Objective： To examine the effects of deltamethrin（DM） and tBhQ on the activation of Nrt2 and expression of GCS-hS in primary astrocytes. Methods： Rat primary astrocytes were treated with DM （10μmol/L） and tBhQ （40μmol/L）, for 1 and 6 h respectively. Results： Analysis using immunofluorescent confocal microscopy revealed that tBhQ treatment led to a dramatic increase in the level of Nrt2. The ratio of cytoplasmic Nrt2 protein to nuclear Nrt2 protein was markedly reduced in astrocytes by either tBhQ or DM treatment. The level of Nrf2 was reduced by combined tBhQ with DM treatment for 1 h, but treatment the ratio of cytoplasmic Nrf2 protein to nuclear Nrf2 protein was increased compared with that of the tBhQ only. The expression of GCS-hS in astrocytes was not altered by DM or tBhQ or both in astrocytes. Conclusion： These findings demonstrated for the first time that Nrf2 was activated by pesticides and antioxidants in astrocytes, implicating a tote of Nrf2 in response to pesticide neurotoxicity ingla.

Clinicopathological Features of Non-familial Colorectal Cancer with high-frequency Microsatellite Instability

Objective To explore the clinicopathological features of non-familial colorectal cancer with high-frequency microsatellite instability (MSI-h). Methods One hundred and fifty patients with colorectal cancer who had no family history were enrolled in this study from June 2006 to June 2008. Five standard microsatellite loci including BAT25, BAT26, D2S123, D5S346, and D17S250 were amplified with immunofluorescent polymerase chain reaction. The patient information including age, sex, and tumor location was recorded. Pathological features including differentiation, mucinous differentiation, histological heterogeneity, and Crohn's-like reaction were observed under light microscope. The presence of tumor-infiltrating lymphocytes (TLs, CD4+ and CD8+) was detected by means of immunohistochemistry. A regression equation was obtained by stepwise logistic regression analysis to evaluate the relationship between MSI-h phenotype in colorectal cancer ands pathological features. Results MSI-h phenotype occurred in 13.33% of the 150 patients with non-familial colorectal cancer. Poor differentiation, histological heterogeneity, Crohn's-like reaction, and presence of TLs were found to be independent factors to identify MSI-h non-familial colorectal cancer. Logistic regression equation showed an overall sensitivity of 70.0%, specificity of 99.2%, and accuracy of 95.3% in identifying MSI-h non-familial colorectal cancer. Conclusion MSI-h non-familial colorectal cancer manifests specific pathological features, which may be relied upon for effective identification of that disease.

Current DNA base editors contain nuclease and DNA deaminase that enables deamination of cytosine(C)or adenine(A), but no method for guanine(G) or thymine(T) editing is available at present. here we developed a deaminase-free glycosylase-based guanine base editor(gGBE) with G editing ability, by fusing Cas9 nickase with engineered N-methylpurine DNA glycosylase protein(MPG). By several rounds of MPG mutagenesis via unbiased and rational screening using an intron-split EGFP reporter, we demonstrated that gGBE with engineered MPG could increase G editing efficiency by more than 1500 ***, this gGBE exhibited high base editing efficiency(up to 81.2%) and high G-to-T or G-to-C(i.e. G-to-Y) conversion ratio(up to 0.95) in both cultured human cells and mouse embryos. Thus, we have provided a proof-of-concept of a new base editing approach by endowing the engineered DNA glycosylase the capability to selectively excise a new type of substrate.

In this report,a series of self-organized TiO_(2)nanotube arrays were prepared by anodization of titanium foil in mixed electrolytes composed of water,ethylene glycol,and Nh_(4)*** photoelectrochemical(PEC)performance as a photoanode was characterized by the PEC water-splitting hydrogen(h_(2))generation *** internal relationship between the TiO_(2)nanotube arrays(TNTAs)morphology and their PECperformance was thoroughly *** results show that when the etching time is 10 hours,the length of the as-prepared TNTAs is about 20.78μm and the measured photocurrent density is around 1.25 m A·cm^(-2)with applied bias voltage 0.6 V(***/Ag Cl)under simulated sunlight irradiation,which is 976 times higher than that of the TiO_(2)substrate without nanotubes architecture(0.00128 m A×cm^(-2)).More interestingly,the results of the IPCEmeasurement show that the band-gap of the as-prepared TNTAs is reduced from 3.20 to 2.83 *** corresponding optical response limit is also extended from 400 nm to TiO_(2)nanotube arrays is 510 nm,which indicates that the increasement of the TNTAs PEC performance benefits from the great improvement of its utilization of both the UVand visible light irradiation.

