expressed microRNAs(miRNAs) in human colon cancer stem cells(SW1116csc) and study their function in SW1116csc ***:SW1116csc were isolated from the humancolon cancer cell line,SW1116 and cultured in serum free medium. A miRNA microarray was used to detect differential expression profiles of miRNAs in SW1116cscand SW1116 cells. Real-time quantitative polymer asechain reaction(PCR) was performed to verify the differential expression of candidate miRNAs obtainedfrom the microarray. Target mRNAs of differentially expressed miRNAs were predicted with target prediction tools. miRNA expression plasmids were transfectedinto SW1116csc using Lipofectamine 2000 reagent. Cellproliferation curves were generated with trypan bluestaining,and the colony formation rate of transfectedcells was measured with the soft agar colony formationassay. Expression of target mRNAs and proteins from differentially expressed miRNAs were detected usingreverse transcription(RT) -PCR and western ***:Compared with expression in SW1116 cells,35 miRNAs(including hsa-miR-192,hsa-miR-29b,hsa-miR-215,hsa-miR-194,hsa-miR-33a and hsa-miR-32) were upregulated more than 1.5-fold,and 11miRNAs(including hsa-miR-93,hsa-miR-1231,hsa-miRPlus-F1080,hsa-miR-524-3p,hsa-miR-886-3p andhsa-miR-561) were downregulated in SW1116csc. The miRNA microarray results were further validated with quantitative RT-PCR. miR-93 was downregulated,and its predicted mRNA targets included BAMBI,CCND2,CDKN1A,HDAC8,KIF23,MAP3K9,MAP3K11,MYCN,PPARD,TLE4 and ZDHHC1. Over expressed miR-93 sig-nificantly in hibited cell proliferation and colony formation by SW1116csc. Furthermore,miR-93 negatively regulated the mRNA and protein levels of HDAC8 and ***:Some miRNAs were differentially expressed during differentiation of SW1116csc into SW1116cells. miR-93 may inhibit SW1116csc proliferation andcolony formation.

Azithromycin-containing versus standard triple therapy for Helicobacter pylori eradication:A meta-analysis

AIM:To evaluate whether adding azithromycin to firstline Helicobacter pylori(H pylori)eradication improved eradication and reduced side ***:Eligible articles were identified by searches of electronic *** included all randomized trials that compared azithromycin-containing with standard triple-therapy regimens for first-line treatment of H pylori *** analysis was performed with Review Manager ***-analyses were also ***:We identified 14 randomized trials(1431 patients).Pooled H pylori eradication rates were 72.01%(95%CI:58.09%-85.93%)and 69.78%(95% CI:66.47%-73.09%)for patients with or without azithromycin by intention-to-treat analysis,and the odds ratio(OR)was 1.17(95%CI:0.64-2.14).The occurrence of side effects differed significantly and was 15.81%(95%CI:12.50%-19.12%)and 25.20%(95%CI:21.44%-28.96%)for treatment with or without azithromycin,respectively,and the summary OR was 0.58(95%CI:0.41-0.82).Furthermore,the azithromycin-containing group had a lower occurrence of diarrhea,nausea and taste ***:Our review suggests that azithromycincontaining triple-therapy regimens could be equally effective in eradication of H pylori compared with standard first-line triple-therapy regimens.

Coffee drinking and pancreatic cancer risk:A meta-analysis of cohort studies

AIM:To quantitatively assess the relationship between coffee consumption and incidence of pancreatic cancer in a meta-analysis of cohort studies. METHODS:We searched MEDLINE,EMBASE,Science Citation Index Expanded and bibliographies of retrieved *** were included if they reported relative risks(RRs)and corresponding 95%CIs of pancreatic cancer with respect to frequency of coffee *** performed random-effects meta-analyses and metaregressions of study-specific incremental estimates to determine the risk of pancreatic cancer associated with a 1 cup/d increment in coffee consumption. RESULTS:Fourteen studies met the inclusion criteria, which included 671 080 individuals(1496 cancer events) with an average follow-up of 14.9 *** with individuals who did not drink or seldom drank coffee per day,the pooled RR of pancreatic cancer was 0.82 (95%CI:0.69-0.95)for regular coffee drinkers,0.86 (0.76-0.96)for low to moderate coffee drinkers,and 0.68(0.51-0.84)for high *** subgroup analyses,we noted that,coffee drinking was associated witha reduced risk of pancreatic cancer in men,while this association was not seen in *** associations were also similar in studies from North America,Europe, and the Asia-Pacific region. CONCLUSION:Findings from this meta-analysis suggest that there is an inverse relationship between coffee drinking and risk of pancreatic cancer.

