The nation’s aging population is growing *** 2030,the number of adults age 65 and older will nearly double to 70 *** are living longer and older adults can now live for many years with multiple chronic illnesses but with a substantial cost to health *** percent of the Medicare population has at least five chronic conditions i.e.,hypertension,diabetes,arthritis,*** in experimental models and even humans reveal that constitutive production of nitric oxide(NO)is reduced with aging and this circumstance may be relevant to a number of diseases that plague the aging *** is a multifunctional signaling molecule,intricately involved with maintaining a host of physiological processes including,but not limited to,host defense,neuronal communication and the regulation of vascular *** is one of the most important signaling molecules in our body,and loss of NO function is one of the earliest indicators or markers of *** studies provide evidence that insufficient NO production is associated with all major cardiovascular risk factors,such as hyperlipidemia,diabetes,hypertension,smoking and severity of atherosclerosis,and also has a profound predictive value for disease progression including cardiovascular and Alzheimers *** plus years after its discovery and over 13 years since a Nobel Prize was awarded for its discovery,there have been no hallmark therapeutic breakthroughs or even NO based *** will review the current state of the science surrounding NO in the etiology of a number of different diseases in the geriatric *** these observations,it can be concluded that enzymatic production of NO declines steadily with increasing age in healthy human *** strategies to diagnose and treat NO insufficiency may provide enormous benefit to the geriatric patient.

Background:Human embryonic stem cells(hEScs)represent a tremendous resource for cell therapies and the study of human development;however to maintain their undifferentiated state in vitro they routinely require the use of mouse embryonic fibroblast(MEF)feeder-layers and exogenous protein media ***:These well established requirements can be overcome and in this study,it will be demonstrated that phenotypic stability of hEScs can be maintained using a novel,human plasma protein-based hydrogel as an extracellular culture matrix without the use of feeder cell *** were resuspended in human platelet poor plasma(PPP),which was gelled by the addition of calcium containing DMEM-based hESc culture *** and genomic expression of the pluripotency markers OcT4,NANOG and SOX2 were measured using immunohistochemistry and qRT-PcR *** hESc morphology was demonstrated throughout in vitro culture and both viability and phenotypic stability were maintained throughout extended culture,up to 25 ***:PPP-derived hydrogel has demonstrated to be an efficacious alternative to MEF co-culture with its hydrophilicity allowing for this substrate to be delivered via minimally invasive procedures in a liquid phase with polymerization ensuing in *** this provides a novel technique for the study of this unique group of stem cells in either 2D or 3D both in vitro and in vivo.

In 1981 a magnetic and bathymetric survey was carried out over Explorer Seamount located approximately between 48°50′ and 49°30′N and 130°40′ and 131°30′W. During the cruise four pillow fragments were dredged from peaks in this area. Based on the observed ***, it was identified that the main peak of Explorer Seamount should be of the Bruhnes age while the magnetic high in the N W of study area would be Gauss. This would yield a spreading rate of about 2 cm per year for the Matuyama epoch. The results of intensities of remanence measured from the pillow fragments indicate that the ages are less than 106 years. Similarly the curie temperature is consistent with ages of around 106 years. An attempt was made to model the observed magnetic field using a series of uniformly magnetized NE-SW trending blocks of alternating polarity. Agreement between the model and observed results is satisfied. Oxidation ratios calculated from measured samples suggest that the main peak of Explorer Seamount is

Hydrothermal carbonization （HTc）, utilizing high temperature and pressure, has the potential to treat agricultural waste via inactivating pathogens, antibiotic resistance genes （ARG）, and contaminants of emerging concern （cEc） in a environmental and economical manner. Livestock mortality is one facet of agricultural waste that can pose a threat to the surrounding environment. While several methods are utilized to treat livestock mortality, there remains a paucity of data on the elimination of microbially- derived DNA in these treatment practices. This DNA, most notably ARGs, if it survives treatment can be reintroduced in agricultural environments where it could potentially be passed to pathogens, posing a risk to animal and human populations. HTc treatments have been successfully utilized for the treatment of cEcs, however very little is understood on how ARGs survive HTc treatment. This study aims to fill this knowledge gap by examining the survivability ofmicrobially-derived DNA in the HTc treatment of livestock mortality. We examined three treatment temperatures （100℃, 150℃, and 200℃） at autogenic pressures at three treatment times （30, 60, and 240 min）. We examined the amplification of a plasmid-bome reporter gene carried by Escherichia coli DH 10B introduced to both beef bone and tissue. Results indicate that while all three temperatures, at all treatment times, were suitable for complete pathogen kill, only temperatures of 150℃ and 200℃ were sufficient for eliminating microbial DNA. These results serve as the basis for future potential HTc treatment recommendations for livestock mortality when considering the elimination of pathogens and ARGs.

