An early-morning gene network controlled by phytochromes and cryptochromes regulates photomorphogenesis pathways in Arabidopsis

Light perception at dawn plays a key role in coordinating multiple molecular processes and in entraining the plant circadian *** Arabidopsis mutant lacking the main photoreceptors,however,still shows clock entrainment,indicating that the integration of light into the morning transcriptome is not well *** this study,we performed a high-resolution RNA-sequencing time-series experiment,sampling every 2 min beginning at *** parallel experiments,we perturbed temperature,the circadian clock,photoreceptor signaling,and chloroplast-derived light *** used these data to infer a gene network that describes the gene expression dynamics after light stimulus in the morning,and then validated key *** sampling time points at high density,we are able to identify three light-and temperature-sensitive bursts of transcription factor activity,one of which lasts for only about 8 *** and cryptochrome mutants cause a delay in the transcriptional bursts at dawn,and completely remove a burst of expression in key photomorphogenesis genes(HY5 and BBX family).Our complete network is available online(http://***/∼de656/dawnBurst/***).Taken together,our results show that phytochrome and cryptochrome signaling is required for fine-tuning the dawn transcriptional response to light,but separate pathways can robustly activate much of the program in their absence.

Objective:To carry out a phyto-chemical characterization of essential oil from Ocimum basilicum L.(***) harvested in dR Congo and to assess the antioxidant potential of crude extracts with respect to the polarity for comparison ***:The phyto-chemical characterization of essential oil produced by hydrodistillation was performed by coupled gas chromatography-mass spectrometer analysis and the antioxidant potential evaluation by in vitro 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity ***:A previously weighed amount of fresh leaves of *** produced 0.65%of essential oil that led to the identification of a set of 84.44%out of 99.98%as major compounds(> 1.5%).The chemo-type of this essential oil was linalool-methyl *** components of oil were characterized by oxygenated aromatic hydrocarbons(46.00%) and oxygenated monoterpenes(26.75%).With respect to the amount of components,methyl chavicol also known as estragole(35.72%) constituted the very large quantity afterward linalool(21.25%) and then epi-α-cadinol(8.02%),α-bergamotene(6.56%),eugenol(4.60%),1,8-cineole(4.04%),germacrene d(2.06%),thymol(1.64%),and(E)-citral(1.55%),*** oil exhibited antioxidant potential and IC_(50) =(1.180 ± 0.015) mg/***-polar crude extracts yields were low compared to the one of polar *** methanol and ethyl acetate had considerably manifested antioxidant potential with IC_(50) values equal to(0.025 ± 0.013) mg/mL and(0.085 ± 0.012) mg/mL,*** concerns to IC50 values,essential oil was less active than methanol and ethyl acetate *** methanol crude extract exhibited the highest ***-polar extracts showed insignificant radical scavenging ability that did not allow assessing IC50 *** results highlighted the occurrence of antioxidant potential compounds in polar ***:Essential oil and crude extracts of *** growing in dR Congo can be advocated as nat

Transcription factor TFIIA is controlled by complex regulatory networks including proteolysis by the protease Taspase 1, though the full impact of cleavage remains elusive. Here, we demonstrate that in contrast to the general assumption,de novoproduced TFIIA is rapidly confined to the cytoplasm via an evolutionary conserved nuclear export signal (NES, amino acids21VINdVRdIFL30), interacting with the nuclear export receptor Exportin-1/chromosomal region maintenance 1 (Crm1). Chemical export inhibition or genetic inactivation of the NES not only promotes TFIIA’s nuclear localization but also affects its transcriptional activity. Notably, Taspase 1 processing promotes TFIIA’s nuclear accumulation by NES masking, and modulates its transcriptional activity. Moreover, TFIIA complex formation with the TATA box binding protein (TBP) is cooperatively enhanced by inhibition of proteolysis and nuclear export, leading to an increase of the cell cycle inhibitor p16INK, which is counteracted by prevention of TBP binding. We here identified a novel mechanism how proteolysis and nuclear transport cooperatively fine-tune transcriptional programs.
