Risk factors for hepatocellular carcinoma in patients with drug-resistant chronic hepatitis B

AIM:To investigate the risk factors and characteristics of hepatocellular carcinoma(HCC) in the patients with drug-resistant chronic hepatitis B(CHB).METHODS:A total of 432 patients with drug-resistant CHB were analyzed retrospectively from January 2004to December 2012. The patients were divided into two groups:the HCC group(n = 57) and the non-HCC group(n = 375). Two groups compared using logistic regression for various patients and viral characteristics in order to identify associated risk factors for ***,patient and tumor characteristics of HCC patients with na ve CHB(N group,n = 117) were compared to the HCC group(R group,n = 57) to identify any difference in HCC characteristics between ***:A significant difference was found for age,platelet count,alpha-fetoprotein(AFP),positivity of HBeAg,seroconversion rate of HBeAg,virologic response,the Child-Pugh score,presence of rtM204I,and the duration of antiviral treatment in non-HCC and HCC group. Cirrhosis,age(> 50 years),HBeAg(+),virologic non-responder status,and rtM204I mutants were independent risk factors for the development of HCC. The R group had lower serum C-reactive protein(CRP) and AFP levels,earlier stage tumors,and a shorter mean tumor surveillance period than the N group. However,the total follow-up duration was not significantly different between the two ***:13.2% of patients with drug-resistant CHB developed HCC. Age,cirrhosis,YIDD status,HBeAg status,and virologic response are associated with risk of HCC. Patients with drug-resistant CHB and these clinical factors may benefit from closer HCC surveillance.

Application of PCR primer sets for detection of <i>Pseudomonas</i>sp. functional genes in the plant rhizosphere

Plant growth promoting pseudomonads play an important role in disease suppression and there is considerable interest in development of bio-marker genes that can be used to monitor these bacteria in agricultural soils. Here, we report the application ofa PCR primer sets targeting genes encoding the main antibiotic groups. Distribution of the genes was variably distributed across type strains of 28 species with no phylogenetic groupingfor the detected antibioticsgenes, phlD for 2,4-diacetylphloroglucinol (2,4-DAPG) and phzCD for phenazine-1-carboxylic acid or hcnBC for hydrogen cyanide production. Analysis of field soils showed that primer sets for phlD and phzCD detected these genes in a fallowed neutral pH soil following wheat production, but that the copy numbers were below the detection limits in bulk soils having an acidic pH. In contrast, PCR products for the phzCD, pltc and hcnBc genes were detectable in mature root zones following plantingwith wheat. The ability to rapidly characterize populations of antibiotics producers using specific primer sets will improve our ability to assess the impacts of management practices on the functional traits of Pseudomonas spp. populations in agricultural soils.

Generation of tumor spheroids using a droplet-based microfluidic device for photothermal therapy

Despite their simplicity,monolayer cell cultures are not able to accurately predict drug behavior in vivo due to their inability to accurately mimic cell-cell and cell-matrix *** contrast,cell spheroids are able to reproduce these interactions and thus would be a viable tool for testing drug ***,the generation of homogenous and reproducible cell spheroids on a large scale is a labor intensive and slow process compared to monolayer cell ***,we present a droplet-based microfluidic device for the automated,large-scale generation of homogenous cell spheroids in a uniform *** the microfluidic system,the size of the spheroids can be tuned to between 100 and 130μm with generation frequencies of 70 *** demonstrated the photothermal therapy(PTT)application of brain tumor spheroids generated by the microfluidic device using a reduced graphene oxide-branched polyethyleneimine-polyethylene glycol(rGO-BPEI-PEG)nanocomposite as the PTT ***,we generated uniformly sized neural stem cell(NSC)-derived neurospheres in the droplet-based microfluidic *** also confirmed that the neurites were regulated by ***,this droplet-based microfluidic device could be a powerful tool for photothermal therapy and drug screening applications.

