The molecular links between extracellular signals and the regulation of localized protein synthesis in plant cells are poorly understood.Here,we show that in Arabidopsis thaliana,the extracellular peptide RALF1 and its receptor,the FERONIA receptor kinase,promote root hair(RH)tip growth by modulating protein synthesis.We found that RALF1 promotes FERONIA-mediated phosphorylation of elF4E1,a eukaryotic translation initiation factor that plays a crucial role in the control of mRNA translation rate.Phosphorylated elF4E1 increases mRNA affinity and modulates mRNA translation and,thus,protein synthesis.The mRNAs targeted by the RALF1-FERONIA-elF4E1 module include ROP2 and RSL4,which are important regulators of RH cell polarity and growth.RALF1 and FERONIA are expressed in a polar manner in RHs,which facilitate elF4E1 polar丨ocalization and thus may control local f?OP2 translation.Moreover,we demonstrated that high-level accumulation of RSL4 exerts negative-feedback regulation of RALF1 expression by directly binding the RALF1 gene promoter,determining the final RH size.Our study reveals that the link between RALF1-FERONIA signaling and protein synthesis constitutes a novel component regulating cell expansion in these polar growing cells.

Plant long noncoding RNAs(lncRNAs)have emerged as important regulators of chromatin dynamics,impacting on transcriptional programs leading to different developmental outputs.The lncRNA AUXIN-REGULATED PROMOTER LOOP(APOLO)directly recognizes multiple independent loci across the Arabidopsis genome and modulates their three-dimensional chromatin conformation,leading to transcriptional shifts.Here,we show that APOLO recognizes the locus encoding the root hair(RH)master regulator ROOT HAIR DEFECTIVE 6(RHD6)and controls RHD6 transcriptional activity,leading to cold-enhanced RH elongation through the consequent activation of the transcription factor gene RHD6-like RSL4.Furthermore,we demonstrate that APOLO interacts with the transcription factor WRKY42 and modulates its binding to the RHD6 promoter.WRKY42 is required for the activation of RHD6 by low temperatures and WRKY42 deregulation impairs cold-induced RH expansion.Collectively,our results indicate that a novel ribonucleoprotein complex with APOLO and WRKY42 forms a regulatory hub to activate RHD6 by shaping its epigenetic environment and integrate signals governing RH growth and development.

BACKGROUND The prognostic value of the Systemic Inflammation Response Index(SIRI)in advanced pancreatic cancer is recognized,but its correlation with patients´nutritional status and outcomes remains unexplored.AIM To study the prognostic significance of SIRI and weight loss in metastatic pancreatic cancer.METHODS The PANTHEIA-Spanish Society of Medical Oncology(SEOM)study is a multicentric(16 Spanish hospitals),observational,longitudinal,non-interventional initiative,promoted by the SEOM Real World-Evidence work group.This pilot study sought to analyze the association between weight loss and inflammatory status as defined by SIRI.The cohort stems from a proof-of-concept pilot study conducted at one of the coordinating centers.Patients with pathologically confirmed metastatic pancreatic adenocarcinoma,treated from January 2020 to January 2023,were included.The index was calculated using the product of neutrophil and monocyte counts,divided by lymphocyte counts,obtained within 15 days before initiation chemotherapy.This study evaluated associations between overall survival(OS),SIRI and weight loss.RESULTS A total of 50 patients were included.66%of these patients were male and the median age was 66 years.Metastasis sites:36%liver,12%peritoneal carcinomatosis,10%lung,and 42%multiple locations.Regarding the first line palliative chemotherapy treatments:50%received gemcitabine plus nab-paclitaxel;28%,modified fluorouracil,leucovorin,irinotecan and oxaliplatin,and 16%were administered gemcitabine.42%had a weight loss>5%in the three months(mo)preceding diagnosis.21 patients with a SIRI≥2.3×10^(3)/L exhibited a trend towards a lower median OS compared to those with a SIRI5%weight loss before diagnosis,the median OS was 6 mo,in contrast to 19 mo for those who did not experience such weight loss(P=0.003).Patients with a weight loss>5%showed higher SIRI levels.This difference was statistically significant(P<0.000).For patients with a SIRI<2.3×10^(3)/L,those who did not

Background:Ocular surface disease in glaucoma patients is a significant ocular co-morbidity that can affect 40%to 59%of these patients worldwide.The current study was aimed at evaluating the potential clinical benefit of an intense pulsed light(IPL)-based treatment in glaucomatous patients with ocular surface disease due to prolonged hypotensive eyedrop treatments.To our knowledge,this is the first series analyzing the therapeutic effect of this treatment option in this type of patients.Methods:This non-comparative prospective case series study enrolled a total of 30 glaucoma patients ranging in age from 57 to 94 years old and treated with hypotensive eyedrops for years with dry eye symptomatology.All patients received four sessions of IPL treatment using the Optima IPL system(Lumenis,Yokneam,Israel)adjusted to the official optimized Lumenis setting.Changes in symptomatology,corneal staining,conjunctival hyperemia,non-invasive break-up time(NIBUT),tear osmolarity,tear meniscus height(TMH),meiboscore and meibomian gland expressibility was analyzed after treatment.Results:Statistically significant reductions were observed after IPL treatment in the symptomatology scores measured with different questionnaires[ocular surface disease index(OSDI),standard patient evaluation of eye dryness(SPEED)and symptom assessment questionnaire in dry eye(SANDE)]as well as with the visual analogue scale(P<0.001).Mean change in OSDI was−15.0±11.3.A significant reduction was found after treatment in the corneal staining score(P<0.001).A significant reduction was found in tear film meniscus height(P=0.012),as well as in tear film osmolarity(P=0.001).A significant reduction was also found in meibomian gland expressibility(P=0.003),changing the percentage of grade 3 eyes from 44.4%before IPL to 17.2%after treatment.Conclusions:IPL therapy combined with meibomian gland expression(MGX)seems to be an effective option to improve symptomatology in glaucomatous patients with ocular surface disease due to prolonged hypotensive eyedrop treatment

