The expression of Lin28B was co-regulated by H3K4me2 and Wnt5a/β-catenin/TCF7L2

Lin28A and Lin28B are homologous RNA-binding proteins that participate in the development of primordial germ cells. The mechanisms underlying expression and regulation of Lin28A have been well documented, but such information for Lin28B is limited. In this study, a fragment of the Lin28B promoter was cloned, the pEGFP-pLin28B vector was constructed. DF-1 chicken fibroblasts were transfected and the expression of green fluorescent protein (GFP) was measured. Furtherly, Lin28B promoter of different lengths fragments was cloned using the chromosome-walking method and the fragments were ligated into the PGL3-Basic vector, and transfected into DF-1 cells. Results of dual-luciferase reporter assay showed that the core of the Lin28B promoter was included in the sequence from –1 431 to –1 034 bp. The binding sites of the transcription factor TCF7L2 was showed within this sequence by bioinformatics analysis. The promoter activity of Lin28B was downregulated (P<0.05) when the TCF7L2 binding site was mutated. Further experiments suggested that Lin28B promoter activity responded to the activation or inhibition of Wnt signaling. Results of chromatin immunoprecipitation and quantitative PCR showed that β-catenin-TCF7L2 may be enriched in the Lin28B promoter core area. In vivo and in vitro activation or inhibition of Wnt signaling significantly up- or down-regulated (P<0.05) Lin28B expression. H3K4me2 enriched in the promoter of Lin28B, which affected the regulation of Wnt signaling to Lin28B. In conclusion, our results showed that H3K4me2 and Wnt5a/β-catenin/TCF7L2 were the positive regulators of Lin28B expression. Findings of this study may lay a theoretical foundation for illuminating the mechanism underlying Lin28B expression.

Study on the role of JAK/STAT signaling pathway during chicken spermatogonial stem cells generation based on RNA-Seq

Spermatogonia! stem cells（SSCs） form the foundation for spermatogenesis and sustain male fertility.To explore the regulatory mechanisms of chicken SSCs generation,we obtained highly purified chicken embryonic stem cells（ESCs）,primordial germ cells（PGCs） and SSCs by fluorescence-activated cell sorting（FACS）.High-throughput analysis methods（RNA-Seq） were used to sequence the transcriptome level of these cells.Gene ontology and Kyoto encyclopedia of genes and genomes（KEGG） pathway enrichment were used to analyze RNA-Seq results.BMP4 was used to induce chicken ESCs differentiation to SSCs-like cells in vitro.The quantitative real-time（qRT）-PCR was used to detect the expression changes of the key genes.The results showed that 22 relevant critical pathways were found by RNA-Seq,one of them was the Janus kinase/signal transducer and activator of transcription（JAK/STAT） signaling pathway.Total of 103 related genes were detected in this pathway.Protein-protein interactions analysis found that 87 proteins were significantly related to 19 key proteins in this pathway.These 87 proteins were enriched in 21 biological processes and 18 signaling pathways.Moreover,during the differentiation of chicken ESCs to SSCs-like cells induced by BMP4 in vitro,JAK2 and STAT3 were activated.The qRT-PCR results showed that the expression trends of JAK2 and STAT3 were basically the same as in vivo.We concluded that JAK/STAT signaling pathway plays an important role in the process of chicken SSCs generation both in vivo and in vitro;it may achieve its function through multiple biological processes and other related pathways.

UBE2I stimulates female gonadal differentiation in chicken(Gallus gallus)embryos

Without known analogous sex-determining factors like SRY(sex determining region Y)in mammals,the chicken(Gallus gallus)sex determination mechanism still remains unclear,which highly restricts the biological research on chicken development and poultry single-sex reproduction.Here we not only characterized a new female-biased gene UBE2I and identified the expression pattern by qRT-PCR,but also described the functional role of UBE2I in the gonadal development of chicken embryos.Results showed that UBE2I exhibited a female-biased expression pattern in the early stage of PGCs(primordial germ cells)in embryonic gonads and robust expression in ovaries of newborn chickens.Most importantly,we successfully developed an effective method to interfere or overexpress UBE2I in chicken embryos through the intravascular injection.The qRT-PCR analysis showed that the sex-related genes(FOXL2,CYP19A1 and HINTW)in females were upregulated(P<0.05)under the overexpression of UBE2I and the sex-related genes(SOX9,DMRT1 and WT1)in females were downregulated(P<0.05)after interfering UBE2I.Furthermore,the change of UBE2I expression was associated with the level of estradiol and its receptors(AR and ESR),which suggests that UBE2I is necessary to initiate the female-specific development in chickens.In conclusion,this work demonstrates that UBE2I is a crucial sex differentiation-related gene in the embryonic development of chickens,which provides insights for further understanding the mechanism of sex determination in chickens.