Sequence Information on Simple Sequence Repeats and Single Nucleotide Polymorphisms through Transcriptome Analysis of Mungbean

Mungbean （Vigna radiata （L.） Wilczek） is a unique species in its ability to fix atmospheric nitrogen, with early maturity, and relatively good drought resistance. We used 454 sequencing technology for transcriptome sequencing. A total of 150 159 and 142 993 reads produced 5 254 and 6 374 large contigs （〉_500 bp） with an average length of 833 and 853 for Sunhwa and Jangan, respectively. Functional annotation to known sequences yielded 41.34% and 41.74% unigenes for Jangan and Sunhwa. A higher number of simple sequence repeat （SSR） motifs was identified in Jangan （1 630） compared with that of Sunhwa （1 334）. A similar SSR distribution pattern was observed in both varieties. A total of 8 249 single nucleotide polymorphisms （SNPs） and indels with 2 098 high-confidence candidates were identified in the two mungbean varieties. The average distance between individual SNPs was -860 bp. Our report demonstrates the utility of transcriptomic data for implementing a functional annotation and development of genetic markers. We also provide large resource sequence data for mungbean improvement programs.

Excessive 5-year weight gain predicts metabolic syndrome development in healthy middle-aged adults

AIM:To quantitatively examine the impacts of an easyto-measure parameter-weight gain-on metabolic syndrome development among middle-aged adults. METHODS:We conducted a five-year interval observational study.A total of 1384 middle-aged adults not meeting metabolic syndrome(MetS)criteria at the initial screening were included in our *** data such as MetS-components and lifestyle factors were collected in *** weight and MetS-components were measured in both 2002 and *** were classified according to proximal quartiles of weight gain(WG)in percentages(%WG≤1%,1%10%, defined as:control,mild-WG,moderate-WG and severe-WG groups,respectively)at the end of the follow-up. Multivariate models were used to assess the association between MetS outcome and excessive WG in the total population,as well as in both genders. RESULTS:In total,175(12.6%)participants fulfilled MetS criteria within five *** comparison to the control group,mild-WG adults had an insignificant risk for MetS development while adults having moderate-WG had a 3.0-fold increased risk for progression to MetS [95%confidence interval(CI),1.8-5.1],and this risk was increased 5.4-fold(95%CI,3.0-9.7)in subjects having *** females having moderate-and severe-WG,the risk for developing MetS was 3.6(95% CI,1.03-12.4)and 5.5(95%CI,1.4-21.4),respectively. For males having moderate-and severe-WG,the odds ratio for MetS outcome was respectively 3.0(95%CI, 1.6-5.5)and 5.2(95%CI,2.6-10.2). CONCLUSION:For early-middle-aged healthy adults with a five-year weight gain over 5%,the severity of weight gain is related to the risk for developing metabolic syndrome.

A new heuristic approach was undertaken for the establishment of a core set for the diversity research of rice. As a result, 107 entries were selected from the 10 368 characterized accessions. The core set derived using this new approach provided a good representation of the characterized accessions present in the entire collection. No significant differences for the mean, range, standard deviation and coefficient of variation of each trait were observed between the core and existing collections. We also compared the diversity of core sets established using this Heuristic Core Collection （HCC） approach with those of core sets established using the conventional clustering methods. This modified heuristic algorithm can also be used to select genotype data with allelic richness and reduced redundancy, and to facilitate management and use of large collections of plant genetic resources in a more efficient way.

Real time sensing of crop yield is critical for a successful implementation of precision *** monitoring system is an optional component of a 55 kW multi-purpose combine harvester,developed in Korea,for both domestic and global markets,especially Asian countries where field sizes are relatively *** aim of the present study was to fabricate and evaluate the performance of a grain flow sensor suitable to the mid-sized full-feed type combine for rice,soybean,and ***,commercially available non-contact type sensing modules(optical,ultrasonic,laser,and microwave modules)were chosen for alternative candidates,to be further tested in a laboratory *** the laboratory tests,the ultrasonic module was selected as a potential approach and the performance was improved by increasing the number of modules and their ***,the improved grain flow sensor was evaluated during field harvesting *** tests with the improved grain flow sensor showed a good potential for rice(R^(2)=0.85,RMSE=126.14 g/s),soybean(R^(2)=0.78,RMSE=43.87 g/s),and barley(R^(2)=0.83,RMSE=37.39 g/s).Further research would be necessary for improvement and commercialization,through various signal processing and field tests under different field and crop conditions.

