Subcellular chemical imaging of structurally similar acridine drugs by near-field laser desorption/laser postionization mass spectrometry

Insights into the pharmacologic effect on cellular processes and the potential toxicological effects are vital to new drug development and evaluation,yet research on these subjects remains a great challenge due to the lack of information regarding the spatiotemporal distribution of drugs and metabolites within a single *** spectrometry imaging(MSI)has proven to be a label-free and high-throughput approach for visualizing drug distribution in spatial and temporal ***,single-cell drug imaging has been limited so far by detection sensitivity and microscale lateral ***,we report near-field laser desorption/laser postionization mass spectrometry(NDPI-MS)for single-cell imaging of two structurally similar drugs,proflavine and ethacridine,and subcellular distributions of proflavine at different drug concentrations were *** NDPI-MS imaging results indicate that proflavine was accumulated in lysosomes,which was verified by laser scanning confocal microscopy(LSCM).Additionally,a distinguished subcellular distribution pattern of ethacridine from proflavine could be visualized,highlighting the complexity of the interaction between the drugs and biological environment even though these two drugs possess similar *** together,the present results demonstrate the great potential of the integrated single-cell MSI platform for characterizing the drug distribution and its phenotype changes within individual cells,expediting the identification and evaluation of newly developed drugs.

Beam quality measurement and consistent safety standard for high power laser products

With the development of laser technology,laser application technology increasingly plays a leading role in the field of industry. High power laser products and their safety requirements are attracting more attention. In laser industrial applications, laser beam quality and the system of laser beam propagation and focusing are greatly ignored. Furthermore, the basic safety standards for laser products have been neither widely publicized nor strictly enforced because of rapid development. This paper examines the underlying relationship between laser beam quality and laser manufacturing, and makes a comparison among different laser systems. It is also shown how the laser safety standards could be better understood and laser beam quality standard is advocated that directly relates to a mission requirement.

Design and discovery of carrier-mediated modified pesticides are vital for reducing pesticide dosage and increasing utilization,yet it remains a great challenge due to limited insights into plant translocation ***/nanoparticle assisted laser desorption/ionization strategy has established itself as a preferential analytical tool for biological tissue analysis,whereas potential applications in plant sciences are hindered with regard to the inability to slice plant leaves and ***,we report gold nanoparticle(AuNP)-immersed paper imprinting mass spectrometry imaging(MSI)for the spatiotemporal visualization of pesticide translocation in plant *** approach plays a dual role in preserving spatial information and improving ionization efficiency for pesticides regardless of imaging artifacts due to homogenous AuNP *** this MSI platform,we proposed the elaborate plant translocation mechanism of agrochemicals for the first time,which is currently poorly *** dynamic processes of carrier-mediated pesticides can be clearly visualized,including crossing of plasma membranes by transporters,translocation downward in stems through the phloem,diffusion to the xylem and,conversely,accumulation at margins of the treated ***,this AuNP-assisted paper imprinting method could be highly compatible with laser-based MSI instruments,expediting researches across a broad range of fields,especially in nanomaterial development and life sciences.

Mass spectrometry imaging(MSI)has made the spatio-chemical characterization of a broad range of small-molecule metabolites within biological tissues ***,available matrix-assisted laser desorption/ionization mass spectrometry(MALDI-MS)suffers from severe background interferences in low-mass ranges and inhomogeneous matrix ***,surface-assisted LDI-MS(SALDI-MS)has been an attractive alternative for high-sensitivity detection and imaging of small *** this study,we construct a new composite substrate,hydrophobic polydopamine(hPDA)-modified TiO_(2)nanotube(TDNT)coated with plasmonic gold nanoparticle(AuNP-hPDA-TDNT),as a dual-polarity SALDI substrate using an easy and cost-effective fabrication *** from the synergistic effects of TDNT semiconductor and plasmonic PDA modification,this SALDI substrate exhibits superior performance for dual-polarity detection of a vast diversity of small *** reduced background interferences,lower detection limits,and spot-to-spot repeatability can be achieved using AuNP-hPDA-TDNT *** to its unique imprinting performance,various metabolites and lipids can be visualized within jatropha integerrima petals,ginkgo leaves,strawberry fruits,and latent *** valuably,the universality of this matrix-free substrate is demonstrated for mapping spatial distribution of lipids within mouse brain tissue *** together,this AuNP-hPDA-TDNT material is expected to be a promising SALDI substrate in various fields,especially in nanomaterial development and life sciences.

The discovery of novel botanical pesticides as a preferred alternative to synthetic pesticides is regarded as an environmentally friendly strategy,yet it remains a great challenge due to limited insights into the synthesis and accumulation of active ingredient ***,we demonstrate the use of gold nanoparticle(AuNP)-assisted laser desorption/ionization mass spectrometry imaging(LDI-MSI)for the tissue-specific distribution and spatiotemporal accumulation effect of rotenone and active ingredient intermediates in its biosynthetic pathway within the roots of Derris *** MSI results revealed that rotenone was mainly concentrated in the epidermis,cortex,and xylem in the first two years and began to accumulate in the phloem since the 3rd ***,the rotenone contents increased in the epidermis,cortex,and xylem in the 4th year and finally decreased in the 5th *** ultra-performance liquid chromatography(UPLC)results revealed that there was a significant difference in rotenone content between the root and non-root bark(p<0.05)except for the 1st *** reached its maximum value at 9.71%in the root bark in the 4th year and 7.98%in the non-root bark in the 1st year,which was in accordance with the MSI ***,the active ingredient intermediates in the Hydroxylation/Methylation and Rotenoid Phases of the biosynthetic pathway of rotenone were concentrated in the root *** together,the collective results provide the scientific guidance for the scientific cultivation of *** and the efficient extraction of botanical pesticides.

Recently, rare earth elements(REEs) have garnered great attention in marine geochemistry, climate change, and anthropogenic pollution as valuable indicators.1, 2 To fully exploit these applications, how to comprehensively give insight into the distribution of REEs in seawater and marine deposits is vital. However, the considerably low concentration(below g/mL, ppm) and similar chemical properties of REEs, as well as the complicated matrix resulting from high salt content in the natural environment have a negative effect upon their accurate determination. To this end, some extensively used methods for direct solid analysis involve laser-induced breakdown spectrometry(LIBS), laser ablation inductively coupled plasma mass spectrometry(LA-ICPMS), and laser ablation/ionization mass spectrometry(LAI-MS) were proposed. Compared to conventional solvent-based methods, these techniques offer high throughput and fast screening without complicated matrix separation by evaporating a microdroplet solution to form the residue, especially when sample amounts are limited. To quantify trace elements with minor sample volumes, an appropriate strategy for achieving low limits of detection with less sample consumption is highly preferred.3 Herein, a laser desorption and laser postionization time-of-flight mass spectrometer(LD-LPI-TOFMS) was developed for direct microtrace determination of rare earth elements(REEs) in residues. Neutral atoms of REEs residue are generated by low-irradiance laser desorption(< 10 W/cm), followed by high-irradiance laser postionization(～ 10 W/cm) of the desorbed atoms, facilitating to decoupling the processes between desorption and ionization spatially and temporally. This strategy plays a dual role in alleviating the barriers of deteriorating spectral resolution at high irradiance, serious matrix effects and elemental fractionation effects at low irradiance.
