Computational Investigation of the Substituent Effect on the Intramolecular Proton Transfer Reaction of 3-Hydroxytropolone

The effects of substituent type and position on the proton transfer reaction of 3-hydroxytropolone(3-OHTRN) have been investigated theoretically by using density functional theory at the level of B3LYP/ 6-31+G** method. The influence of solvent on the proton transfer reactions of substituted 3-OHTRN has been examined using the self-consistent isodensity polarized continuum model(SCI-PCM) in water. As a result, while the proton transfer reaction is kinetically the easiest by substitution on position 3 of-NH2 group in the gas phase, it is kinetically the easiest by substitution on position 5 of the same group in water. In addition, these reactions are either kinetically or thermodynamically easier in the gas phase than that in water, except the reaction of structure with-NH2 group at position 6.

RNA targets of TDP-43: Which one is more important in neurodegeneration?

Nuclear loss and cytoplasmic aggregation of TDP-43 is the most common shared pathological feature of fronto-temporal dementia(FTD)and amyotrophic lateral scle-rosis(ALS),a motor neuron disease also known as Lou Gehrig’s disease[1].TDP-43 pathology is found in 97%of ALS cases,approximately 45%of FTD cases,and in some other neurodegenerative diseases such as Alzheimer’s disease[2].However,the direct mechanisms by which TDP-43 dysregulation contributes to neurodegenera-tion remain largely ***-43 is an RNA-binding protein with multiple roles in RNA metabolism,includ-ing transcription,splicing,transport,and the localization and stability of its target mRNAs,as well as in microRNA biogenesis[3].TDP-43 has thousands of RNA targets,but it is largely unknown which targets,if any,directly contribute to *** is it known whether any of them can be either manipulated as therapeutic targets to influence disease progression or,if not,could serve as biomarkers of TDP-43 pathology.

ddPCR在儿童异基因造血干细胞移植后EB病毒感染监测中的初步应用

目的本研究应用微滴式数字PCR(droplet digital PCR,ddPCR)检测儿童异基因造血干细胞移植术(allogeneic hematopoietic stem cell transplantation,alloHSCT)术后EB病毒(Epstein-Barr virus,EBV)感染的情况,初步探讨其在临床中作为alloHSCT移植术后EBV感染监测的补充检测方法的可行性。方法收集47例alloHSCT术后儿童共290份外周血样本,使用ddPCR和实时定量PCR(real-time quantitative PCR,qPCR)分别检测血浆中EBV DNA载量,比较两者的检出效能。通过对其中39例患儿进行连续检测,观察EBV DNA载量的动态变化,并分析两种方法的优劣势。结果ddPCR的EBV阳性检出率显著高于qPCR(χ^(2)=20.25,P<0.001),尤其在低病毒载量样本中表现更佳。在连续监测EBV DNA期间,39例患儿中有14例出现ddPCR阳性结果,其中有2例在ddPCR检测阳性、qPCR检测阴性的同时出现了皮疹和发热,提示ddPCR在早期发现EBV感染方面更为敏感。结论ddPCR在alloHSCT术后患儿EBV感染监测中具有一定优势,尤其是对于低病毒载量的样本。然而,由于该技术在临床应用中尚处于探索阶段,仍需更多的研究和实践来证实ddPCR的应用价值。

Evaluation of nintedanib as a new postoperative antiscarring agent in experimental extraocular muscle surgery

AIM:To investigate the efficacy of nintedanib on reducing postoperative inflammation,fibrosis and adhesion formation following extraocular muscle surgery in rabbits in comparison with triamcinolone acetonide(TA).METHODS:Reinsertion of superior rectus muscle in right eyes of 30 New Zealand white rabbits were *** were randomized to receive one of the following treatments:0.9%normal saline,one of 1-,5-,and 10μmol doses of nintedanib subconjunctivally immediately after surgery and on postoperative day 1,2,3,5,and 7,and TA immediately after *** a control group,unoperated left eyes(n=6)were *** the 28 th day,six eyes from each group were enucleated and histopathologically and immunohistochemically analyzed to assess the postoperative inflammatory changes,fibrosis and *** growth factor beta,matrix metalloproteinase-2 and alpha smooth muscle actin expressions were ***:Conjunctival and scleral inflammation in TA and nintedanib groups were significantly reduced compared to saline(sham)*** vascularity and rectus muscle fibrosis were significantly reduced in 10μmol nintedanib *** groups were the most effective groups in reduction of perimuscular *** three nintedanib groups nor TA group differed statistically from sham group with regard to *** expressions of transforming growth factor beta,alpha smooth muscle actin and matrix metalloproteinase-2 were reduced in nintedanib groups compared to saline ***:Nintedanib appears to attenuate postoperative inflammation and fibrosis after extraocular muscle *** may be a safer and stronger alternative agent in extraocular muscle surgery when compared to *** investigation is needed to prove antiadhesive effect of nintedanib.

