Papaya is a rich source of nutrients,antioxidants,and vitamins;however,it is highly perishable and hence needs to be preserved using suitable unit *** is an economical method of papaya treatment,but results in the loss of essential nutrients and also degrades its pigments,color,and ***,this work aims to utilize ultrasound and osmotic dehydration as pretreatment strategies to preserve the rich composition of papaya during conventional hot air *** dried papaya slices were evaluated for moisture content,water activity,total phenolic content,β-carotene,color,antioxidant activity,microstructure,and *** showed that sample pretreatment with 33 kHz ultrasound frequency and dehydrated with 35°Brix osmotic solution resulted in lower moisture content(<12%w.b.)and water activity(<0.41),high phenolic content(88.5 mg GAE/mL),highβ-carotene content(184.54 mg/g)and antioxidant capacity(48.3%),and preserved color after *** texture(hardness)of the pretreated samples was also more desirable from an industrial standpoint,which was further validated using SEM micrographs that showed a more porous *** study concludes that pretreatment of papaya slices with ultrasound and osmotic dehydration is recommended before drying.

Osteoarthritis （OA） is a degenerative joint disease and a major cause of pain and disability in older adults. We have previously identified epidermal growth factor receptor （EGFR） signaling as an important regulator of cartilage matrix degradation during epiphyseal cartilage development. To study its function in OA progression, we performed surgical destabilization of the medial meniscus （DMM） to induce OA in two mouse models with reduced EGFR activity, one with genetic modification （, was/＋ mice） and the other one with pharmacological inhibition （gefitinib treatment）. Histological analyses and scoring at 3 months post-surgery revealed increased cartilage destruction and accelerated OA progression in both mouse models. TUNEL staining demonstrated that EGFR signaling protects chondrocytes from OA-induced apoptosis, which was further confirmed in primary chondrocyte culture. Immunohistochemistry showed increased aggrecan degradation in these mouse models, which coincides with elevated amounts of ADAMTS5 and matrix metalloproteinase 13 （MMP13）, the principle proteinases responsible for aggrecan degradation, in the articular cartilage after DMM surgery. Furthermore, hypoxia-inducible factor 2α （HIF2α）, a critical catabolic transcription factor stimulating MMP13 expression during OA, was also upregulated in mice with reduced EGFR signaling. Taken together, our findings demonstrate a primarily protective role of EGFR during OA progression by regulating chondrocyte survival and cartilage degradation.

Radiation therapy is a common treatment regimen for cancer ***,its adverse effects on the neighboring bone could lead to fractures with a great impact on quality of *** underlying mechanism is still elusive and there is no preventive or curative solution for this bone *** hormone(PTH)is a current therapy for osteoporosis that has potent anabolic effects on *** this study,we found that focal radiation from frequent scans of the right tibiae in 1-month-old rats by micro-computed tomography severely decreased trabecular bone mass and deteriorated bone ***,PTH daily injections remarkably improved trabecular bone in the radiated tibiae with increases in trabecular number,thickness,connectivity,structure model index and stiffness,and a decrease in trabecular *** analysis revealed that radiation mainly decreased the number of osteoblasts and impaired their mineralization activity but had little effects on *** reversed these adverse effects and greatly increased bone formation to asimilar level in both radiated and non-radiated ***,PTH protects bone marrow mesenchymal stem cells from radiation-induced damage,including a decrease in number and an increase in adipogenic *** radiation generated the same amount of free radicals in the bone marrow of vehicle-treated and PTH-treated animals,the percentage of apoptotic bone marrow cells was significantly attenuated in the PTH *** together,our data demonstrate a radioprotective effect of PTH on bone structure and bonemarrow and shed new light on a possible clinical application of anabolic treatment in radiotherapy.

Prospects of diagnostic and prognostic biomarkers of pyometra in canine

Maintaining bone architecture requires continuous generation of osteoblasts from osteoprogenitor *** previous study of mice with epidermal growth factor receptor(EGFR)specifically inactivated in osteoblast lineage cells revealed that EGFR stimulates bone formation by expanding the population of mesenchymal *** ligands are potent regulators for the osteoprogenitor pool but the underlying mechanisms are largely *** we demonstrate that activation of EGFR increases the number of osteoprogenitors by promoting cell proliferation and suppressing either serum depletion-or TNFα-induced apoptosis mainly through MAPK/Erk *** calvarial organ culture revealed that EGF elevated the number of proliferative cells and decreased the number of apoptotic cells,which lead to increased *** analysis of MC3T3 cells,an osteoprogenitor cell line,revealed that EGFR signaling stimulates the expression of Mcl1,an anti-apoptotic protein,and a family of Egr transcription factors(Egr1,2,and 3).The up-regulation of Mcl1 and Egr2 by EGF were further confirmed in osteoprogenitors close to the calvarial bone *** of Nab2,a co-repressor for Egrs,attenuated the EGF-induced increase in osteoprogenitor ***,knocking down the expression of Egr2,but not Egr1or 3,resulted in a similar *** inhibitor,adenovirus overexpression,and si RNA approaches,we demonstrate that EGFR signaling activates MAPK/Erk pathway to stimulate the expression of Egr2,which in turn leads to cell growth and Mcl1-mediated cell *** together,our data clearly demonstrate that EGFR-induced Egr2expression is critical for osteoprogenitor maintenance and new bone formation.

