Hypoxia-inducible factor(HIF-1α),a core transcription factor responding to changes in cellular oxygen levels,is closely associated with a wide range of physiological and pathological ***,its differential impacts on vascular cell types and molecular programs modulating human vascular homeostasis and regeneration remain largely ***,we applied CRISPR/Cas9-mediated gene editing of human embryonic stem cells and directed differentiation to generate HIF-ia-deficient human vascular cells including vascular endothelial cells,vascular smooth muscle cells,and mesenchymal stem cells(MsCs),as a platform for discovering cell type-specific hypox-ia-induced response *** comparative molecular profiling across cell types under normoxic and hypoxic conditions,we provide insight into the indispensable role of HIF-1αin the promotion of ischemic vascular *** found human MSCs to be the vascular cell type most susceptible to HIF-1a deficiency,and that transcriptional inactivation of ANKZF1,an effector of HIF-1a,impaired pro-angiogenic ***,our findings deepen the understanding of HIF-ia in human angiogenesis and support further explorations of novel therapeutic strategies of vascular regeneration against ischemic damage.

A Truncated Arabidopsis NUCLEOSOME ASSEMBLY PROTEIN 1, AtNAP1;3T, Alters Plant Growth Responses to Abscisic Acid and Salt in the Atnap 1;3-2 Mutant

Chromatin remodeling is thought to have crucial roles in plant adaptive response to environmental stimulus. Here, we report that, in Arabidopsis, the evolutionarily conserved histone chaperone, NUCLEOSOME ASSEMBLY PROTEIN 1 （NAP1）, is involved in plant response to abscisic acid （ABA）, a phytohormone important in stress adaptation. We show that simultaneous loss-of-function of AtNAP1;1, AtNAP1;20 and AtNAP1;3 （the triple mutant m123-1） caused a slight hypersensitive response to ABA in seedling growth. Strikingly, the other triple mutant m123-2 containing a different mutant allele of AtNAP1;3, the Atnap 1;3-2 allele, showed a hyposensitive response to ABA and a decreased tolerance to salt stress. This ABA- hyposensitive and salt response phenotype specifically associated with the Atnapl;3-2 mutant allele. We show that this mutant allele produced a truncated protein, AtNAP1;3T, which lacks 34 amino acids at the C-terminus compared to the wild-type protein AtNAP1;3. We further show that the heterozygous plants containing the Atnapl;3-2 mutant allele as well as transgenic plants overexpressing AtNAP1;3Texhibit ABA-hyposensitive phenotype. It thus indicates that AtNAP1;3T functions as a dominant negative factor in ABA response. The expression of some ABA-responsive genes, including genes encoding protein kinases and transcription regulators, was found perturbed in the mutant and in theAtNAP1;3Ttransgenic plants. Taken together, our study uncovered AtNAP1 proteins as positive regulators and AtNAP1;3Tas a negative regulator in ABA signaling pathways, providing a novel link of chromatin remodeling to hormonal and stress responses.

Evaporation and drying characteristics of the sessile ferrofluid droplet under a horizontal magnetic field

In this study,the evaporation characteristics and drying patterns of various sessile ferrofluid droplets on certain substrate under horizontal magnetic fields of controlled intensities are *** effects of droplet concentration and magnetic field intensity on the duration of each evaporation stage and drying patterns of droplets have been systematically *** turned out that a plateau appears at the initial stage of evaporation in the absence of magnetic field and it was found that the plateau value is positively correlated with the concentration of ferrofluid *** the external magnetic field,the evaporation time of droplets decreases,the stage of contact line retreat extends,the stage of late pinning mode shortens,and the deposition area of ferrofluid droplet decreases compared to that of without magnetics *** deposition area increases gradually and becomes more uniform with the increase of magnetic *** decrease of friction force which is due to the decrease of the number of nanoparticles at the contact line under external magnetic field is the main reason for the observed *** found that the coffee ring and the uniform deposition inside the droplet will be destroyed when the magnetic field intensity is higher than a critical *** work has a significant reference value for the evaporation of sessile magnetic fluid droplets under the applied magnetic field,especially when the drying pattern needs to be precisely controlled,such as in spray or biomedicine.

