Effect of Freeze-Thaw and Urea in Solubility of GPC3-Csub Protein Expressed in Escherichia coli

Glypican-3 is a protein encoded by the Glypican-3 gene located on human X chromosome (Xq26), composed of two subunits, a 40 kDa n-terminal subunit, and a 30 kDa C-terminal subunit. Glypican-3 is a currently potential target molecule for liver cancer treatments because of its over-expression and growth effects on hepatocellular carcinoma (HCC). This study examined the expression and purification of a C-terminal subunit of Glypican-3 protein (GPC3-Csub) due to its application in both diagnosis and therapy for hepatocellular carcinoma. The gene encoding for GPC3-Csub was successfully cloned into plasmid pET28a fused with an affinity tag composed of six consecutive histidine residues (His-tag). Recombinant protein GPC3-Csub was expressed in Escherichia coli BL21 (DE3) in the condition of adding 3% ethanol with IPTG induction. GPC3-Csub was extracted using repeated freeze-thaw cycles with lysozyme, and inclusion bodies were solubilized by 8M Urea, SDS 10% in pH 12. His-tag fused GPC3-Csub proteins allowed it to be purified by affinity chromatography method using the nickel-nitrilotriacetic acid (ni-nTA) column. High expression of GPC3-Csub was confirmed by Coomassie staining and western-blot. GPC3-Csub could be isolated with a ni-nTA column and have a purity of about 90%.

目的:研究非糖尿病心力衰竭(Heart Failure,HF)患者应用达格列净治疗后血浆中血浆肾素浓度(Plasma renin concentration,PRC)、血浆血管紧张素Ⅱ(angiotensinⅡ,AnG-II)、血浆醛固酮(Aldosterone,ALD)的浓度水平变化情况,探讨达格列净对不伴糖尿病心力衰竭患者RAAS系统影响。方法:选取2021年7月至2022年10月于大理大学第一附属医院心内科收治102例非糖尿病心力衰竭患者为研究对象,44名常规治疗的患者纳入对照组,58名在常规治疗基础上口服达格列净的患者纳入实验组。记录患者一般情况,并分别在服药前及服药后第7天测定患者一般实验室指标(血尿酸、血糖、肌酐、总胆固醇、甘油三酯、血钾、血钠、低密度脂蛋白胆固醇)、BnP值、RAAS系统(PRC、AnG-II、ALD)、左室射血分数(LVEF)、心功能等级。记录两组患者基础资料以及治疗期间心衰症状、心肾功能及RAAS系统相关指标的变化情况并进行对比。结果:1.实验组患者nYHA心功能分级I级从1例(1.72%)经治疗后升到12例(20.68%);心功能II级从5例(8.62%)上升至45例(77.59%);心功能III级从40例(68.89%)降低0例(0.00%),心功能IV级从12例(20.68%)降到1例(1.72%),治疗前后心功能分级分布差异有统计学差异(Pna无显著变化(P>0.05)。治疗前后实验组患者均进行生命体征、Glu、TC、TG、Cr、UA,K、na复查未出现明显异常,未发生不良反应事件,说明达格列净治疗于心衰是安全的。3.治疗后实验组BnP值为(434.55±418.40)pg/ml,对照组为(619.33±453.37)pg/ml,实验组低于对照组,组间差异具有统计学意义(PnP平均下降(869.12±1143.25)pg/ml,对照组平均下降(276.79±740.19)pg/ml,组间差异具有统计学意义(PnP值显著下降,差异均具有统计学意义(P0.05);经治疗,实验组患者的PRC值显著下降至(11.58±4.77)pg/ml,差异具有统计学意义(P0.05);经治疗,实验组患者的ALD值显著下降至(65.47±12.01)pg/ml,差异具有统计学意义(P0.05)。比较两组患者治疗前后差值,实验组ALD(pg/ml)(19.05±20.16),与对照组(2.29±16.07)结果显示,实验组显著高于对照组具有统计学意义(PngⅡ(35.41±5.61)pg/ml,与对照组(33.21±7.47)pg/ml差异无统计学意义(P>0.05);经治疗,实验组与对照组均显著下降,治疗前后差异均具有统计学意义(PngⅡ(pg/ml)(14.49±5.87),与对照组(10.16±6.46)结果显示,实验组显著高于对照组具有统计学意义(Png-II、ALD浓度水平差异无统计学意义(P>0.05)。结论:1.达格列净可显著降低不伴糖尿病心力衰竭患者心功能分级和BnP值,且安全性佳。2.达格列净可显著降PRC、Ang-II、ALD浓度水平,且治疗效果不受性别和其他药物的影响。提示达格列净可能具有抑制RASS系统的作用。

Properties of poly（1-naphthylamine）/Fe3O4 composites and arsenic adsorption capacity in wastewater

The research results of poly（1-naphthylamine）lFe3O4 （PnA/Fe3O4） nanocomposites synthesized by a chemical method for As（Ⅲ） wastewater treatment are presented in this paper. XRD patterns and TEM images showed that the Fe3O4 grain size varied from 13 to 20 nm. The results of Raman spectral analysis showed that PnA participated in part of the PnA/Fe3O4 composite samples. The grain size of PnA/Fe3O4 composite samples is about 25-30 nm measured by SEM. The results of vibrating sample magnetometer measurements at room temperature showed that the saturation magnetic moment of PnA/Fe3O4 samples decreased from 63.13 to 43.43 emu/g, while the PnA concentration increased from 5% to 15%. The nitrogen adsorption-desorption isotherm of samples at 77 K at a relative pressure PIPo of about 1 was studied in order to investigate the surface and porous structure of nanoparticles by the BET method. Although the saturation magnetic moments of samples decreased with the polymer concentration increase, the arsenic adsorption capacity of the PnA/Fe3O4 sample with the PnA concentration of 5% is better than that of Fe3O4 in a solution with pH = 7. In the solution with pH 〉 14, the arsenic adsorption of magnetic nanoparticles is insignificant.