Nitric oxide(NO)is an essential reactive oxygen species and a signal molecule in plants.Although several studies have proposed the occurrence of oxidative NO production,only reductive routes for NO production,such as the nitrate(NO_(3)^(-))-upper-reductase pathway,have been evidenced to date in land plants.However,plants grown axenically with ammonium as the sole source of nitrogen exhibit contents of nitrite and NO3−,evidencing the existence of a metabolic pathway for oxidative production of NO.We hypothesized that oximes,such as indole-3-acetaldoxime(IAOx),a precursor to indole-3-acetic acid,are intermediate oxidation products in NO synthesis.We detected the production of NO from IAOx and other oximes catalyzed by peroxidase(POD)enzyme using both 4-amino-5-methylamino-2′,7′-difluorescein fluorescence and chemiluminescence.Flavins stimulated the reaction,while superoxide dismutase inhibited it.Interestingly,mouse NO synthase can also use IAOx to produce NO at a lower rate than POD.We provided a full mechanism for POD-dependent NO production from IAOx consistent with the experimental data and supported by density functional theory calculations.We showed that the addition of IAOx to extracts from Medicago truncatula increased the in vitro production of NO,while in vivo supplementation of IAOx and other oximes increased the number of lateral roots,as shown for NO donors,and a more than 10-fold increase in IAOx dehydratase expression.Furthermore,we found that in vivo supplementation of IAOx increased NO production in Arabidopsis thaliana wild-type plants,while prx33-34 mutant plants,defective in POD33-34,had reduced production.Our data show that the release of NO by IAOx,as well as its auxinic effect,explain the superroot phenotype.Collectively,our study reveals that plants produce NO utilizing diverse molecules such as oximes,POD,and flavins,which are widely distributed in the plant kingdom,thus introducing a long-awaited oxidative pathway to NO production in plants.This knowledge has essential implications for und

Sejima and Nishizawa have developed their work without explicitly referencing a theoretical discourse.However,they have stated that their housing projects describe their"architectural biography."This study presents the hypothesis that systermic theories had infu-enced their housing projects at a spatial and constructive Level.With the aim of testing it,the research produces four lines of results.Firstly,we observe that the word system is prominently current in a signifcant sample of their published texts.We describe how architects like Tomi-naga,Ito,Taki,and Shinohara,who infuenced Sejima when she was still a student,discussed this concept inspired by Christopher Alexander's texts.The second line brings together all the couple's single-family housing projects published between 1987 and 2010 for the first time in a single document,as well as an original graphic-constructive reconstruction of the most outstanding ones.Thirdly,we unveil the emergence of four constructive patterns by applying these theories to the graphic material developed.Finally,we summarize the results in dia-grams,showing the frequency,intensity,and continuity of the identified patterns in the four phases throughout the studied period.Verifying the hypothesis facilitates further research relate their domestic work and their other types of projects.

Cyclin-dependent kinases 4 and 6(CDK4/6)inhibitors demonstrated activity in terms of progression-free survival(PFS)in advanced dedifferentiated liposarcoma(DD-LPS),a sarcoma with CDK4 amplification.CDK4 overexpression is by far more common than amplification in sarcomas and it might be a rational target for CDK inhibitors.Preclinical investigators of this study found that CDK4 overexpression,while not of CDKN2A,was the most consistent predictive factor for palbociclib efficacy in sarcomas.Advanced adult-type soft-tissue sarcoma,excluding DD-LPS,or bone sarcoma patients,progressing after at least one systemic line,whose tumors overexpressed CDK4,but not CDKN2A at baseline biopsy,were accrued in this single-arm phase II trial(EudraCT number:2016-004039-19).With the main endpoint of a 6-month PFS rate,40%was considered promising in this population.Palbociclib was administered orally at 125 mg/day for 21 days in 28-day cycles.A total of 214 patients with 236 CDK4/CDKN2A determinations were assessed for prescreening,archival material(141),and screening,baseline biopsy(95).There were 28(29%)with favorable mRNA profiles from 95 screened patients at baseline.From 23 enrolled patients,21 evaluable,the 6-month PFS rate was 29%(95%CI 9–48),and there were 6 patients out of 21 with a PFS longer than 6 months.The median PFS and overall survival were 4.2(95%CI 3.6–4.8)and 12(95%CI 8.7–15.4)months,respectively.Translational research showed a significant correlation between CDK4 mRNA and protein expression.Palbociclib was active in a variety of sarcoma subtypes,selected by CDK4/CDKN2A,and deserves further investigation in the sarcoma context.

As for unmanned aircraft, the knowledge of the aircraft performance is directly related with the navigation, guidance, and control system programming. Therefore, the measured data in each phase of the flight must be sufficiently precise to obtain a good characterization of aircraft.This article proposes new methods of sending information to ground, which make it possible to know the aircraft behavior accurately, and for this purpose, four contributions have been made for ALO(Avión Ligero de Observación, Spanish acronym for Light Observation Aircraft). Currently, the characterization is based on data obtained at ten samples per second, insufficient to acquire detailed knowledge of what happened during the whole flight of an aircraft. As a result of these contributions, many more samples per second of accelerations and angular velocities are obtained at the most critical moments of the flight, such as takeoff or landing. Among the improvements included are data compression techniques, providing references to locate the measured data in time and identifying labels of each parameter.