Research on the appropriate way to transfer exogenous substances into chicken embryos

In biological research, chicken embryos are a classic experimental model for the exploration of the embryonic development and cell differentiation. Transferring exogenous substances into chicken embryos for producing medical antibodies has been widely used in the production practice. However, there are few studies about the effect of the different injection site and dosage on chicken embryos. The aim of this study was to explore the effects of different injection sites and dosages on chicken embryo hatching rate and development, so as to provide a basis for further studies using the chicken embryo model. Freshly laid eggs （Rugao yellow chicken） were injected with different doses of saline at the tip, equatorial plane and the blunt end of the egg shell, respectively. Egg hatching rate was recorded and compared among injection sites and different doses. A trypan blue stain was also injected at the aforementioned sites and the growth of chicken embryos was observed. The SPSS （statistical package for the social science） software was used to analyze the relationship between the chicken eggs hatching rate and the different injection sites or the different dosages. The experimental results showed that there were significant differences on egg hatching rates among the different injection sites and doses （P〈0.05）. The hatchability of the blunt end injection group was significantly higher than that of the other two sites. The egg hatching rate decreased with increased saline doses. The egg hatching rate of the 100 pL saline injection group was higher than the 200 and 300 μL dosage groups. Ultimately, we suggest that the optimal chicken embryo injection process is during early development, at the blunt end site with a dose less than 100 μL to minimize damage to the egg.

Fractal model of spontaneous imbibition in low-permeability reservoirs coupled with heterogeneity of pore seepage channels and threshold pressure

Spontaneous imbibition(SI)is an important mechanism for enhancing oil recovery in low-permeability reservoirs.Due to the strong heterogeneity,and the non-Darcy flow,the construction of SI model for lowpermeability reservoirs is extremely challenging.Commonly,traditional SI models based on single or averaged capillary tortuosity ignore the influence of heterogeneity of pore seepage channels and the threshold pressure(TP)on imbibition.Therefore,in this work,based on capillary model and fractal theory,a mathematical model of characterizing SI considering heterogeneity of pore seepage channels is established.On this basis,the threshold pressure was introduced to determine the pore radius at which the wetted phase can displace oil.The proposed new SI model was verified by imbibition experimental data.The study shows that for weakly heterogeneous cores with permeability of 0-1 m D,the traditional SI model can characterize the imbibition process relatively accurately,and the new imbibition model can increase the coefficient of determination by 1.05 times.However,traditional model has serious deviations in predicting the imbibition recovery for cores with permeability of 10-50 m D.The new SI model coupling with heterogeneity of pore seepage channels and threshold pressure effectively solves this problem,and the determination coefficient is increased from 0.344 to 0.922,which is increased by2.68 times.For low-permeability reservoirs,the production of the oil in transitional pores(0.01-0.1μm)and mesopores(0.1-1μm)significantly affects the imbibition recovery,as the research shows that when the heterogeneity of pore seepage channels is ignored,the oil recovery in transitional pores and mesopores decreases by 7.54%and 4.26%,respectively.Sensitivity analysis shows that increasing interfacial tension,decreasing contact angle,oil-water viscosity ratio and threshold pressure will increase imbibition recovery.In addition,there are critical values for the influence of these factors on the imbibition recovery,which provides theoret

Molecular Chaperone-Dependent Polymer Translocation through Nanopores: The Effects of Chaperone Concentration and Chaperone-Polymer Interaction

The polymer translocation through a nanopore from a donor space(or named cis side) to a receiver space(trans side) in the chaperone-induced crowded environment has attracted increasing attention in recent years due to its significance in biological systems and technological applications. In this work, we mainly focus on the effects of chaperone concentration and chaperone-polymer interaction on the polymer translocation. By assuming the polymer translocation to be a quasi-equilibrium process, the free energy F of the polymer can be estimated by Rosenbluth-Rosenbluth method and then the translocation time τ can be calculated by Fokker-Plank equation based on the obtained free energy landscape. Our calculation results show that the translocation time can be controlled by independently tuning the chaperone concentration and chaperone-polymer interaction at the cis side or the trans side. There exists a critical chaperone-polymer attraction ε~*=-0.2 at which the volume exclusion and interaction effects of the chaperone can balance each other. Additionally, we also find that at large chaperone-polymer attraction, the translocation time is mainly governed by the diffusion coefficient of the polymer.