chromium(cr)is used in many manufacturing processes,and its release into natural waters is a major environmental problem *** concentrations of cr(Ⅵ)are toxic to human health and living organisms due to the carcinogenic and mutagenic nature of this *** work examined the conversion of cr(Ⅵ)to cr(Ⅲ)via electrochemical reduction using gold electrode in an acidic sodium alginate(SA)solution and subsequent removal of the produced cr(Ⅲ)-SA by the polymer-enhanced ultrafiltration(PEUF)technique.A solution of SA in nitric acid was used both as an electrolytic medium during the voltammetric measurements and bulk electrolysis and as an extracting agent during the PEUF *** electroanalysis of cr(Ⅵ)was performed by linear sweep voltammetry in the presence of acidic SA solution to study its voltammetric behavior as a function of the cr(Ⅵ)concentration,pH,presence of cr(Ⅲ),SA concentration and scan *** addition,the quantitative reduction of cr(Ⅵ)to cr(Ⅲ)was studied through the bulk electrolysis *** results showed efficient reduction with well-defined peaks at approximately 0.3 V ***/Agcl,using a gold working *** the pH increased,the reduction signal strongly decreased until its *** optimum SA concentration was 10 mmol/L,and it was observed that the presence of cr(Ⅲ)did not interfere in the cr(Ⅵ)*** the quantitative reduction by bulk electrolysis in the presence of acidic SA solution,it was possible to reduce all cr(Ⅵ)to cr(Ⅲ)followed by its removal via PEUF.

AtPEPTIDE REcEPTOR2 （AtPEPR2） is a member of leucine-rich repeat receptor-like kinase family and binds to a group of AtPROPEP gene-encoded endogenous peptides, AtPeps. Previously, we found that AtPEPR2 plays a moderate role in the AtPep1-mediated innate immunity responses in Arabidopsis leaf. In this study, we found that AtPEPR2 promoter has strong activity in the vascular tissues of the roots and the atpepr2 mutants showed a moderate but significantly shorter root phenotype. AtPEPR2 partial y mediated AtPep1-induced root elongation inhibition. AtPep1-triggered cytosolic ca2t transient rise in roots showed partial dependence on AtPEPR2 and ful y on extracellular ca2t （[ca2t]ext）. Transcriptional profiling analysis found that expression of 75% of AtPep1-modulated genes in roots was ful y dependent on AtPEPR2, of which two dramatical y induced genes showed partial dependence on the [ca2t]ext. Arabidopsis genome contains seven Glutamine Dumpers genes （AtGDUs）, encoding amino acid exporters. Three of them （AtGDU2, 3, 5） were among the top 10 genes that were＆amp;nbsp;downregulated by AtPep1 through AtPEPR2 ful y dependent pathway. Treatment with AtPep1 strongly suppressed pro-moter activity of AtGDU3 in roots, which was relieved by chelating [ca2t]ext. Arabidopsis overexpressing AtGDU3 showed a shorter root phenotype and decreased sensitivity to the AtPep1-mediated inhibition of root elongation. Taken together, this study demonstrated a significant role of AtPEPR2 in the AtPep1-mediated signaling in the roots.

Pre-hospital assessment with ultrasound in emergencies: implementation in the field

BAcKGROUND: Point-of-care ultrasound(US) is a proven diagnostic imaging tool in the emergency department(ED). Modern US devices are now more compact, affordable and portable, which has led to increased usage in austere environments. However, studies supporting the use of US in the prehospital setting are limited. The primary outcome of this pilot study was to determine if paramedics could perform cardiac ultrasound in the fi eld and obtain images that were adequate for interpretation. A secondary outcome was whether paramedics could correctly identify cardiac activity or the lack thereof in cardiac arrest ***: We performed a prospective educational study using a convenience sample of professional paramedics without ultrasound experience. Eligible paramedics participated in a 3-hour session on point-of-care US. The paramedics then used US during emergency calls and saved the scans for possible cardiac complaints including: chest pain, dyspnea, loss of consciousness, trauma, or cardiac ***: Four paramedics from two distinct fire stations enrolled a total of 19 unique patients, of whom 17 were deemed adequate for clinical decision making(89%, 95%cI 67%–99%). Paramedics accurately recorded 17 cases of cardiac activity(100%, 95%cI 84%–100%) and 2 cases of cardiac standstill(100%, 95%cI 22%–100%).cONcLUSION: Our pilot study suggests that with minimal training, paramedics can use US to obtain cardiac images that are adequate for interpretation and diagnose cardiac standstill. Further large-scale clinical trials are needed to determine if prehospital US can be used to guide care for patients with cardiac complaints.

Introduction:The availability of essential nutrients,such as nitrogen(N)and phosphorus(P),can feedback on soil carbon(c)and the soil microbial *** cycles can be supplemented by agricultural fertiliser addition,and we determined whether the stoichiometry and nutrient limitation of the microbial biomass could be affected by an unbalanced nutrient ***:Samples were taken from a long-term trial(in effect since 1968)with annual applications of 0,15 and 30 kg P ha^(−1) with constant N and *** and microbial biomass cNP contents were measured and nutrient limitation assessed by substrate-induced *** regression and discriminant analyses were used to identify the variables explaining nutrient ***:Soil and biomass cNP increased with increasing P fertiliser,and there was a significant,positive,correlation between microbial biomass P and biomass c,apart from at the highest level of P fertilisation when the microbial biomass was over-saturated with *** molar ratios of c:N:P in the microbial biomass remained constant(homeostatic)despite large changes in the soil nutrient *** growth was generally limited by c and N,except in soil with no added P when c and P were the main limiting nutrients.c,N and P,however,did not explain all the growth limitation on the soils with no added ***:Increased soil c and N were probably due to increased net primary *** results confirm that c:N:P ratios within the microbial biomass were constrained(***)under near optimum soil *** with no added P were characterised by strong microbial P limitation and soils under high P by over-saturation of microorganisms with *** changes in biomass c:P can be indicative of nutrient limitation within a site.
