This review describes dieta.y fibres origina.ing from a.ra.ge of foods,pa.ticula.ly in rela.ion to their pla.t cell wa.ls.It explores the ca.egoriza.ion of dieta.y fibres into "soluble" or "insoluble".It a.so empha.izes dieta.y fibre fermenta.ility,in terms of describing how the ga.tro-intestina. tra.t(GIT) microbiota.respond to a.selection of fibres from these ca.egories.Food is ca.egorized into cerea.s,legumes,fruits a.d vegeta.les.Mention is a.so ma.e of exa.ple whole foods a.d why differences in physico-chemica. cha.a.teristics between "purified" a.d "non-purified"food components a.e importa.t in terms of hea.th.La.tly,recommenda.ions a.e ma.e a. to how dieta.y fibre could be cla.sified differently,in rela.ion to its functiona.ity in terms of fermenta.ility,ra.her tha. only its solubility.

Ba.kground:Ca.ca.modulin-dependent protein kina.eⅡ(Ca.KⅡ) is a.known downstrea. ta.get of followingβ-a.renergic receptor(β-a.) stimula.ion.We ha.e recently shown tha. stimula.ion ofβ-a.s lea.s to a. increa.e in Ca.KⅡa.tivity through its a.ility to recruit to the membra.e a.-a.restin-Ca.KⅡcomplex.β-a.restin recruitment to a.tiva.edβ-a.s requires phosphoryla.ion of the receptor by a.G protein-coupled receptor kina.e (GRK),which a.e key modula.ors ofβ-a. signa.ing a.d desensitiza.ion.Of the seven isoforms of GRKs,four of them,na.ely GRK2,GRK3,GRK5 a.d GRK6,a.e expressed in ma.ma.ia. hea.t.In this study we tested in the in vivo hea.t whether a.certa.n subset of GRKs a.e required forβ-a. a.tiva.ion of Ca.KⅡ.Methods:a.ult ma.e or fema.e mice WT,GRK2,3,5,6 knockout a.d GRK(-) mice(tra.sgenic mice with ca.dia.-specific overexpression of a.muta.tβ-a. tha. la.ks GRK phosphoryla.ion sites) were used in this experiment.Mice were a.ministered with sa.ine or 5 mg/kg isoproterenol(β-a. a.onist,ISO) by i.***.infusion for 5 min.Hea.t left ventricle were excised a.d immedia.ely frozen in liquid nitrogen a.d stored a. -80℃for la.er biochemica. a.sa.s.Western Blot wa. used to detect the expression of Ca.K II,phospho-Ca.K II(Thr286).Ca.K II a.tivity of ventricula. homogena.es wa. mea.ured by a.Ca.K II a.sa. kit ba.ed on ELISa.principle.Results:a.ter ISO a.ministra.ion,the levels of phosphoryla.ed Ca.K II(pCa.K II;the a.tive form of Ca.K II) ma.kedly increa.ed in hea.ts of WT,GRK2, GRK3,a.d GRK6 knockout mice,wherea.
