A Systems Biology Approach for Studying Heterotopic Ossification： Proteomic Analysis of clinical Serum and Tissue Samples

Heterotopic ossification （HO） refers to the abnormal formation of bone in soft tissue. Although some of the underlying processes of HO have been described, there are currently no clinical tests using validated biomarkers for predicting HO formation. As such, the diagnosis is made radiographically after HO has formed. To identify potential and novel biomarkers for HO, we used isobaric tags for relative and absolute quantitation （iTRAQ） and high-throughput antibody arrays to produce a semi-quantitative proteomics survey of serum and tissue from subjects with （HO ＋） and without （HO-） heterotopic ossification. The resulting data were then analyzed using a systems biology approach. We found that serum samples from subjects experiencing traumatic injuries with resulting HO have a different proteomic expression controls. Subsequent quantitative ELISA identified profile compared to those from the matched five blood serum proteins that were differentially regulated between the HO-- and HO- groups. compared to HO- samples, the amount of insulin-like growth factor I （IGF1） was up-regulated in HO＋ samples, whereas a lower amount of osteopontin （OPN）, myeloperoxidase （mPO）, runt-related transcription factor 2 （RUNX2）,and growth differentiation factor 2 or bone morphogenetic protein 9 （BmP-9） was found in HO ＋ samples （Welch two sample t-test; P 〈 0.05）. These proteins, in combination with potential serum biomarkers previously reported, are key candidates for a serum diagnostic panel that may enable early detection of HO prior to radiographic and clinical manifestations.

Polycyclic aromatic hydrocarbons(PAHs)are large class of hydrophobic,semi-volatile organic contaminants that may enter the environment from both natural sources and anthropogenic *** PAHs arise from the incomplete combustion of fossil fuels and organic matter and following dispersal via long-range transport and may subsequently deposit in surface waters,soils and sediments of remote regions,including the *** current review summarizes and discusses Arctic data that is available for combustion-derived PAHs between 2004 and early 2018,focusing largely on data collected from remote,unexploited Arctic regions and from studies that provide some evidence of a pyrogenic *** increasing use of attribution ratios,which aid in discriminating PAHs from petrogenic or pyrogenic sources,suggest PAHs found in Arctic marine waters and sediment predominantly originate from natural underwater seeps,while those measured in air,freshwater,and terrestrial environments are likely to have originated from atmospheric and combustion-derived *** efforts indicate that atmospheric PAHs in the canadian and Norwegian Arctic are likely to have originated in the northern hemisphere e predominantly from Western Russia,northern Europe,and North *** Asia appears to be a minor source of PAHs to the Arctic,despite contributing more than 50%of global PAH *** comparison to the growing data for atmospheric PAHs,environmental data for these compounds in terrestrial and freshwater environments remain *** have been detected in Arctic biota from terrestrial,freshwater and marine environments,indicating exposure,however,levels are generally low,as most organisms efficiently metabolize parent ***,PAH emissions are expected to decline in the future,however models suggest the Arctic may not experience the same magnitude of decline projected for other world ***,future changes in climate may contribute to a re-volatilizati

Hexachlorobutadiene(HcBD)is a halogenated hydrocarbon that is primarily produced as an unintentional byproduct in the manufacture of chlorinated *** between HcBD and other persistent organic pollutants(POPs)led to its listing in 2015 for global regulation under the Stockholm convention on ***'s toxicity and propensity for long-range transport means there is special concern for its potential impacts on Arctic *** present review comprehensively summarizes all available information of the occurrence of HcBD in the Arctic environment,including its atmospheric,terrestrial,freshwater and marine ecosystems and ***,reports of HcBD in Arctic environmental media are *** has been measured in Arctic air collected from monitoring stations in Finland and canada,yet there is a dearth of data for other abiotic matrices(***,sediments,glacier ice,freshwaters and seawater).Low HcBD concentrations have been measured in Arctic terrestrial and marine biota,which is consistent with laboratory studies that indicate that HcBD has the potential to bioaccumulate,but not to *** data for Arctic biota suggest that terrestrial birds and mammals and seabirds,have comparatively higher HcBD concentrations than fish and marine mammals,warranting additional *** spatial and temporal trends in HcBD concentrations in the Arctic are currently limited,future monitoring of HcBD in the Arctic will be important for assessing the impact of global regulations newly-imposed by the Stockholm convention on POPs.

Informing management of rare species with an approach combining scenario modeling and spatially explicit risk assessment

Wildlife managers are tasked with identifying and managing stressors that threaten persistence of *** demonstrate an approach to land-use planning that combines scenario modeling and ecological risk assessment to map and quantify risk to population persistence for three rare prairie species in Washington State,*** corroboration of model output,we found that of the management scenarios considered,only a scenario with year-round restrictions on use of off-road vehicles,digging,and camping enforced in all potential habitat reduces risk to the *** risk is focused primarily in two patches of prairie habitat in our study area,indicating stringent restrictions need not be applied ***,one area is not easily accessed by two of the three species considered,suggesting reintroductions to suitable but inaccessible habitat may play an important role in management of these *** analyses suggest changes in land use and management that might improve habitat for rare species,with options for minimizing monetary and social *** the proposed approach relies on hypothetical management scenarios and uses a model flexible in data requirements to provide spatially explicit output,it can be used to inform adaptive management of rare species in diverse land-planning processes and will be especially useful when management decisions must be made under time or cost constraints.