Bearing fault diagnosis based on a multiple-constraint modal-invariant graph convolutional fusion network

Multisensor data fusionmethod can improve the accuracy of bearing fault diagnosis,in order to address the problems of single-sensor data types and the insufficient exploration of redundancy and complementarity between different modal data in most existing multisensor data fusion methods for bearing fault diagnosis,a bearing fault diagnosis method based on a Multiple-Constraint Modal-Invariant Graph Convolutional Fusion Network(MCMI-GCFN)is proposed in this ***,a Convolutional Autoencoder(CAE)and Squeeze-and-Excitation Block(SE block)are used to extract features of raw current and vibration ***,the model introduces source domain classifiers and domain discriminators to capture modal invariance between different modal data based on domain adversarial training,making use of the redundancy and complementarity between multimodal ***,the spatial aggregation property of Graph Convolutional Neural Networks(GCN)is utilized to capture the dependency relationship between current and vibration modes with similar time step features for accurately fusing contextual semantic ***,the validation is conducted on the public bearing damage current and vibration dataset from Paderborn *** experimental results showed that the delivered fusion method achieved a bearing fault diagnosis accuracy of 99.6%,which was about 9%–11.4%better than that with nonfusion methods.

Autoantibodies in Chinese patients with chronic hepatitis B:Prevalence and clinical associations

AIM: To investigate the prevalence of autoantibodies and their associations with clinical features in Chinesepatients with chronic hepatitis B(CHB).METHODS: A total of 325 Chinese patients with CHB were enrolled in this retrospective,hospitalbased *** with chronic hepatitis C(CHC),autoimmune hepatitis(AIH),or primary biliary cirrhosis(PBC) were included,with healthy donors acting as controls.A panel of autoantibodies that serologically define AIH and PBC was tested by indirect immunofluorescence assay and line *** AIH-related autoantibody profile included homogeneous anti-nuclear antibodies(ANA-H),smooth-muscle antibodies,anti-liver kidney microsome type 1,antiliver cytosolic antigen type 1,and anti-soluble liver antigen/liver pancreas; the PBC-related antibodies were characterized by ANA-nuclear dots/membranous rimlike,anti-mitochondrial antibodies-M2(AMA-M2),antiBPO(recombinant antigen targeted by AMA-M2),antiSp100,anti-promyelocytic leukemia protein(anti-PML),and *** dichotomization of clustering was used to unequivocally designate the AIH or PBC profiles for each ***-Ro52 antibodies were also ***: The prevalence of any autoantibody in CHB amounted to 58.2%,which was similar to the 66.2% prevalence in CHC,significantly higher than the 6.7% in the healthy controls(P < 0.001),and lower than the 100% found in AIH and PBC(P = 0.004 and P < 0.001,respectively).There were more anti-PML and anti-gp210 antibodies among the CHB patients than the CHC patients(11.1% vs 0%,P = 0.003; 12.6% vs 0%,P < 0.001,respectively).The prevalence and titer of AMA,anti-BPO,anti-PML,and anti-gp210 were higher in PBC than in those with *** the CHB patients,the prevalence of ANA,especially ANA-H,was significantly lower in patients with compensated and decompensated cirrhosis compared with patients without ***-eight cases of hepatocellular carcinoma(HCC) in CHB showed a significant differencecompared with non-HCC patients i

Non-obstructive azoospermia(NOA),the lack of sperm in the ejaculate due to failure of spermatogenesis,can result from a variety of diverse factors,with genetic factors accounting for~30%of NOA cases,including chromosomal aberrations,Y-chromosome microdeletions,and monogenic variants(Tang et al.,2022).More than 2000genes are known to participate in spermatogenesis,and>400 genes specifically linked to azoospermia have been identified through studies in mouse models(Krausz and Riera-Escamilla,2018).However,relatively few genes associated with azoospermia in mice have been verified in humans.

Discovery of IHMT-MST1-39 as a novel MST1 kinase inhibitor and AMPK activator for the treatment of diabetes mellitus

Insulin-producing pancreaticβcell death is the fundamental cause of type 1 diabetes(T1D)and a contributing factor to type 2 diabetes(T2D).Moreover,metabolic disorder is another hallmark of *** sterile 20-like kinase 1(MST1)contributes to the progression of diabetes mellitus through apoptosis induction and acceleration of pancreaticβcell ***-activated protein kinase(AMPK)is an energy sensing kinase and its activation has been suggested as a treatment option for metabolic ***,pharmacological inhibition of MST1 and activation of AMPK simultaneously represents a promising approach for diabetes ***,we discovered a novel selective MST1 kinase inhibitor IHMT-MST1-39,which exhibits anti-apoptosis efficacy and improves the survival of pancreaticβcells under diabetogenic conditions,as well as primary pancreatic islets in an ex vivo disease ***,IHMT-MST1-39 activated AMPK signaling pathway in hepatocytes in vitro,combination of IHMT-MST1-39 and metformin synergistically prevented hyperglycemia and significantly ameliorated glucose tolerance and insulin resistance in diabetic *** together,IHMT-MST1-39 is a promising anti-diabetic candidate as a single agent or in combination therapy for both T1D and T2D.
