Blood flow restriction(BFR)during exercise bouts has been used to induce hypertrophy of skeletal muscle,even with low ***,the effects of BFR during the rest periods between sets are not *** have tested the hypothesis that BFR during rest periods between sets of high-intensity resistance training would enhance ***-two young adult male university students were recruited for the current study,with n=11 assigned to BFR and n=11 to a control *** results revealed that four weeks training at 70%of 1 RM,five sets and 10 repetitions,three times a week with and without BFR,resulted in similar progress in maximal strength and in the number of maximal *** miR-1 and miR-133a decreased significantly in the vastus lateralis muscle of BFR group compared to the group without BFR,while no significant differences in the levels of miR133b,miR206,miR486,and miR499 were found between *** conclusion,it seems that BFR restrictions during rest periods of high-intensity resistance training,do not provide benefit for enhanced performance after a four-week training ***,BFR-induced downregulation of miR-1 and miR-133a might cause different adaptive responses of skeletal muscle to high intensity resistance training.

＜正＞ The peptide hormone somatostatin has been shown to inhibit the release of inflammatory neuropeptides from the nerve endings of capsaicin sensitive neurons and thus prevent inflammation. Previous studies indicated that somatostatin exerts its anti-inflammatory effects through the SSTR4 and possibly the SSTR1 receptor. To facilitate large scale screening for pharmacologically active compounds acting on these targets, two new cell lines were created which express the human SSTR1 and SSTR4 receptors. CHO-K1 cells were transfected by alentiviral system containing the human SSTR1 and SSTR4 cDNAs. In the constructs the SSTR cDNA is followedby an internal ribosome entry site allowing separate expression of the enhanced green fluorescent protein from the same mRNA. Stable and uniform expression of the SSTR receptors were demonstrated by RT-PCR. flow cytometry and immunohistochemistry. Binding of somatostatin-14. somatostatin analogue peptid TT-232. the selective SSTR4 ligand KD-5621 and the selective SSTR1 ligand KD-7825 were demonstrated by a radioactive binding assay.

AIM To analyze the effect of intralesional steroid injections in addition to endoscopic dilation of benign refractory esophageal *** A comprehensive search was performed in three databases from inception to 10 April 2017 to identify trials, comparing the efficacy of endoscopic dilation to dilation combined with intralesional steroid injections. Following the data extraction, meta-analytical calculations were performed on measures of outcome by the randomeffects method of Der Simonian and Laird. Heterogeneity of the studies was tested by Cochrane's Q and I^2 statistics. Risk of quality and bias was assessed by the Newcastle Ottawa Scale and JADAD assessment *** Eleven articles were identified suitable for analyses, involving 343 patients, 235 cases and 229 controls in total. Four studies used crossover design with 121 subjects enrolled. The periodic dilation index(PDI) was comparable in 4 studies, where the pooled result showed a significant improvement of PDI in the steroid group(MD:-1.12 dilation/month, 95% CI:-1.99 to -0.25 P = 0.012; I^2 = 74.4%). The total number of repeat dilations(TNRD) was comparable in 5 studies and showed a non-significant decrease(MD:-1.17, 95%CI:-0.24-0.05, P = 0.057; I^2 = 0), while the dysphagia score(DS) was comparable in 5 studies and did not improve(SMD: 0.35, 95%CI:-0.38, 1.08, P = 0.351; I^2 = 83.98%) after intralesional steroid *** Intralesional steroid injection increases the time between endoscopic dilations of benign refractory esophageal strictures. However, its potential role needs further research.

The results of an experimental study of radiation characteristics of a barrier discharge in a mixture ArCCl 4 were presented in this paper. Formation of molecules ArCl(B) and Cl 2 (D") in the barrier discharge plasma depends strongly on the partial pressure of freon vapor is shown. The intensity of the radiation band Cl 2 (D"-A") on the value of the pulse repetition rate current at a charging voltage U d = 13 kV was close to the line that indicates the possibility of increasing the average power of the VUV-UV discharge at frequencies f > 1 kHz.