Herbivory and Plant Genotype Influence Fitness-Related Responses of<i>Arabidopsis thaliana</i>to Indirect Plant-Plant Interactions

Previous studies have demonstrated that genetic identity between interacting perennial plants results in more effective defense when emitter and receiver neighbors have greater genetic similarity. However, the effects of both genetic relatedness and presence of herbivores on fitness-related responses of neighboring plants have not yet been explored. Our aim was to examine how manipulating these two important factors genetic and environmental factors can influence indirect plant-plant communication in the annual crucifer Arabidopsis thaliana. Plants of a single genotype (receivers) were exposed to volatile emissions of neighboring emitter plants with a similar or different genotype, and either intact or damaged by larvae of a specialist herbivore for ten days. Each of the four treatments was isolated in separate environmental chambers and the full experiment was replicated twice. Receiver plant growth and reproductive-related traits were measured ten days after exposure to treatments, and at senescence. Results showed that the effect of herbivory and plant genotype of emitter plants influenced responses related growth and reproduction in receiver plants. Receiver plants grew taller, had more inflorescence branching, and produced more fruits (60% more) when exposed to undamaged emitters of a different genotype than receivers exposed to the other emitter plant treatments. Therefore, genotype identity and environmental context (presence of herbivory) may be important factors influencing indirect plant-plant communication, which could, in turn, result in selection for genotypes showing increased fitness-related responses.

As the novel coronavirus disease 2019(cOVID-19)pandemic spreads rapidly across the globe many unanswered questions about the basic biology and epidemiology of the disease hamper our response strategies and limit our ability to achieve control and prevent a rebound or so-called“second wave”.One such crucial question is:To what degree do asymptomatic cases contribute to transmission?Early,small studies on this subject have found wide ranging estimates of the prevalence of asymptomatic carriers,and just a handful of studies so far have documented viral shedding by asymptomatic cases[1].We recently re-examined china’s cOVID-19 case report data to investigate this question[2].This Editorial aims to describe how asymptomatic cases contribute to transmission and what the implications are for control strategies.

Automatic Detection of Nephrops Norvegicus Burrows from Underwater Imagery Using Deep Learning

The Norway lobster,Nephrops norvegicus,is one of the main commercial crustacean fisheries in *** abundance of Nephrops norvegicus stocks is assessed based on identifying and counting the burrows where they live from underwater videos collected by camera systems mounted on *** Spanish Oceanographic Institute(IEO)andmarine Institute Ireland(mIIreland)conducts annual underwater television surveys(UWTV)to estimate the total abundance of Nephrops within the specified area,with a coefficient of variation(cV)or relative standard error of less than 20%.currently,the identification and counting of the Nephrops burrows are carried out manually by the marine *** is quite a time-consuming *** a solution,we propose an automated system based on deep neural networks that automatically detects and counts the Nephrops burrows in video footage with high *** proposed system introduces a deep-learning-based automated way to identify and classify the Nephrops *** research work uses the current state-of-the-art Faster RcNN models Inceptionv2 and mobileNetv2 for object detection and *** conduct experiments on two data sets,namely,the Smalls Nephrops survey(FU 22)and cadiz Nephrops survey(FU 30),collected by marine Institute Ireland and Spanish Oceanographic Institute,*** the results,we observe that the Inception model achieved a higher precision and recall rate than *** best mean Average Precision(mAP)recorded by the Inception model is 81.61%compared to mobileNet,which achieves the best mAP of 75.12%.

AIm To investigate the impact of medication beliefs, illness perceptions and quality of life on medication adherence in people with decompensated *** One hundred adults with decompensated cirrhosis completed a structured questionnaire when they attended for routine outpatient hepatology review. measures of self-reported medication adherence(morisky medication Adherence Scale), beliefs surrounding medications(Beliefs about medicines Questionnaire), perceptions of illness and medicines(Brief Illness Perception Questionnaire), and quality of life(chronic Liver Disease Questionnaire) were examined. clinical data were obtained via patient history and review of medical records. Least absolute shrinkage and selection operator and stepwise backwards regression techniques were used to construct the multivariable logistic regression model. Statistical significance was set at alpha = *** medication adherence was " High " in 42 % o f participants, "medium" in 37%, and "Low" in 21%. compared to patients with "High" adherence, those with "medium" or "Low" adherence were more likely to report difficulty affording their medications(P ception of treatment helpfulness(P = 0.003) and stronger medication concerns relative to medication necessity beliefs(P = 0.003). People with "Low" adherence also experienced greater symptom burden and poorer quality of life, including more frequent abdominal pain(P = 0.023), shortness of breath(P = 0.030), and emotional disturbances(P = 0.050). multivariable analysis identified having stronger medication concerns relative to necessity beliefs(Necessity-concerns Differential ≤ 5, OR = 3.66, 95%cI: 1.18-11.40) and more frequent shortness of breath(shortness of breath score ≤ 3, OR = 3.87,95%cI: 1.22-12.25) as independent predictors of "Low"*** The association between "Low" adherence and patients having strong concerns or doubting the necessity or helpfulness of their medications should be explor
