Brain antigens in functionally distinct antigen-presenting cell populations in cervical lymph nodes in MS and EAE

Drainage of central nervous system(CNS) antigens to the brain-draining cervical lymph nodes(CLN) is likely crucial in the initiation and control of autoimmune responses during multiple sclerosis(MS).We demonstrate neuronal antigens within CLN of MS *** monkeys and mice with experimental autoimmune encephalomyelitis(EAE) and in mouse models with non-inflammatory CNS damage,the type and extent of CNS damage was associated with the frequencies of CNS antigens within the cervical lymph *** addition,CNS antigens drained to the spinal-cord-draining lumbar lymph *** human MS CLN,neuronal antigens were present in pro-inflammatory antigen-presenting cells(APC),whereas the majority of myelincontaining cells were *** may reflect a different origin of the cells or different drainage ***,neuronal antigen-containing cells in human CLN did not express the lymph node homing receptor CCR7,whereas myelin antigen-containing cells in situ and in vitro ***,CLN from EAE-affected CCR7-deficient mice contained equal amounts of myelin and neuronal antigens as wild-type *** conclude that the type and frequencies of CNS antigens within the CLN are determined by the type and extent of CNS ***,the presence of myelin and neuronal antigens in functionally distinct APC populations within MS CLN suggests that differential immune responses can be evoked.

AIM： To evaluate accuracy of in vivo diagnosis of ad- enomatous vs non-adenomatous polyps using/-SCAN digital chromoendoscopy compared with high-definition white light. METHODS： This is a single-center comparative effec- tiveness pilot study. Polyps （n = 103） from 75 average- risk adult outpatients undergoing screening or surveil- lance colonoscopy between December 1, 2010 and April 1, 2011 were evaluated by two participating en- doscopists in an academic outpatient endoscopy center. Polyps were evaluated both with high-definition white light and with/-SCAN to make an/n vivo prediction of adenomatous vs non-adenomatous pathology. We de- termined diagnostic characteristics of/-SCAN and high- definition white light, including sensitivity, specificity, and accuracy, with regards to identifying adenomatous vs non-adenomatous polyps. Histopathologic diagnosis was the gold standard comparison. RESULTS： One hundred and three small polyps, de- tected from forty-three patients, were included in the analysis. The average size of the polyps evaluated in the analysis was 3.7 mm （SD 1.3 mm, range 2 mm to 8 mm）. formal histopathology revealed that 54/103 （52.4%） were adenomas, 26/103 （25.2%） were hyper- plastic, and 23/103 （22.3%） were other diagnoses in- clude ＂lymphoid aggregates＂, ＂non-specific colitis,＂ and ＂no pathologic diagnosis.＂ Overall, the combined accu- racy of endoscopists for predicting adenomas was iden- tical between/-SCAN （71.8%, 95%CI： 62.1%-80.3%） and high-definition white light （71.8%, 95%CI： 62.1%-80.3%）. However, the accuracy of each endosco- pist differed substantially, where endoscopist A demon- strated 63.0% overall accuracy （95%CI： 50.9%-74.0%） as compared with endoscopist B demonstrating 93.3% overall accuracy （95%CI： 77.9%-99.2%）, irrespective of imaging modality. AIM： To correlate the significance of liver biochemical tests in diagnosing post orthotopic liver transplantation （OLT） biliary complications and to study their profile before and after endoscopic therapy.METHODS： Patients who developed biliary complications were analysed in detail for the clinical information, laboratory tests, treatment offered, response to it, follow up and outcomes. The profile of liver enzymes was determined. The safety, efficacy and outcomes of endoscopic retrograde cholangiography （ERC） were also analysed. RESULTS： 40 patients required ERC for 70 biliary complications. GGT was found to be 〉 3 times （388.1 ± 70.9 U/mL vs 168.5 4± 34.2 U/L, P = 0.007） and SAP 〉 2 times （345.1 ± 59.1 U/L vs 152.7 ± 21.4 U/L, P = 0.003） the immediate post OLT values. Most frequent complication was isolated anastomotic res in 28 （40%）. Sustained success was achieved in 26 （81%） patients. CONCLUSION： Biliary complications still remain an important problem post OLT. SAP and GGT can be used as early, non-invasive markers for diagnosis and also to assess the adequacy of therapy. Endoscopic management is usually effective in treating the majority of these biliary complications.

We present modeling work on three young stellar objects that are promis-ing targets for future high-resolution observations to investigate circumstellar disk evolution. The currently available data comprise the spectral energy distribution from optical to millimeter wavelengths which allow constraining the structure of the cir-cumstellar disk using self-consistent radiative transfer models. The results suggest that the assumption of well-mixed dust and gas leads to overestimation of flux in the far-infrared. Observational and theoretical arguments suggest that an overall decrease in far-infrared excess can be explained by dust settling towards the midplane. A new disk model is hence employed to take the effect of dust sedimentation into account. The extended model satisfactorily reproduces all existing observations. The three tar-gets studied here therefore deserve follow-up observations to reveal the evolutionary state of their protoplanetary disks.

We present a detailed comparison of two approaches, the use of a precalculated database and simulated annealing （SA）, for fitting the continuum spectral energy distribution （SED） of astrophysical objects whose appearance is dominated by surrounding dust. While pre-calculated databases are commonly used to model SED data, only a few studies to date employed SA due to its unclear accuracy and convergence time for this specific problem. from a methodological point of view, different approaches lead to different fitting quality, demand on computational resources and calculation time. We compare the fitting quality and computational costs of these two approaches for the task of SED fitting to provide a guide to the practitioner to find a compromise between desired accuracy and available resources. To reduce uncertainties inherent to real datasets, we introduce a reference model resembling a typical circumstellar system with 10 free parameters. We derive the SED of the reference model with our code MC3 D at 78 logarithmically distributed wavelengths in the range [0.3 μm, 1.3 mini and use this setup to simulate SEDs for the database and SA. Our result directly demonstrates the applicability of SA in the field of SED modeling, since the algorithm regularly finds better solutions to the optimization problem than a precalculated database. As both methods have advantages and shortcomings, a hybrid approach is preferable. While the database provides an approximate fit and overall probability distributions for all parameters deduced using Bayesian analysis, SA can be used to improve upon the results returned by the model grid.

Aim： To determine the androgenic effects of Basella alba and Hibiscus macranthus extracts in the rat and the bull, and to develop a novel in vitro test system using Leydig cells from bull testes. Methods： The effect of methanol extracts from both plants on testosterone production in isolated Leydig cells from the rat and the bull was analyzed using ^125I-radioimmunoassay （^125I-RIA）. Rat Leydig cells were obtained by common methods, whereas a novel technique was used to purify Leydig cells from bull testes. Results： Bull testes from the slaughter house were a cheap source of pure Leydig cells. In culture, these cells produced testosterone for 5-6 days, which can be stimulated by human chorionic gonadotrophin （hCG）. Basella alba extracts significantly enhanced testosterone production in bull and rat Leydig cells in a concentration-dependent manner. Hibiscus macranthus showed no androgenic effect but was shown to inhibit testosterone production at higher concentrations. Conclusion： Leydig cells purified from bull testes can be used as an alternative tool in experimental animal research. Certain fractions of Basella alba extract demonstrated androgenic potential whereas Hibiscus macranthus extracts did not.
