Methionine adenosyltransferases(MATs)are essential for cell survival because they catalyze the biosynthesis of the biological methyl donor S-adenosylmethionine(SAMe)from methionine and adenosine triphosphate(ATP).Mammalian cells express two genes,MAT1A and MAT2A,which encode two MAT catalytic subunits,α1 andα2,***α1 subunit organizes into dimers(MATIII)or tetramers(MATI).Theα2 subunit is found in the MATII isoform.A third gene MAT2B,encodes a regulatory subunit b,that regulates the activity of MATII by lowering the inhibition constant(Ki)for SAMe and the Michaelis constant(Km)for ***1A expressed mainly in hepatocytes maintains the differentiated state of these cells whereas MAT2A and MAT2B are expressed in non-parenchymal cells of the liver(hepatic stellate cells[HScs]and Kupffer cells)and extrahepatic tissues.A switch from the liverspecific MAT1A to MAT2A has been observed during conditions of active liver growth and *** injury,fibrosis,and cancer are associated with MAT1A silencing and MAT2A/MAT2B *** though both MAT1A and MAT2A are involved in SAMe biosynthesis,they exhibit distinct molecular interactions in liver *** review provides an update on MAT genes and their roles in liver pathologies.

In this paper, a finite element model was developed for a turbine blade with thermal barrier coatings to investigate its failure behavior under cyclic thermal loading. Based on temperature and stress fields obtained from finite element simulations, dangerous regions in ceramic coating were determined in terms of the maximum principal stress criterion. The results show that damage preferentially occurs in the chamfer and rabbet of a turbine blade with thermal barrier coatings and its thermal fatigue life decreases with the increase of thermal stress induced by high service temperature.

To study the temperature distribution and thermal-stress field in different service stages, a two-dimensional model of a turbine blade with thermal barrier coatings is developed, in which the conjugate heat transfer analysis and the decoupled thermal-stress calculation method are adopted. Based on the simulation results, it is found that a non-uniform distribution of temperature appears in different positions of the blade surface, which has directly impacted on stress field. The maximum temperature with a value of 1030 ℃ occurs at the leading edge. During the steady stage, the maximum stress of thermally grown oxide （TGO） appears in the middle of the suction side, reaching 3.75 GPa. At the end stage of cooling, the maximum compressive stress of TGO with a value of-3.5 GPa occurs at the leading edge. Thus, it can be predicted that during the steady stage the dangerous regions may locate at the suction side, while the leadine edge mav be more Drone to failure on cooling.

The silkworm, Bombyx mori, is an economically important insect that was domesticated more than 5000 years ago. Its major economic traits focused on by breeders are quantitative traits, and an accurate and efficient QTL mapping method is necessary to explore their genetic architecture. However, current widely used QTL mapping models are not well suited for silkworm because they ignore female achiasmate and gender effects. In this study, we propose a composite method combining rational population selection and special mapping methods to map QTL in silkworm. By determining QTL for cocoon shell weight（cSW）, we demonstrated the effectiveness of this method. In the cSW mapping process, only 56 markers were used and five loci or chromosomes were detected, more than in previous studies. Additionally, loci on chromosomes 1 and 11 dominated and accounted for 35.10% and 15.03% of the phenotypic variance respectively. Unlike previous studies, epistasis was detected between loci on chromosomes 11 and 22. These mapping results demonstrate the power and convenience of this method for QTL mapping in silkworm, and this method may inspire the development of similar approaches for other species with special genetic characteristics.

Glutathione S-transferases（GSTs, Ec 2.***.1.1 8） are a family of super enzymes with multiple functions that play a major role in the detoxification of endogenous and xenobiotic compounds. In our previous study, we have predicted 23 putative cytosolic GSTs in the silkworm genome using bioinformatic methods. In this study, we cloned and studied the insect-specific epsilon-class GST gene GSTe4 from the silkworm, Bombyx mori. The recombinant Bm GSTe4（Bac-Bm GSTe4） was overexpressed in SF- 9 cell lines, and it was found to have effective GST activity. We also found that the expression of Bm GSTe4 was especially down-regulated after the silkworms were fumigated with or ingested phoxim. Moreover, Bm GSTe4 protected HEK293 cells against UV-induced cell apoptosis. These results demonstrated that Bm GSTe4 has GST activity, is sensitive to phoxim, and plays a role in inhibition of UV-induced cell apoptosis.

Anthocyanins from mulberry fruits are used in medicine. However, little anthocyanin can be detected in other tissues and sometimes also mulberry fruits are colorless. The aim of this study was to investigate which gene or genes have the strongest correlation with the anthocyanin biosynthesis. The expression of several anthocyanin synthesis genes were determined in different tissues of two white and two purple fruit cultivars. Genes encoding dihydroflavonol reductase（MaDFR） and anthocyanidin synthase（MaANS） showed a high expression only in fruit tissue of purple-fruit *** the development of mulberry fruits, the anthocyanin content was well correlated with the transcripts abundance of MaDFR, MaANS, and MacHS（encoding chalcone synthase）. The skin of female mulberry flowers turns red under irradiance because of up-regulated expressions of MacHS, MaDFR, and MaANS. These three genes may control the anthocyanin biosynthesis in mulberry and up-regulation of them may greatly increase the anthocyanin content.