针对沽源460低品位复杂钼矿中钼矿物以微细脉状形式被褐铁矿所包裹,矿石细碎后泥化现象严重的问题,研究了以拌入高酸加温熟化破解铀、钼矿物的包裹体,并对处于硫化态的钼矿物进行氧化,进而实现难处理铀钼矿的高效浸出,再通过加入耐酸耐高温的天然粘合剂凹凸棒,解决了熟化后的矿石物料在筑堆浸出过程中遇水易泥化而导致渗透性差的问题。确定的最佳熟化条件为:矿石粒度-2.5 mm,熟化酸用量10%,黏合剂凹凸棒拌入量8%,熟化温度90℃,熟化时间24 h,铀渣计浸出率63.4%,钼渣计浸出率75.0%。

The development of life beyond planet Earth is a long-standing quest of the human race, but whether normal mammalian embryonic development can occur in space is still unclear. here, we show unequivocally that preimplantation mouse embryos can develop in space, but the rate of blastocyst formation and blastocyst quality are compromised. Additionally, the cells in the embryo contain severe DNA damage, while the genome of the blastocysts developed in space is globally hypomethylated with a unique set of differentially methylated regions. The developmental defects, DNA damage and epigenetic abnormalities can be largely mimicked by the treatment with ground-based low-dose radiation. however,the exposure to simulated microgravity alone does not cause major disruptions of embryonic development,indicating that radiation is the main cause for the developmental defects. This work advances the understanding of embryonic development in space and reveals long-term extreme low-dose radiation as a hazardous factor for mammalian reproduction.

采用高温固相法制备了一系列单掺或双掺Pr3+和Yb3+的GdBO3材料,分别测试分析了材料的物相结构和发光性质。在446 nm蓝光(Pr3+:3h4→3P2)激发下,检测到Yb3+的近红外特征发射,表明样品中存在Pr3+到Yb3+的能量传递。Pr3+的掺杂浓度一定时,样品的发光会随着Yb3+掺杂浓度的改变而发生变化。通过对比不同掺杂情况下Pr3+:3P0能级的衰减曲线,发现随着Yb3+的掺杂浓度的增加,该能级的荧光寿命不断缩短;同时利用不同条件下的衰减特性计算得出不同Yb3+掺杂浓度样品的能量传递效率。用Inokutihirayama模型分析表明Pr3+-Yb3+能量传递类型为偶极子-偶极子相互作用。

Background:Pulmonary hypertension(Ph)is a progressive disease characterized by lung endothelial cell dysfunction and Vascular *** progenitor cells(EPCs)have been proved to be a potential therapeutic strategy to treat *** has been found to be protective to hypoxia-induced *** this study,we applied high shear stress(hSS)-induced Ph,and examined whether EPCs confer resistance against hSS-induced Ph through ***:Pulmonary microvascular endothelial cells(PMVECs)were cultured under hSS with pro-inflammatory factors in an artificial capillary system to mimic the Ph *** of p62,a selective autophagy substrate,were quantified by western *** viability was determined by trypan blue exclusion ***:The p62 level in PMVECs was increased at 4 hours after hSS,peaked at 12 hours and declined at 24 *** cell viability gradually *** with PMVECs cultured by empty medium,in cells cultured by EPC-conditioned medium(EPC-CM),the cell viability was significantly higher;however,p62 levels were also significantly higher,suggesting inhibition of autophagy by *** choloquine to suppress autophagy decreased the cell viability of PMVECs under ***:EPC-CM could suppress the autophagic activity of PMVECs in hSS-induced ***,suppression of autophagy leads to ceil *** could fight against Ph through cellular or molecular pathways independent of *** it is not proved if induction of autophagy could be a potential strategy to treat hSS-induced Ph as hypoxia-induced Ph.

Lanthanum coordination compound with l-alanine[La2（ala）4（h2O）8]·（ClO4）6（ala=l-alanine） was synthesized by reaction of lanthanum perchlorate with l-alanine in aqueous solution and obtained in the form of single *** X-ray crystal structure has been determined and the IR spectrum has been *** title compound Cl6La2C12h44O40N4,Mr=1375.2,crystallizes in the triclinic,space group P1 , with parameters: a= 11.227（3）,b=11.445（2）,c=11.014（3）,α=114.46（2）,β=114.51（2）,γ=78.62（2）°,V=1171.73,Z=1,Dc=1.95 g/cm￣3,F（000）=680,μ=22.9 cm￣-1（MoKα）.The final R=0.050,Rω=*** the crystal,two lanthanum ions are coordinated by four alanine molecules via four carboxylate bridges to form a dinuclear coordination *** water molecules are coordinated to each lanthanum ion.
