Protective effect of probiotics on intestinal barrier function in malnourished rats after liver transplantation

BACKGROUND: Most patients waiting for liver transplantation have end-stage liver diseases with malnutrition, which is prone to induce intestinal barrier dysfunction after liver transplantation. We aimed to study the effect of probiotics on intestinal barrier function in malnourished rats following liver transplantation with long-term antibiotics. METHODS: Twelve Lewis rats were selected as donors. Twelve BN rats, which served as recipients, were subjected to malnutrition by semi-starvation for 4-5 weeks. They were randomly divided into two groups: a control group which received phosphate-buffered saline and a probiotics group which received Bifidobacterium and Lactobacillus. All recipients were injected with intramuscular imipenem and subcutaneous cyclosporine A. Furthermore, six normal BN rats without any drugs or operations served as a normal group. Eight days after operation, all rats were sacrificed for examination of the following parameters: serum levels of endotoxin and TNF-α, bacterial translocation, intestinal microflora, ileocecal sIgA, lymphocyte numbers, and phenotypes (CD4, CD8, αβTCR, γδTCR)ofPeyer’spatches. RESULTS: In recipients subjected to malnutrition, weight decreased by 20% and they survived until 8 days after operation. Compared with the normal group, all recipients on postoperative day 8 showed increased levels of serum endotoxin and TNF-α as well as increased counts oftranslocated bacteria. Meanwhile, there were decreases in counts of Bifidobacterium and Lactobacillus in the ileocecum, sIgA concentration, and lymphocytes of Peyer’s patches. Moreover, partial alteration in lymphocyte phenotypes was evidenced by elevated ratios of CD8 + and γδTCR + lymphocytes. In contrast, compared to the control group, supplementation with probiotics reduced the levels of serum endotoxin, TNF-α and bacterial translocation, increased the counts of Bifidobacterium and Lactobacillus, the concentration of sIgA and lymphocytes of Peyer’s patches, and also slightly

What we know about ST13, a co-factor of heat shock protein, or a tumor suppressor?

This article is to summarize the molecular and functional analysis of the gene “suppression of tumorigenicity 13” (ST13). ST13 is in fact the gene encoding Hsp70 interacting protein (Hip), a co-factor (co-chaperone) of the 70-kDa heat shock proteins (Hsc/Hsp70). By collaborating with other positive co-factors such as Hsp40 and the Hsp70-Hsp90 organizing protein (Hop), or competing with negative co-factors such as Bcl2-associated athanogen 1 (Bag1), Hip facilitates may facilitate the chaperone function of Hsc/Hsp70 in protein folding and repair, and in controlling the activity of regulatory proteins such as steroid receptors and regulators of proliferation or apoptosis. Although the nomenclature of ST13 implies a role in the suppression of tumorigenicity (ST), to date available experimental data are not sufficient to support its role in cancer development, except for the possible down-regulation of ST13 in gastric and colorectal cancers. Further investigation of this gene at the physiological level would benefit our understanding of diseases such as endocrinological disorders, cancer, and neurodegeneration commonly associated with protein misfolding.

Changes of phospholipase D activity of rat peritoneal mast cells in degranulation

IM: To study the changes of phospholipase D (PLD) activity of actively sensitized rat peritoneal mast cells (RPMC) in degranulation. METHODS: Degranulation of RPMC was determined by measurement of β-hexosaminidase release. PLD activity assay was carried out by measurement of PLD product, choline, with chemiluminescent oxidation of luminol. RESULTS: Actively sensitized RPMC challenged with ovalbumin (0.5-8 mg/L for 120 s, 4 mg/L for 15-120 s) resulted in significant activation of PLD accompanied with the increment of P-hexosaminidase release. PLD activity of sensitized RPMC was increased by more than 2-fold compared with that of unsensitized RPMC which contained low levels of PLD activity [(35±13) pmol choline/min in 1×106cells], but β-hexosaminidase releases of the sensitized cells were as low as spontaneous releases.

