Fluorescence lifetime imaging of molecular rotors to map microviscosity in cells

Fluorescence liftime imaging （FLIM） of modified hydrophobic bodipy dyes that act as fluorescent molecular rotors shows that the fluorescence lifetime of these probes is a function of the microviscosity of their environment. Incubating cells with these dyes, we find a punctate and continuous distribution of the dye in cells. The viscosity value obtained in what appears to be endocytotic vesicles in living cells is around 100 times higher than that of water and of cellular ***-resolved fluorescence anisotropy measurements also yield rotational correlation times consistent with large microviscosity values. In this way, we successfully develop a practical and versatile approach to map the microviscosity in cells based on imaging fluorescent molecular rotors.

Indoor Air Quality(IAQ)has been an area of growing concern with the increasing knowledge of health hazards associated with contaminants,particularly in high occupancy buildings where residents may be exposed to high levels of nuisance dust and other *** and Energy in Environmental Design(LEED®)certification,which is awarded to buildings that prioritize sustainability and efficient resource use,has been increasingly sought in new *** LEED-certified buildings become more commonplace,it is worthwhile to consider whether these new building practices improve IAQ for its *** study compares particulate matter(PM)concentrations in 12 LEED-certified buildings to 12 analogous non-LEED certified buildings on the University of Utah ***-time air sampling was conducted in each building for PM measurements and a Wilcoxon signed rank test was conducted to compare PM levels.A statistically significant difference was found between LEED certification and PM concentrations,with LEED-certified buildings containing,on average,approximately half the PM of their non-LEED *** findings suggest that LEED certification is worth the financial investment,as it may lead to improved IAQ for ***,further research on other contaminants is warranted,including the characterization and comparison of formaldehyde and carbon dioxide levels.

In response to increasing concerns over climate change,soil health and wine quality,grape growers are seeking new practices(e.g.,biochar application)to minimize their environmental footprint while increasing productivity and the quality of their *** explore the potential of biochar-based amendments to achieve these goals in wine grape production,vineyard trials were established in the fall of *** Oregon sites were chosen with distinct soil types and climates(Willamette Valley and Rogue Valley)but planted with the same grapevine scion/rootstock Pinot noir *** treatments were applied under vines at each location:no biochar-no tillage(NT);no biochar+tillage(B0);18 tons ha^(−1)biochar+tillage(B18);35 tons ha^(−1)biochar+tillage(B35).In 2019,a suite of soil health,plant,and crop variables were measured,and wines were produced after *** incorporation of biochar modified the chemical and physical composition of soils at the two studied locations,increasing the bioavailability of carbon and nitrogen,their gravimetric water content and the concentration of plant available micro and macro *** responses of plant physiology parameters or productivity at either site were found after biochar incorporation when compared with ***,a significant and gradual decrease in the amount of wine tannins was found as a result of biochar application in wines produced from grapes from the Woodhall ***-term field experiments are required to assess the effects of biochar on soil properties,vine physiol-ogy,productivity,and grape and wine quality several years after incorporation.

Background TDP-43 proteinopathies represent a spectrum of neurological disorders,anchored clinically on either end by amyotrophic lateral sclerosis(ALS)and frontotemporal degeneration(FTD).The ALS-FTD spectrum exhibits a diverse range of clinical presentations with overlapping phenotypes,highlighting its *** study was aimed to use disease progression modeling to identify novel data-driven spatial and temporal subtypes of brain atrophy and its progression in the ALS-FTD *** We used a data-driven procedure to identify 13 anatomic clusters of brain volume for 57 behavioral variant FTD(bvFTD;with either autopsy-confirmed TDP-43 or TDP-43 proteinopathy-associated genetic variants),103 ALS,and 47 ALS-FTD patients with likely TDP-43.A Subtype and Stage Inference(SuStaIn)model was trained to identify subtypes of individuals along the ALS-FTD spectrum with distinct brain atrophy patterns,and we related subtypes and stages to clinical,genetic,and neuropathological features of *** SuStaIn identified three novel subtypes:two disease subtypes with predominant brain atrophy in either prefrontal/somatomotor regions or limbic-related regions,and a normal-appearing group without obvious brain *** limbic-predominant subtype tended to present with more impaired cognition,higher frequencies of pathogenic variants in TBK1 and TARDBP genes,and a higher proportion of TDP-43 types B,E and *** contrast,the prefrontal/somatomotor-predominant subtype had higher frequencies of pathogenic variants in C9orf72 and gRN genes and higher proportion of TDP-43 type *** normal-appearing brain group showed higher frequency of ALS relative to ALS-FTD and bvFTD patients,higher cognitive capacity,higher proportion of lower motor neuron onset,milder motor symptoms,and lower frequencies of genetic pathogenic *** overall SuStaIn stages also correlated with evidence for clinical progression including longer disease duration,higher King’s stage,and cog

The impact of climate change may be felt most keenly by tropical *** these taxa,it is argued,thermal specialization means a given shift in temperature will have a larger effect on *** species with limited dispersal ability,the impact of climate change depends on the capacity for their climate-relevant traits to *** shifts can occur through genetic adaptation,various forms of plasticity,or a combination of these *** we assess the extent and causes of shifts in 7 physiological traits in a tropical lizard,the rainforest sunskink(Lampropholis coggeri).Two populations were sampled that differ from each other in both climate and physiological *** compared trait values in each animal soon after field collection versus following acclimation to laboratory *** also compared trait values between populations in:(i)recently field-collected animals;(ii)the same animals following laboratory acclimation;and(iii)the laboratory-reared offspring of these *** results reveal high trait lability,driven primarily by acclimation and local *** contrast,developmental plasticity,resulting from incubation temperature,had little to no effect on most *** results suggest that,while specialized,tropical ectotherms may be capable of rapid shifts in climate-relevant traits.