Pyometra is one of the most common uterine pathologies of intact bitch at middle to advanced age. In the early stages, the disease shows subtle changes, making diagnosis a challenge. In contrast, at later stages, it manifests as potentially life-threatening systemic inflammatory response syndrome. Ultrasonographic examination of the uterus aids in the diagnosis, although it has limitation in ascertaining the clinical severity of pyometra. Moreover, differentiation of cystic endometrial hyperplasia from pyometra could not be discerned with greater accuracy. Therefore, false negative diagnosis of pyometra patients leads to development of systemic inflammatory response, which delays administration of therapies and results in deaths during early course of treatment. Further, indiscriminate use of broad-spectrum antimicrobials at higher dose in false positive cases considerably contributes to the rising pool of drug resistant pathogens, thereby increasing the risk of case fatality due to sepsis in a long-term. Monitoring the circulating pro-inflammatory cytokines, acute phase proteins, endotoxin, growth factors and inflammatory mediators is the current trend in pyometra research, especially for developing diagnostic and prognostic biomarkers. The present review deals with the prospects of developing diagnostic and prognostic biomarkers in the canine pyometra.

Spinal cord injury（SCI）-induced bone loss represents the most severe osteoporosis with no effective *** animal studies have focused primarily on long bones at the acute stage using adolescent rodents. To mimic chronic SCI in human patients, we performed a comprehensive analysis of long-term structural and mechanical changes in axial and appendicular bones in adult rats after SCI. In this experiment, 4-month-old Fischer 344 male rats received a clinically relevant T13 contusion injury. Sixteen weeks later, sublesional femurs, tibiae,and L4 vertebrae, supralesional humeri, and blood were collected from these rats and additional non-surgery rats for micro-computed tomography（m CT）, micro-finite element, histology, and serum biochemical *** trabecular sites, extreme losses of bone structure and mechanical competence were detected in the metaphysis of sublesional long bones after SCI, while the subchondral part of the same bones showed much milder damage. Marked reductions in bone mass and strength were also observed in sublesional L4 vertebrae but not in supralesional humeri. At cortical sites, SCI induced structural and strength damage in both sub- and supralesional long bones. These changes were accompanied by diminished osteoblast number and activity and increased osteoclast number and activity. Taken together, our study revealed site-specific effects of SCI on bone and demonstrated sustained inhibition of bone formation and elevation of bone resorption at the chronic stage of SCI.

Environmental, biological, socio-cultural and economic status variation existing in the Central Himalaya have led to the evolution of diverse and unique traditional agroecosystems, crop species and livestock, which facilitate the traditional mountain farming societies to sustain themselves. Indigenous agroecosystems are highly site specific and differ from place to place, as they have evolved along divergent lines. For maintenance of traditional agrodiversity management the farmers of the Central Himalaya have evolved various types of crop rotations in consonance with the varied environmental conditions and agronomic requirements. In irrigated fiat lands two crops are harvested in a year with negligible fallow period but in rainfed conditions if a cropping sequence is presumed to be starting after winter fallow phase then four major cropping seasons can be identified namely first kharif season （first crop season）, first rabi season （second crop season）, second kharif season （third crop season） and second rabi season （fourth crop season）. Highest crop diversity is present in kharif season in comparison to rabi season. Traditionally the fields are left fallow after harvest of the second kharif season crop. Important characteristics of agrodiversity management are the use of bullocks for draughtpower, human energy as labour, crop residues as animal feed and animal waste mixed with forest litter as organic input to restore soil fertility levels. Women provide most of the human labour except for ploughing and threshing grain. The present study deals with assessment of traditional agrodiversity management such as （i） crop diversity, （ii） realized yield under the traditional practices and （iii） assess the differences of realized yields under sole and mixed cropping systems. It indicated that crop rotation is an important feature of the Central Himalayan village ecosystem which helps to continue the diversity of species grown, as are the distribution of crops in the growing

Chemotherapy has been linked with Takotsubo cardiomyopathy. Most of the literature on chemotherapy associated Takotsubo cardiomyopathy is on the drug 5-fluorouracil. In this report, we describe the case of a 55-year-old Asian male who developed Takotsubo cardiomyopathy while receiving dual chemotherapy with cytarabine and daunorubicin for acute myeloid leukemia. To our knowledge, it is the first case of Takotsubo cardiomyopathy associated with daunorubicin and/or cytarabine.