Extracellular matrix remodelling in dental pulp tissue of carious human teeth through the prism of single-cell RNA sequencing

Carious lesions are bacteria-caused destructions of the mineralised dental tissues,marked by the simultaneous activation of immune responses and regenerative events within the soft dental pulp *** major molecular players in tooth decay have been uncovered during the past years,a detailed map of the molecular and cellular landscape of the diseased pulp is still *** this study we used single-cell RNA sequencing analysis,supplemented with immunostaining,to generate a comprehensive single-cell atlas of the pulp of carious human *** data demonstrated modifications in the various cell clusters within the pulp of carious teeth,such as immune cells,mesenchymal stem cells(MSC)and fibroblasts,when compared to the pulp of healthy human *** immune response in the carious pulp tissue is accompanied by specific changes in the fibroblast and MSC *** changes include the upregulation of genes encoding extracellular matrix(ECM)components,including COL1A1 and Fibronectin(FN1),and the enrichment of the fibroblast cluster with *** incremental changes in the ECM composition of carious pulp tissues were further confirmed by immunostaining *** of the Fibronectin fibres under mechanical strain conditions showed a significant tension reduction in carious pulp tissues,compared to the healthy *** present data demonstrate molecular,cellular and biomechanical alterations in the pulp of human carious teeth,indicative of extensive ECM remodelling,reminiscent of fibrosis observed in other *** comprehensive atlas of carious human teeth can facilitate future studies of dental pathologies and enable comparative analyses across diseased organs.

A Study on the Evaluation of Heating and Cooling Energy Efficiency in Residential Buildings for Climatic Zones of Turkey

In Turkey, most of the common type projects of mass production residential buildings are being developed and constructed by TOK1 （Housing Development Administration of Turkey）. These buildings, in which energy efficient approach has been disregarded for years, cause to gradually increase on heating and cooling energy consumptions. In regards to national economics, it is essential to evaluate energy efficiency and to develop suggestions to decrease energy consumptions in residential buildings. To achieve appropriate solutions, cost evaluation also becomes necessary. Therefore, this paper aims to introduce a study which serves the purpose of producing a choice of energy efficient solutions in order to reduce energy consumptions and energy cost. In this study, different heating and cooling energy efficient scenarios have been developed for a selected residential building, constructed by TOKI, for climatic zones of Turkey. For each scenario, energy simulations have been executed by means of the simulation program--DesignBuilder, the user-friendly visual interface of EnergyPlus, and cost analysis has been carried out by using the net present value and discounted payback period method. As a result, energy and cost effective solutions have been presented and discussed for different climatic zones.

Thromboembolism is an important complication in children undergoing therapy for ALL as it has the potential to impact adversely on both their survival and quality of life. The incidence of thrombosis in children with ALL varies between 1.1% and 36.7% and the actual mean is 3.2%. The aim of our study is to review the available reported data on the effect of FVL on thrombotic risk in pediatric patients with acute leukemia.

Energy and Cost Analyses of Solar Photovoltaic (PV) Microgeneration Systems for Different Climate Zones of Turkey

The environmental and energy problems that have arisen in Turkey because of the dramatically increase in energy consumption require the implementation of energy efficiency and microgeneration measures in the building sector which is the main sector of primary energy consumption. Since Turkey is highly dependent on exported energy resources, the basic energy policy approach is based on providing the supply security. In this regard, supporting for in situ energy production, encouraging the use of renewable energy sources and the systems such as microgeneration systems in order to meet the energy requirements of buildings would be considered as a key measure for resolving the energy related challenges of Turkey and dealing with the sustainability issues. Turkey’s geographical location has several advantages for extensive use of most of the renewable energy sources such as especially solar energy. However, this huge solar energy potential is not being used sufficiently in residential building sector which is responsible for the great energy consumption of Turkey. Therefore, this paper aims to introduce a study which investigates, on a life cycle basis, the environmental and the economic sustainability of solar Photovoltaic (PV) microgenerators to promote the implementation of this system as an option for the renovation of existing residential buildings in Turkey. In this study, main parameters which were related to the distribution of modules to be installed in flat roofs and facades and the evaluation of the PV systems were taken into account. The effect of these parameters on energy generation of PV systems was analyzed in a case study considering different climate zones of Turkey;and the decrease in the existing energy consumption of the reference building was calculated.
