Dissimilarity nitrate reduction to ammonium(DNRA)is of significance in agriculture ecosystems as the process is beneficial to N retention in ***,how fertilization regimes influence DNRA rates and functional microbes in agriculture was rarely *** the present study,a 2-year pot experiment was conducted in two contrasting paddy soils to evaluate the effects of straw and nitrogen addition on DNRA process and the related functional microbes,using stable isotope tracer and molecular ecology *** results showed that the abundance and transcription activity of nitrite reductase encoding gene(nrfA)involved in DNRA process and DNRA rates were significantly higher in alkaline soils than in acidic *** incorporation significantly enhanced nrfA gene abundance and transcription activity,with a greater effect in alkaline soil than in acidic *** rates of DNRA,abundance and transcription activity of nrfA gene positively correlated to soil C/N and C/NO_(3)^(-) induced by straw *** analysis based on nrfA gene transcript showed that Deltaproteobacteria was the most dominant group in both soil types(30.9%-67.4%),while Gammaproteobacteria,Chloroflexi,Actinobacteria were selectively enriched by straw *** results demonstrated that DNRA activity can be improved by straw return practice in paddy soils while the effect will vary among soil types due to differentiated functional microbial communities and edaphic properties.

Genetic Analysis and Molecular Mapping of Novel White Striped Leaf Mutant Gene in Rice

A new white striped leaf mutant wsll was discovered from Nipponbare mutated by ethyl methanesulfonate. The mutant showed white striped leaves at the seedling stage and the leaves gradually turned green after the tillering stage. The chlorophyll content of wsll was significantly lower than that of wild-type during the fourth leaf stage, tillering stage and booting stage. The numbers of chloroplast, grana and grana lamella were reduced and the thylakoids were degenerated in wsll compared with wild type. Genetic analysis showed that the wsll was controlled by a single recessive gene. Molecular mapping of the wsll was performed using an F2 population derived from wsll/Nanjing 11. The wsll was finally mapped on the telomere region of chromosome 9 and positioned between simple sequence repeat markers RM23742 and RM23759 which are separated by approximately 486.5 kb. The results may facilitate map-based cloning of wsll and understanding of the molecular mechanism of the regulation of leaf-color by WSL1 in rice.

Load-induced error identification of hydrostatic turntable and its influence on machining accuracy

In heavy duty machine tools, hydrostatic turntable is often used as a means for providing rotational motion and supporting workpiece, so the accuracy of turntable is crucial for part machining. In order to analyze the influence of load-indcued errors on machining accuracy, an identification model of load-induced errors based on the deformation caused by applied load of hydrostatic turntable of computerized numerical control(CNC) gantry milling heavy machine is proposed. Based on multi-body system theory and screw theory, the space machining accuracy model of heavy duty machine tool is established with consideration of identified load-induced errors. And then, the influence of load-induced errors on space machining accuracy and the roundness error of a milled hole is analyzed. The analysis results show that load-induced errors have a big influence on the roundness error of machined hole, especially when the center of the milled hole is far from that of hydrostatic turntable.

Micromechanical modeling of hollow cylinder torsional shear test on sand using discrete element method

Previous studies on the hollow cylinder torsional shear test (HCTST) have mainly focused on the macroscopic behavior, while the micromechanical responses in soil specimens with shaped particles have rarely been investigated. This paper develops a numerical model of the HCTST using the discrete element method (DEM). The method of bonded spheres in a hexagonal arrangement is proposed to generate flexible boundaries that can achieve real-time adjustment of the internal and external cell pressures and capture the inhomogeneous deformation in the radial direction during shearing. Representative angular particles are selected from Toyoura sand and reproduced in this model to approximate real sand particles. The model is then validated by comparing numerical and experimental results of HCTSTs on Toyoura sand with different major principal stress directions. Next, a series of HCTSTs with different combinations of major principal stress direction (a) and intermediate principal stress ratio (b) is simulated to quantitatively characterize the sand behavior under different shear conditions. The results show that the shaped particles are horizontally distributed before shearing, and the initial anisotropic packing structure further results in different stress-strain curves in cases with different a and b values. The distribution of force chains is affected by both a and b during the shear process, together with the formation of the shear bands in different patterns. The contact normal anisotropy and contact force anisotropy show different evolution patterns when either a orb varies, resulting in the differences in the non-coaxiality and other macroscopic responses. This study improves the understanding of the macroscopic response of sand from a microscopic perspective and provides valuable insights for the constitutive modeling of sand. Tetrasphaera have been recently identified based on the 16S ribosomal RNA(rRNA)gene as among the most abundant polyphosphate-accumulating organisms(PAOs)in global full-scale wastewater treatment plants(WWTPs)with enhanced biological phosphorus removal(EBPR).However,it is unclear how Tetrasphaera PAOs are selectively enriched in the context of the EBPR *** this study,an EBPR microbiome enriched with Tetrasphaera(accounting for 40%of 16S sequences on day 113)was built using a top-down design approach featuring multicarbon sources and a low dosage of *** microbiome showed enhanced nutrient removal(phosphorus removal~85%and nitrogen removal~80%)and increased phosphorus recovery(up to 23.2 times)compared with the seeding activated sludge from a local full-scale *** supply of 1 mg·L^(-1)allylthiourea promoted the coselection of Tetrasphaera PAOs and Microlunatus PAOs and sharply reduced the relative abundance of both ammonia oxidizer Nitrosomonas and putative competitors Brevundimonas and Paracoccus,facilitating the establishment of the EBPR *** on 16S rRNA gene analysis,a putative novel PAO species,EBPR-ASV0001,was identified with Tetrasphaera japonica as its closest *** study provides new knowledge on the establishment of a Tetrasphaera-enriched microbiome facilitated by allylthiourea,which can be further exploited to guide future process upgrading and optimization to achieve and/or enhance simultaneous biological phosphorus and nitrogen removal from high-strength wastewater.