Design Strategy of Diagonal Compressors in Compressed Air Energy Storage System

As a kind of large-scale physical energy storage,compressed air energy storage(CAES)plays an important role in the construction of more efficient energy system based on renewable energy in the future.Compared with traditional industrial compressors,the compressor of CAES has higher off-design performance requirements.From the perspective of design,it needs to pay attention not only to the performance of the design point,but also to the performance of all the stable working range.However,from the previous literature,no diagonal compressor was used in CAES which can meet the requirements,which also reflects the design program can be further improved.Therefore,this paper studies the design strategy of high efficient diagonal compressor for large-scale CAES,and gives the complete strategy algorithms used for different program modules.The pressure ratio,isentropic efficiency and stable working range are comprehensively considered.In the design process,the criteria for the key parameters of the diagonal flow angle of the diagonal compressor are given for the first time.The results show that the isentropic efficiency at the design point is 92.7%,the total pressure ratio is1.97,and the stable working range exceeds 20%,which meets the design requirements of the compressor for CAES and exceeds the overall performance of the previous compressors in the entire working range.

The increasingly serious environmental challenges have gradually aroused people's interest in electric vehicles.Over the last decade,governments and automakers have collaborated on the manufacturing of electric vehicles with high performance.Cutting-edge battery technologies are pivotal for the performance of electric vehicles.Zn-air batteries are considered as potential power batteries for electric vehicles due to their high capacity.Zn-air battery researches can be classified into three categories:primary batteries,mechanically rechargeable batteries,and chemically rechargeable batteries.The majority of current studies aim at developing and improving chemically rechargeable and mechanically rechargeable Zn-air batteries.Researchers have tried to use catalytic materials design and device design for Zn-air batteries to make it possible for their applications in electric vehicles.This review will highlight the state-of-the-art in primary batteries,mechanically rechargeable batteries,and chemically rechargeable batteries,revealing the prospects of Zn-air batteries for electric vehicles.

Energy-saving and environmentally friendly photocatalysis has emerged as a popular research area in response to issues with energy scarcity and environmental degradation.Due to the unique layer-like structure,BiOX（Cl,Br,I） is frequently used in photocatalysis.However,inherent flaws in BiOX,such as an inappropriate band gap and low carrier separation efficiency,restrict its capacity for photocatalysis.Owing to the tunable grouping layer,alloying engineering is employed to optimize the intrinsic properties of BiOX and alloyed BiOX becomes a promising photocatalytic material.This review describes the structure of BiOX,where tunable halogen layers provide favorable conditions for the implementation of alloying engineering to improve intrinsic properties.The article compares the effects and mechanisms of alloying engineering on the optimization of the energy band structure and carrier behavior of BiOX,and lists various modification methods used to improve the optimization of the intrinsic properties by alloying engineering,including defect engineering,morphology control as well as the synergy between alloying and other modification methods（bismuth-rich strategies,cation doping,construction of heterojunctions and plasma resonance effects）.Subsequently,applications of alloyed BiOX in energy and environmental fields are summarized,including contaminant degradation,antibacterial,CO2reduction,nitrogen fixation and organic synthesis.Finally,we summarize the current challenges and future directions of alloyed BiOX.It is expected that this work will provide guidance and assistance for an in-depth study and understanding of the mechanisms of alloying engineering to optimize intrinsic properties and design alloyed BiOX with higher photocatalytic activity.

A Two-Layer Encoding Learning Swarm Optimizer Based on Frequent Itemsets for Sparse Large-Scale Multi-Objective Optimization

Traditional large-scale multi-objective optimization algorithms(LSMOEAs)encounter difficulties when dealing with sparse large-scale multi-objective optimization problems(SLM-OPs)where most decision variables are zero.As a result,many algorithms use a two-layer encoding approach to optimize binary variable Mask and real variable Dec separately.Nevertheless,existing optimizers often focus on locating non-zero variable posi-tions to optimize the binary variables Mask.However,approxi-mating the sparse distribution of real Pareto optimal solutions does not necessarily mean that the objective function is optimized.In data mining,it is common to mine frequent itemsets appear-ing together in a dataset to reveal the correlation between data.Inspired by this,we propose a novel two-layer encoding learning swarm optimizer based on frequent itemsets(TELSO)to address these SLMOPs.TELSO mined the frequent terms of multiple particles with better target values to find mask combinations that can obtain better objective values for fast convergence.Experi-mental results on five real-world problems and eight benchmark sets demonstrate that TELSO outperforms existing state-of-the-art sparse large-scale multi-objective evolutionary algorithms(SLMOEAs)in terms of performance and convergence speed.