DNA extraction from paraffin embedded colorectal carcinoma samples: A comparison study of manual vs automated methods,using four commercially kits

BACKGROUND Nucleic acid isolation from formalin-fixed, paraffin-embedded tissue(FFPET)samples is a daily routine in molecular pathology laboratories, but extraction from FFPET is not always easily achieved. Choosing the right extraction technique is key for further *** To compare the performance of four commercially available kits used for DNA extraction in routine *** DNA isolation was performed on 46 randomly selected formalin-fixed, paraffinembedded(FFPE) colorectal adenocarcinoma(CRC) surgical specimens. Four commercially available extraction kits were used: two for manual DNA extraction(the Pure Link Genomic DNA Mini Kit from Invitrogen and the High Pure FFPE DNA Isolation Kit from Roche) and two for automated DNA extraction(the i Prep Genomic DNA Kit from Invitrogen and the Magna Pure LC DNA Isolation Kit from Roche). The DNA concentration and quality(odds ratio) among the four systems were compared. The results were correlated with the clinicopathological aspects of CRC cases: age, gender, localization, macro-and microscopic features,lymph node metastases, and the lymph node *** The highest DNA concentration was obtained using the manual kits: 157.24 ±62.99 ng/μL for the Pure Link Genomic DNA Mini Kit and 86.64 ng/μL± 43.84 for the High Pure FFPE DNA Isolation Kit(P < 0.0001). Lower concentrations were obtained with automated systems: 20.39 ± 21.19 ng/μL for the Magna Pure LC DNA Isolation Kit and 8.722 ± 6.408 ng/μL for the i Prep Genomic DNA Kit,with differences between the systems used(P < 0.0001). The comparison between age, gender, tumor localization, pT or pN stage and the lymph node ratio indicated no statistically significant difference in DNA concentration using any of the nucleic acid isolation kits. DNA concentration was influenced by the macroscopic features and grade of differentiation. ＜正＞ Vagal nerve is likely to have a prominent role in centrally induced ***, different neuronal pathways project to dorsal vagal complex which may influence the vagal efferent activity. The present work analysed the chain of events involved in alpha - 2 - adrenoceptor - initiated gastric mucosal defense. Methods Gastric damage was induced by ethanol. Gastric acid secretion was measured in pylorus ligated *** were given either intracerebroventricularly （icv.） or intraperitoneally （ip.）.Results: Rilmenidine - alpha - 2/11 - imidazolinereceptors agonist - inhibited the ethenol - induced lesion （ED50:2 nmol/ kg ip.,6 pmol/rat icv.）. Yohimbine,prazosin,ARC 239 （icv.）（alpha-2B - adrenoreceptor antagonists） and the opioid receptor antagonist naloxone inhibited the gastroprotective effect. The gastroprotection was also blocked by NMDA receptor antagonist dizocilpine and NO synthase inhibitor L - NNA （icv.）. Conclusion: Activation of central alpha - 2B - adrenoceptor subtype initiates an opioid - excitatory amino acid - NO - mediated process resulted in gastric mucosal protection.

Single skip metastasis in sentinel lymph node: In an early gastric cancer

Lymph node status is considered a key prognostic and predictive factor in patients with gastric cancer(GC).Although there is a practical approach to the intraoperative detection of sentinel lymph nodes(SLNs),such a procedure is not included in the European surgical *** this report,we present a practical approach to SLN mapping in a representative case with early gastric cancer(EGC).A 74-year-old female was hospitalized with an endoscopically observed,superficially ulcerated tumor located in the antral *** gastrectomy with D2 lymphadenectomy and SLN mapping was performed by injecting methylene blue dye into the peritumoral submucosal *** incidentally detected blue-stained lymph node located along the middle colic artery was also *** was detected 40 min after injection of the methylene *** examination showed a p T1b-staged well-differentiated HER-2-negative *** of the 41 LNs located at the first,third,and fifth station of the regional LN compartments were found to be free of tumor *** only lymph node with metastasis was located along the middle colicartery and was considered a non-regional lymph *** incidentally identified skip metastasis indicated stage Ⅳ GC.A classic chemotherapy regimen was given,and no recurrences were observed six months after *** this representative case,low-cost SLN mapping,with a longer intraoperative waiting time,totally changed the stage of the tumor in a patient with EGC.