A meta-analysis of the effects of energy intake on risk of digestive cancers

Background:There is no consensus on whether triplet regimen is better than doublet regimen in the first-line treatment of advanced gastric cancer(AGC).We aimed to compare the efficacy and safety of oxaliplatin plus capecitabine(XELOX)and epirubicin,oxaliplatin,plus capecitabine(EOX)regimens in treating ***:This phase III trial enrolled previously untreated patients with AGC whowere randomly assigned to receive the XELOXor *** primary endpoint was non-inferiority in progression-free survival(PFS)for XELOX as compared with EOX on an intention-to-treat ***:Between April 10,2015 andAugust 20,2020,448AGCpatientswere randomized to receive XELOX(n=222)or EOX(n=226).The median PFS(mPFS)was 5.0 months(95%confidence interval[CI]=4.5-6.0 months)in the XELOX arm and 5.5 months(95%CI=5.0-6.0 months)in the EOX arm(hazard ratio[HR]=0.989,95%CI=0.812-1.203;P_(non-inferiority)=0.003).There was no significant difference inmedian overall survival(mOS)(12.0 vs.12.0months,P=0.384)or objective response rate(37.4%vs.45.1%,P=0.291)between the two *** patients with poorly differentiated adenocarcinoma and liver metastasis,the EOX arm had a significantly longer mOS(P=0.021)and a trend of longer mPFS(P=0.073)than the XELOX *** rate of grade 3/4 adverse events(AEs)was 42.2%(90/213)in the XELOX arm and 72.5%(156/215)in the EOX arm(P=0.001).The global health-related quality of life(QoL)score was significantly higher in the XELOX arm than in the EOX arm during ***:This non-inferiority trial demonstrated that the doublet regimen was as effective as the triplet regimen and had a better safety profile and QoL as a first-line treatment for AGC ***,the triplet regimen might have a survival advantage in patients with poorly differentiated adenocarcinoma and liver metastasis.

The continuously arising of SARS-CoV-2 variants has been posting a great threat to public health safety globally,from B.1.17(Alpha), B.1.351(Beta), P.1(Gamma), B.1.617.2(Delta) to B.1.1.529(Omicron). The emerging or reemerging of the SARS-CoV-2 variants of concern is calling for the constant monitoring of their epidemics,pathogenicity and immune escape. In this study, we aimed to characterize replication and pathogenicity of the Alpha and Delta variant strains isolated from patients infected in Laos. The amino acid mutations within the spike fragment of the isolates were determined via sequencing. The more efficient replication of the Alpha and Delta isolates was documented than the prototyped SARS-CoV-2 in Calu-3 and Caco-2 cells, while such features were not observed in Huh-7, Vero E6 and HPA-3 cells. We utilized both animal models of human ACE2(hACE2)transgenic mice and hamsters to evaluate the pathogenesis of the isolates. The Alpha and Delta can replicate well in multiple organs and cause moderate to severe lung pathology in these animals. In conclusion, the spike protein of the isolated Alpha and Delta variant strains was characterized, and the replication and pathogenicity of the strains in the cells and animal models were also evaluated.

Monkeypox is a zoonotic *** the first human monkeypox case was detected in 1970,it has been prevalent in some countries in central and western *** May 2022,monkeypox cases have been reported in more than 96 non-endemic countries and regions *** of September 14,2022,there have been more than 58,200 human monkeypox cases,and there is community *** cessation of smallpox vaccination in 1980,which had some cross-protection with monkeypox,resulted in a general lack of immunity to monkeypox,which caused global concern and *** of Sep-tember 14,2022,there are four monkeypox cases in China,including three in Taiwan province and one in Hong Kong *** foreign studies have shown that children are vulnerable to monkeypox and are also at high risk for severe disease or *** order to improve pediatricians'understanding of monkeypox and achieve early detection,early diagno-sis,early treatment,and early disposal,we have organized national authoritative experts in pediatric infection,respiratory,dermatology,critical care medicine,infectious diseases,and public health and others to formulate this expert consensus,on the basis of the latest"Clinical management and infection prevention and control for monkeypox"released by The World Health Organization,the"guidelines for diagnosis and treatment of monkeypox(version 2022)"issued by National Health Commission of the People's Republic of China and other relevant *** the development of this consensus,multidisciplinary experts have repeatedly demonstrated the etiology,epidemiology,transmission,clinical manifestations,laboratory examinations,diagnosis,differential diagnosis,treatment,discharge criteria,prevention,disposal process,and key points of prevention and control of suspected and confirmed cases.

Fish consumption and risk of gastrointestinal cancers:A meta-analysis of cohort studies

AIM:To assess quantitatively the relationship between fish intake and the incidence of gastrointestinal cancers in a meta-analysis of cohort ***:We searched MEDLINE,Embase,Science Citation Index Expanded,and the bibliographies of retrieved *** cohort studies were included if they reported relative risks(RRs) and corresponding 95% confidence intervals(CIs) of various cancers with respect to fish *** RRs were not available in the published article,they were computed from the exposure *** investigators extracted the data independently and discrepancies were resolved by discussion with a third *** performed random-effect meta-analyses and metaregressions of study-specific incremental estimates to determine the risk of cancer associated with a 20-g/d increment of fish ***:Forty-two studies,comprising 27 independent cohorts,met our inclusion *** studies included 2325040 participants and 24115 incident cases of gastrointestinal cancer,with an average follow-up of 13.6 *** with individuals who did not eat,or seldom ate,fish,the pooled RR of gastrointestinal cancers was 0.93(95%CI:0.88-0.98) for regular fish consumers,0.94(0.89-0.99) for low to moderate fish consumers,and 0.91(0.84-0.97) for high fish ***,a 20-g increase in fish consumption per day was associated with a 2% reduced risk of gastrointestinal cancers(RR = 0.98; 95%CI:0.96-1.01).In subgroup analyses,we noted that fish consumption was associated with reduced risk of colorectal(RR = 0.93; 95%CI:0.87-0.99; P < 0.01),esophageal(RR = 0.91; 95%CI:0.83-0.99; P < 0.05) and hepatocellular cancers(RR = 0.71; 95%CI:0.48-0.95; P < 0.01).CONCLUSION:This meta-analysis suggested that fish consumption may reduce total gastrointestinal cancer *** relationships were also detected between fish consumption and specific types of cancers.