Immunosuppression in Human Peripheral Blood T Lymphocytes by Fluvastatin

To investigate the immunosuppressive effect of fluvastatin on the PHA-activated T lymphocytes. T lymphocytes were isolated from the blood of healthy volunteers, cell proliferation and the activation mar- kers expression were examined by flow cytometric analysis. Cytokine secretion was assayed by ELISA. LDH-release assay was used to detect activity of killer cells. NFAT activation was evaluated by TransAMTM ELISA kit. Results were as following. (1) Whereas no modification in CD25 expression was seen, fluvastatin at 5 μM caused a lower level of CD69 expression, accompanied by an essential suppression on proliferation, IL-2 production and cytotoxicity development in PHA-stimulated T cells. However, the level of secreted IL-10 had no change, and the level of IL-4 even experienced a significant increase. (2) Combined with cyclosporine A (CsA), fluvastatin would further repress CD69 expression, cells proliferation and activity of killer cells, meanwhile significantly induced the secretion of IL-4 and IL-10. (3) Fluvastatin treatment also resulted in a strong inhibition of NFAT activation. In conclusion, partly involving the blockage of activation of NFAT, fluvastatin exhibited an immunosuppressive effect in vitro.

Shear wave velocity-based liquefaction evaluation in the great Wenchuan earthquake: a preliminary case study

The great Wenchuan earthquake （Ms= 8.0） in 2008 caused severe damage in the western part of the Chengdu Plain. Soil liquefaction was one of the major causes of damage in the plain areas, and proper evaluation of liquefaction potential is important in the definition of the seismic hazard facing a given region and post-earthquake reconstruction. In this paper, a simplified procedure is proposed for liquefaction assessment of sandy deposits using shear wave velocity （V）, and soil liquefaction from the Banqiao school site was preliminarily investigated after the earthquake. Boreholes were made at the site and shear wave velocities were measured both by SASW and down-hole methods. Based on the in-situ soil information and V profiles, the liquefaction potential of this site was evaluated. The results are reasonably consistent with the actual field behavior observed after the earthquake, indicating that the proposed procedure is effective. The possible effects of gravel and fines contents on liquefaction of sandy soils were also briefly discussed.

Effect of IFNα-2a on Fas expression and apoptosis rate of peripheral blood cytotoxic T cells in patients with hepatitis B

Interferon(IFN) with antiviral and im-munomodulatory activities is one of the most important therapeutic agents for the treatment of chronic hepatitis. The apoptotic effect of IFN is influenced by cell type and the types of IFN, which suppresses proliferation and induces apoptosis in some cell types while inhibiting apoptosis in others. The aim of this study was to explore the effect of IFNα-2a on Fas expression and the apoptosis rate of peripheral blood cytotoxic T cells (CTLs) in patients with hepatitis B. METHODS:Peripheral blood mononuclear cells were isolated from 26 patients with hepatitis B including 16 patients with chronic hepatitis B and 10 patients with chronic severe hepatitis B. Fas expression and apoptosis rate of CTLs were analyzed with flow cytometry before and after IFNα-2a treatment. RESULTS:Before IFNα-2a treatment, Fas expression and apoptosis rate of CTLs from patients with chronic hepatitis B were significantly higher than those from patients with chronic severe hepatitis B and healthy controls respectively. No significant difference was observed between Fas expression and apoptosis rate of CTLs from patients with chronic severe hepatitis B and healthy controls. After IFNα-2a treatment,Fas expression and apoptosis rate of CTLs from different groups were compared with those before IFNα-2a treatment, showing no significant difference despite alternation of different degree. CONCLUSIONS:Activation induced cell death (AICD) exists in peripheral blood CTLs from patients with hepatitis B. No effect of IFNα-2a exerts on Fas expression and apoptosis rate of Fas in patients with hepatitis B.

