Sphingosine-1-phosphate(S1P)is an important lipid mediator that regulates a diverse range of intracellular cell signaling pathways that are relevant to tissue engineering and regenerative ***,the precise function of S1P in dental pulp stem cells(DPSCs)and its osteogenic differentiation remains *** here investigated the function of S1P/S1P receptor(S1PR)-mediated cellular signaling in the osteogenic differentiation of DPSCs and clarified the fundamental signaling *** results showed that S1P-treated DPSCs exhibited a low rate of differentiation toward the osteogenic phenotype in association with a marked reduction in osteogenesis-related gene expression and AKT *** note,both S1PR1/S1PR3 and S1PR2 agonists significantly downregulated the expression of osteogenic genes and suppressed AKT activation,resulting in an attenuated osteogenic capacity of *** importantly,an AKT activator completely abrogated the S1P-mediated downregulation of osteoblastic markers and partially prevented S1P-mediated attenuation effects during ***,the pro-inflammatory TNF-αcytokine promoted the infiltration of macrophages toward DPSCs and induced S1P production in both DPSCs and *** findings indicate that the elevation of S1P under inflammatory conditions suppresses the osteogenic capacity of the DPSCs responsible for regenerative endodontics.

Background Sarcopenia is a risk factor for metabolic disorders and cardiovascular disease, but the association between sarcopenia and cardiovascular risk factors according to age and obesity status in the general population remains unknown. We thus investigated these associations in the Korean population. Methods We included 8,958 and 8,518 subjects from the fifth Korean National Health and Nutrition Examination Survey （KNHANES）（from 2010 and 2011, respectively）. The study was restricted to participants 〉 20 years old who had completed the health examination survey, including whole body dual-energy X-ray absorptiometry scans. After exclusion, 7,366 subjects （3,188 men, 4,178 women） were included in our final analysis. Age was categorized according to three age groups （20-39, 40-59, mad 〉 60 years）, and subjects were catego- rized according to their sarcopenic and obesity status. Cardiovascular risk was assessed with Framingham risk score （FRS）. Results The sarco- penic obese group had a higher FRS than the non-sarcopenic obese group, which had a higher FRS than the non-sarcopenic non-obese group. Age-wise, the 20-39 year-old group showed a non-significant association between sarcopenia and FRS. In the 40-59 year-old group, regardless of obesity status, sarcopenic subjects had a higher FRS than non-sarcopenic subjects. In the 〉 60 year-old group, sarcopenic subjects had a higher FRS than non-sarcopenic subjects for the non-obese group. Conclusions Sarcopenia was associated with cardiovascular disease and may be an early predictor of its susceptibility in both elderly and middle-aged subjects. Thus, management of sarcopenia is necessary to prevent cardiovascu- lar disease.

The NOD-,LRR-,and pyrin domain-containing protein 3(NLRP3)inflammasome is a multiprotein complex involved in the release of mature interleukin-1βand triggering of pyroptosis,which is of paramount importance in a variety of physiological and pathological *** the past decade,considerable advances have been made in elucidating the molecular mechanisms underlying the priming/licensing(Signal 1)and assembly(Signal 2)involved in NLRP3 inflammasome ***,a number of studies have indicated that the priming/licensing step is regulated by complicated mechanisms at both the transcriptional and posttranslational *** this review,we discuss the current understanding of the mechanistic details of NLRP3 inflammasome activation with a particular emphasis on protein-protein interactions,posttranslational modifications,and spatiotemporal regulation of the NLRP3 inflammasome *** also present a detailed summary of multiple positive and/or negative regulatory pathways providing upstream signals that culminate in NLRP3 inflammasome complex assembly.A better understanding of the molecular mechanisms underlying NLRP3 inflammasome activation will provide opportunities for the development of methods for the prevention and treatment of NLRP3 inflammasome-related diseases.

Iron sulfides have bee n con sidered as one of the most promising can didates for sodium ion battery anode materials due to their high theoretical capacity and low cost. I n this work, spin dle-like Fe7S8 with n itroge rvdoped carb on (FeiS/N-C) nano hybrids are successfully syn thesized via a solvothermal method by sulfidation iron-based metal organic framework (FeMOF). As sodium ion battery an odes, FeySs/N-C nano hybrids exhibit high reversible capacity of 450.8 mAh g-1 at 200 mA·g^-1, and 406.7 mAh·g^-1 at 500 mA·g^-1 even after 500 cycles. They also show excellent rate properties and delivering the capacity of 327.8 mAh·g^-1 at a very high current density of 3.2 A·g^-1. These outstanding electrochemical performa nces can be attributed to the unique structure of Fe7S8/ N-C nan ohybrids. The nano scale dime nsion in their size can be ben eficial for facile ion and electro n tran sports. Furthermore, the stable n itroge n doped carb on frameworks can also improve electrical conductivity and relieve the problems related to volume expansion. X-ray absorption spectroscopy and X-ray photoelectron spectroscopy analyses have been performed to study reactions occurred in spindle-like FeySs/N-C nanohybrid electrode at both bulk and surface.

Inflammatory fibroid polyp(IFP) is a rare benign lesion of the gastrointestinal tract. We report a case of computed tomography(CT) imaging finding of a gastric IFP with massive fibrosis. CT scans showed thickening of submucosal layer with overlying mucosal hyperenhancement in the gastric antrum. The submucosal layer showed increased enhancement on delayed phase imaging. An antrectomy with gastroduodenostomy was performed because gastric cancer was suspected, particularly signet ring cell carcinoma. The histopathological diagnosis was an IFP with massive fibrosis. The authors suggest that when the submucosal layer of the gastric wall is markedly thickened with delayed enhancement and preservation of the mucosal layer, an IFP with massive fibrosis should be considered in the differential diagnosis.
