In 2013, a human influenza outbreak caused by a novel H7n9 virus occurred in China. Recently, the H7n9 virus acquired multiple basic amino acids at its hemagglutinin（HA） cleavage site, leading to the emergence of a highly pathogenic virus. The development of an effective diagnostic method is imperative for the prevention and control of highly pathogenic H7n9 influenza. Here, we designed and synthesized three pairs of primers based on the nucleotide sequence at the HA cleavage site of the newly emerged highly pathogenic H7n9 influenza virus. One of the primer pairs and the corresponding probe displayed a high level of amplification efficiency on which a real-time RT-PCR method was established. Amplification using this method resulted in a fluorescent signal for only the highly pathogenic H7n9 virus, and not for any of the H1–H15 subtype reference strains, thus demonstrating high specificity. The method detected as low as 39.1 copies of HA-positive plasmid and exhibited similar sensitivity to the virus isolation method using embryonated chicken eggs. Importantly, the real-time RT-PCR method exhibited 100% consistency with the virus isolation method in the diagnosis of field samples. Collectively, our data demonstrate that this real-time RT-PCR assay is a rapid, sensitive and specific method, and the application will greatly aid the surveillance, prevention, and control of highly pathogenic H7n9 influenza viruses.

Physical mechanical stimulation can maintain and even increase bone ***,we report an important role of osteocytic integrinα5 in regulating the anabolic response of bone to mechanical loading using an Itga5 conditional gene knockout(cKO)mouse ***α5 gene deletion increased apoptotic osteocytes and reduced cortical anabolic responses to tibial compression including decreased endosteal osteoblasts and bone formation,and increased endosteal osteoclasts and bone resorption,contributing to the decreased bone area fraction and biomechanical properties,leading to an enlarged bone marrow area in cKO *** disruption of anabolic responses to mechanical loading was also detected in cKO trabecular ***,integrinα5 deficiency impeded load-induced Cx43 hemichannel opening,and production and release of PGE2,an anabolic factor,resulting in attenuated effects of the loading on catabolic sclerostin(SOST)reduction and anabolicβ-catenin ***,this study shows an indispensable role of integrinα5 in osteocytes in the anabolic action of mechanical loading on skeletal tissue through activation of hemichannels and PGE2-evoked gene ***α5 could act as a potential new therapeutic target for bone loss,especially in the elderly population with impeded mechanical sensitivity.

Estrogen deficiency in postmenopausal women is a major cause of bone loss,resulting in osteopenia,osteoporosis,and a high risk for bone *** 43 (Cx43) hemichannels (HCs) in osteocytes play an important role in osteocyte viability,bone formation,and *** showed here that estrogen deficiency reduced Cx43 expression and HC *** determine if functional HCs protect osteocytes and bone loss during estrogen deficiency,we adopted an ovariectomy model in wild-type (WT) and two transgenic Cx43 mice:R76W (dominant-negative mutant inhibiting only gap j.nction channels) and Cx43 Δ130–136 (dominant-negative mutant compromising both gap j.nction channels and HCs).The bone mineral density (BMD),bone structure,and histomorphometric changes of cortical and trabecular bones after ovariectomy were *** results showed that the Δ130–136 transgenic cohort had greatly decreased vertebral trabecular bone mass compared to WT and R76W mice,associated with a significant increase in the number of apoptotic osteocyte and empty ***,osteoclast surfaces in trabecular and cortical bones were increased after ovariectomy in the R76W and WT mice,respectively,but not in Δ130–136 *** data demonstrate that impairment of Cx43 HCs in osteocytes accelerates vertebral trabecular bone loss and increase in osteocyte apoptosis,and further suggest that Cx43 HCs in osteocytes protect trabecular bone against catabolic effects due to estrogen deficiency.

目的探讨精神分裂症应用基于时效激励理论的团队绘画艺术疗法干预后,患者的自我效能、认知功能以及症状缓解情况。方法选取2022年1月至2023年1月聊城市第四人民医院就诊的精神分裂症患者(n=104)为本次探究对象,根据随机数字表法将入选患者分为观察组及对照组。对照组(52例)行常规护理干预,观察组(52例)在对照组的基础上行基于时效激励理论的团队绘画艺术疗法干预。观察两组患者的服药依从性、总自杀率、致死率;比较干预前后患者的自我效能评分(GSES)、症状缓解程度(BPRS)及认知功能评分。结果观察组患者的依从性为96.15%,明显高于对照组患者的82.69%,两者相较差异具有统计学意义(P0.05);干预后,两组患者的GSES、总应答数、正确应答数、完成分类数评分较干预前呈现增高趋势,BPRS、持续错误数评分有所降低,观察组优于对照组,差异具有统计学意义(P0.05)。与对照组患者的25.00%相比,观察组患者的总自杀率为9.62%,组间相较具有统计学意义(P<0.05)。结论基于时效激励理论的团队绘画艺术疗法能提高精神分裂症的服药依从性,患者的自我效能感明显提高,症状可快速得到缓解,认知功能明显改善,减少自杀风险,患者的致死率低,干预效果安全、有效。

A comprehensive dataset from 594 fracturing wells throughout the Duvernay Formation near Fox Creek, Alberta, is collected to quantify the influences of geological, geomechanical, and operational features on the distribution and magnitude of hydraulic fracturing-induced seismicity. An integrated machine learning-based investigation is conducted to systematically evaluate multiple factors that contribute to induced seismicity. Feature importance indicates that a distance to fault, a distance to basement, minimum principal stress, cumulative fluid injection, initial formation pressure, and the number of fracturing stages are among significant model predictors. Our seismicity prediction map matches the observed spatial seismicity, and the prediction model successfully guides the fracturing job size of a new well to reduce seismicity risks. This study can apply to mitigating potential seismicity risks in other seismicity-frequent regions.

由于受到轮轨激励的干扰,轮对轴承故障特征易被背景噪声所淹没,且在变转速模式下故障特征具有时变性,进一步加剧了提取难度。为此,提出一种基于快速谱平均峭度图的列车轮对轴承故障特征提取方法。根据频谱趋势将频谱划分为若干子带,采用Meyer小波构造带通滤波器,计算每个窄带滤波信号的平均峭度并构造快速谱平均峭度图;选取最优频带进行解调,消除背景噪声的影响;通过相位函数对滤波信号进行迭代广义解调,解决时变故障冲击间隔带来的频谱模糊问题。仿真和试验分析结果表明所提方法有效克服了强背景噪声和时变故障特征的干扰,为变转速工况下轮对轴承故障特征提取提供了新的解决策略。
