Gene-edited pigs for agricultural and biomedical applications are typically generated using somatic cell nuclear transfer(SCNT).However, SCNT requires the use of monoclonal cells as donors, and the time-consuming and laborious monoclonal selection process limits the production of large populations of gene-edited animals. Here, we developed a rapid and efficient method named RE-DSRNP(reporter RNA enriched dual-sg RNA/CRISPR-Cas9 ribonucleoproteins) for generating gene-edited donor cells. RE-DSRNP takes advantage of the precise and efficient editing features of dual-sg RNA and the high editing efficiency, low off-target effects, transgene-free nature, and low cytotoxic characteristics of reporter RNA enriched RNPs(CRISPR-Cas9ribonucleoproteins), thus eliminating the need for the selection of monoclonal cells and thereby greatly reducing the generation time of donor cells from 3–4 weeks to 1 week, while also reducing the extent of apoptosis and chromosomal aneuploidy of donor cells. We applied RE-DSRNP to produce cloned pigs bearing a deletion edit of the wild-type p53-induced phosphatase 1(WIP1)gene: among 32 weaned cloned pigs, 31(97%) carried WIP1 edits, and 15(47%) were homozygous for the designed fragment deletion, and no off-target event was detected. The WIP1 knockout(KO) pigs exhibited male reproductive disorders, illustrating the utility of RE-DSRNP for rapidly generating precisely edited animals for functional genomics and disease research. REDSRNP's strong editing performance in a large animal and its marked reduction in the required time for producing SCNT donor cells support its application prospects for rapidly generating populations of transgene-free cloned animals.

Streptococcus agalactiae, Streptococcus dysgalactiae and Streptococcus uberis are three common pathogens causing bovine mastitis. The GapC of Streptococcus is a highly conserved surface layer protein that is responsible for inducing protective immune response in mice. To identify protective epitope of GapC, a monoclonal antibody (mAb1F2) against the recombinant GapC of S. dysgalactiae was generated by screening a phage-displayed random 12-peptide library. After three rounds of biopanning, 25 phage clones were randomly selected and their specificity to the mAb was verified by sandwich ELISA. Their amino acid of 13 among total 25 phage colones were sequenced and deduced. It was found that their amino aicd sequences can match amino acids 30TRINDLT36 of GapC. It showed that mAb1F2 could react with the motif 30TRINDLT36 fused GST in Western bloting. So it proposed that the motif 30TRINDLT36 represented a linear B-cell epitope of the GapC. A single replacement of amino acid for the epitope 30TRINDLT36 demonstrated that residues R31A, I32A, N33A, D34A and L35A constitute the core of the epitope. The high homology was found after alignment of the epitope sequences from different streptococcus species. The identified epitope would be useful in development of epitope-based marker vaccines against Streptococcus infection.

This paper investigates the impact of non-ideal user equipment(UE) hardware on a cell-free(CF) massive MIMO(mMIMO) network with centralized operation under spatially correlated channels. The minimum mean-squared error(MMSE) estimator can be derived with the help of the generic non-ideal UE hardware model. It is demonstrated that even if the effective signal-to-noise ratio approaches infinity, pilot contamination and imperfect hardware can cause a non-zero estimation error floor. After that, a lower bound is determined for the ergodic uplink capacity of the centralized CF mMIMO network under non-ideal UE hardware. Moreover, the optimal receive combining vector is obtained to maximize the uplink spectral efficiency(SE). The maximum ratio(MR) and regularized zero-forcing(RZF) combining schemes are offered as alternatives in light of the computational complexity of the MMSE receiver. Comparing the RZF to the MMSE scheme under different levels of hardware impairments, our findings indicate that the RZF receiver suffers a negligible loss in total SE. For MR combining, a novel closed-form uplink achievable SE expression is obtained based on the MMSE estimator and the use-and-then-forget bounding technique. This expression gives vital insights into the achievable uplink performance with UE hardware impairments. Besides, for various hardware impairment factors, the impact of pilot sequence length on average sum SE is disclosed for different receive combining schemes. To increase the overall SE of the max-min fairness scheme, a heuristic fractional power control scheme with UE hardware impairments is developed, which can essentially avoid sacrificing the SE of other UEs while maximizing the SE of the unluckiest UE in the whole network. Finally,our theoretical performance analysis and power control algorithm are validated by simulation results, and fundamental design guidelines are provided for selecting hardware satisfying the practical UE requirements.