In this paper,a novel method to automatically detect protein spots on a two-dimensional (2-D) electrophoresis gel image is proposed to implement proteomics analysis of complex *** the basis of the identifying spots results based on color variation and spot size features,morphological feature is introduced as a new criterion to distinguish protein spots from non-protein ***-sharpening,edge-detecting and morphological feature extraction methods were consequently combined to detect protein spots on a 2-D electrophoresis gel image subject to strong *** proposed method was applied to detect the protein spots of proteomic gel images from *** cell,human kidney tissue and human *** results demonstrated that this method is more accurate and reliable than previous methods such as PDQuest 7.2 and ImageMaster 5.0 software for detecting protein spots on gel images with strong interferences.

Determination of low level HBsAg in serum by microparticle enzyme immunoassay

Objective: To investigate the distribution of the popula- tion with iow level HBsAg in serum and the characteri- stics of their HBV serologic markers. Methods: We detected HBsAg, HBeAg, anti-HBc, an- ti-HBe, and anti-HBs in the serum samples from 8089 non-hepatitis inpatients and defined the patients with HBsAg concentration equal to or less than 5 μg/L by using the AxSYM microparticle enzyme immunoassay (MEIA) system. Results: 189 patients with HBsAg equal to or Iow than 5 μg/L (2.34% of all 8089 patients and 23.16% of 816 HBsAg positive patients) included 84 (44.40%), 33 (17.5%) and 72 (38.10%) with HBsAg equal to or less than 1 μg/L, 1-2 μg/L and 2-5 μg/L respec- tively. Most of these patients were detected with 'posi- tive for HBsAg, anti-HBc and anti-HBe but negative for HBeAg and anti-HBs', and anti-HBc antibodies were frequently present in their sera. Conclusions: The population with low Level HBsAg in serum should not be neglected and it is important to improve the test sensitivity of HBsAg and to detect the other HBV serologic markers.

BACKGROUND:Fatty liver is a common chronic liver disease *** is associated with an increasing morbidity in china in recent *** aim of this study was to analyze the effect of drinking alcohol on the hemoglobin and biochemical values of patients with fatty ***:We investigated the clinical and laboratory data of 669 patients with fatty *** the 669 patients,166 consumed alcohol more than 60 g per week for at least 2 years,and 503 did not have a history of long-term alcohol *** further analyzed the relationship between alcohol consumption and clinical characteristics of these ***:The values of aspartate transaminase (AST),gammaglutamyl transpeptidase (GGT),and hemoglobin in the long-term consumption group were significantly higher than those in the non long-term consumption group (P<0.05).In the patients without long-term alcohol consumption,the values of GGT and hemoglobin in patients with light alcohol consumption were significantly higher than those in non alcohol consumers (P<0.05).CONCLUSION:Alcohol consumption is associated with significantly increased values of AST,GGT,and hemoglobin in patients with fatty liver,suggesting their potential roles in hepatic steatosis.

Minocycline inhibits 5-lipoxygenase activation and brain inflammation after focal cerebral ischemia in rats

Aim:To determine whether the anti-inflammatory effect of minocycline on postis-chemic brain injury is mediated by the inhibition of 5-lipoxygenase （5-LOX）expression and enzymatic activation in ***:Focal cerebral ischemiawas induced for 30 min with middle cerebral artery occlusion,followed *** ischemic injuries,endogenous IgG exudation,the accumulationof neutrophils and macrophage/microglia,and 5-LOX mRNA expression weredetermined 72 h after reperfusion.5-LOX metabolites （leukotriene B4and cysteinylleukotrienes） were measured 3 h after ***:Minocycline （22.5and 45 mg/kg,ip,for 3 d） attenuated ischemic injuries,IgG exudation,and theaccumulation of neutrophils and macrophage/microglia 72 h after *** inhibited 5-LOX expression 72 h after reperfusion and the production ofleukotrienes 3 h after reperfusion,Conclusion:Minocycline inhibited postis-chemic brain inflammation,which might be partly mediated by the inhibition of5-LOX expression and enzymatic activation.