Effects of increasing the degree of building height asymmetry on ventilation and pollutant dispersion within street canyons

Rational urban design helps to build sustainable cities with high ventilation capacity and pollutant removal ca-pacity,but the effect of building height on ventilation and pollutant dispersion inside asymmetric canyons has not been fully *** this paper,we studied the effect of increasing the degree of building height asym-metry(DBHA)on canyon ventilation and pollutant diffusion in shallow and deep asymmetric street canyons by considering six different building height ratios(BHR=3/4,1/2,1/3,4/3,2/1 and 3/1).The results show that increasing the DBHA in asymmetric canyons can improve the ventilation and pollutant removal *** step-up canyons,increasing the downwind building height is very useful to improve ventilation and pollutant *** shallow/deep step-up canyons with BHR=1/3,the air exchange rate(ACH)increased to 211.2%and 380.1%of the flat canyons,*** spatially-average pollutant concentration in the pedestrian zones(leeward Kavg∗ang windward Kavg∗)decreases significantly with the increase of DBHA,especially for the deep step-up canyon with BHR=1/3,the leeward Kavg∗and windward Kavg∗decrease to 15.3%and 3%,***,increasing the upwind building height can also improve the ventilation capacity in the step-down *** the deep step-down canyon with BHR=3/1,the leeward Kavg∗and windward Kavg∗decreased to 40.6%and 24.1%of the deep flat canyon,***,the ventilation capacity is very low for step-down canyons with BHR=4/3,and for step-down canyons with BHR≥2/1,the ventilation capacity and pollutant removal capacity increase significantly with the increase of ***,in urban planning,step-down canyons with BHR=4/3 should be avoided and designed to satisfy the condition of BHR≥2/*** findings will be a valuable reference for urban designers to build sustainable cities with high ventilation capacity.

Critical limb ischemia(CLI)is a devastating disease characterized by the progressive blockage of blood *** the paracrine effect of growth factors in stem cell therapy made it a promising angiogenic therapy for CLI,poor cell survival in the harsh ischemic microenvironment limited its ***,an imperative need exists for a stem-cell delivery method that enhances cell ***,a collagen microgel(CMG)cell-delivery scaffold(40×20μm)was fabricated via micro-fragmentation from collagen-hyaluronic acid polyionic complex to improve transplantation *** human adipose-derived stem cells(hASCs)with CMG enabled integrin receptors to interact with CMG to form injectable 3-dimensional constructs(CMG-hASCs)with a microporous microarchitecture and enhanced mass ***-hASCs exhibited higher cell survival(p<0.0001)and angiogenic potential in tube formation and aortic ring angiogenesis assays than cell *** of CMG-hASCs intramuscularly into CLI mice increased blood perfusion and limb salvage ratios by 40%and 60%,respectively,compared to cell aggregate-treated *** immunofluorescent analysis revealed that transplanted CMG-hASCs have greater muscle regenerative and angiogenic potential,with enhanced cell survival than cell aggregates(p<0.05).Collectively,we propose CMG as a cell-assembling platform and CMG-hASCs as promising therapeutics to treat CLI.

Antioxidative phytoceuticals to ameliorate pancreatitis in animal models: An answer from nature

Despite enthusiastic efforts directed at elucidating critical underlying mechanisms towards the identification of novel therapeutic targets for severe acute pancreatitis(SAP), the disease remains without a specific therapy to be executed within the first hours to days after onset of symptoms. Although earlier management for SAP should aim to either treat organ failure or reduce infectious complications, the current standard of care for the general management of AP in the first hours to days after onset of symptoms include intravenous fluid replacement, nutritional changes, and the use of analgesics with a close monitoring of vital signs. Furthermore, repeated evaluation of severity is very important, as the condition is particularly unstable in the early stages. In cases where biliary pancreatitis is accompanied by acute cholangitis or in cases where biliary stasis is suspected,an early endoscopic retrograde cholangiopancreatography is ***,practice guidelines regarding the treatment of pancreatitis are *** chronic pancreatitis,conservative management strategies include lifestyle modifications and dietary changes followed by analgesics and pancreatic enzyme ***,attention has been focused on phytoceuticals or antioxidants as agents that could surpass the limitations associated with currently available *** oxidative stress has been shown to play an important role in the pathogenesis of pancreatitis,antioxidants alone or combined with conventional therapy may improve oxidativestress-induced organ *** in phytoceuticals stems from their potential use as simple,accurate tools for pancreatitis prognostication that could replace older and more tedious ***,the use of antioxidative nutrition or phytoceuticals may represent a new direction for clinical research in *** this review article,recent advances in the understanding of the pathogenesis of pancreatitis are discussed and the
