Anti-osteosarcoma trimodal synergistic therapy using NiFe-LDH and MXene nanocomposite for enhanced biocompatibility and efficacy

Osteosarcoma is usually resistant to immunotherapy and,thus primarily relies on surgical resection and high-dosage chemotherapy.Unfortunately,less invasive or toxic therapies such as photothermal therapy(PTT)and chemodynamic therapy(CDT)generally failed to show satisfactory outcomes.Adequate multimodal therapies with proper safety profiles may provide better solutions for osteosarcoma.Herein,a simple nanocomposite that synergistically combines CDT,PTT,and chemotherapy for osteosarcoma treatment was fabricated.In this composite,small 2D Ni Fe-LDH flakes were processed into 3D hollow nanospheres via template methods to encapsulate 5-Fluorouracil(5-FU)with high loading capacity.The nanospheres were then adsorbed onto larger 2D Ti3C2MXene monolayers and finally shielded by bovine serum albumin(BSA)to form 5-FU@Ni Fe-LDH/Ti3C2/BSA nanoplatforms(5NiTiB).Both in vitro and in vivo data demonstrated that the 5-FU induced chemotherapy,Ni Fe-LDH driven chemodynamic effects,and MXene-based photothermal killing collectively exhibited a synergistic“all-in-one”anti-tumor effect.5NiTiB improved tumor suppression rate from<5%by 5-FU alone to~80.1%.This nanotherapeutic platform achieved higher therapeutic efficacy with a lower agent dose,thereby minimizing side effects.Moreover,the composite is simple to produce,enabling the fine-tuning of dosages to suit different requirements.Thus,the platform is versatile and efficient,with potential for further development.

Cognitive impairment in cerebral small vessel disease induced by hypertension

Hypertension is a primary risk factor for the progression of cognitive impairment caused by cerebral small vessel disease,the most common cerebrovascular disease.Howeve r,the causal relationship between hypertension and cerebral small vessel disease remains unclear.Hypertension has substantial negative impacts on brain health and is recognized as a risk factor for cerebrovascular disease.Chronic hypertension and lifestyle factors are associated with risks for stro ke and dementia,and cerebral small vessel disease can cause dementia and stroke.Hypertension is the main driver of cerebral small vessel disease,which changes the structure and function of cerebral vessels via various mechanisms and leads to lacunar infarction,leukoaraiosis,white matter lesions,and intracerebral hemorrhage,ultimately res ulting in cognitive decline and demonstrating that the brain is the to rget organ of hypertension.This review updates our understanding of the pathogenesis of hypertensioninduced cerebral small vessel disease and the res ulting changes in brain structure and function and declines in cognitive ability.We also discuss drugs to treat cerebral small vessel disease and cognitive impairment.

Photosynthetic response dynamics in the invasive species Tithonia diversifolia and two co-occurring native shrub species under fluctuating light conditions

To determine the invasiveness of invasive plants,many studies have compared photosynthetic traits or strategies between invasive and native species.However,few studies have compared the photosynthetic dynamics between invasive and native species during light fluctuations.We compared photosynthetic induction,relaxation dynamics and leaf traits between the invasive species,Tithonia diversifolia and two native species,Clerodendrum bungei and Blumea balsamifera,in full-sun and shady habitats.The photosynthetic dynamics and leaf traits differed among species.T.diversifolia showed a slower induction speed and stomatal opening response but had higher average intrinsic water-use efficiency than the two native species in full-sun habitats.Thus,the slow induction response may be attributed to the longer stomatal length in T.diversifolia.Habitat had a significant effect on photosynthetic dynamics in T.diversifolia and B.balsamifera but not in C.bungei.In shady habitat,T.diversifolia had a faster photosynthetic induction response than in full-sun habitat,leading to a higher average stomatal conductance during photosynthetic induction in T.diversifolia than in the two native species.In contrast,B.balsamifera had a larger stomatal length and slower photosynthetic induction and relaxation response in shady habitat than in full-sun habitat,resulting in higher carbon gain during photosynthetic relaxation.Nevertheless,in both habitats,T.diversifolia had an overall higher carbon gain during light fluctuations than the two native species.Our results indicated that T.diversifolia can adopt more effective response strategies under fluctuating light environments to maximize carbon gain,which may contribute to its successful invasion.

Emerging mechanisms and implications of c GAS-STING signaling in cancer immunotherapy strategies

The intricate interplay between the human immune system and cancer development underscores the central role of immunotherapy in cancer treatment.Within this landscape,the innate immune system,a critical sentinel protecting against tumor incursion,is a key player.The cyclic GMP-AMP synthase(c GAS)and stimulator of interferon genes(STING)pathway has been found to be a linchpin of innate immunity:activation of this signaling pathway orchestrates the production of type I interferon(IFN-α/β),thus fostering the maturation,differentiation,and mobilization of immune effectors in the tumor microenvironment.Furthermore,STING activation facilitates the release and presentation of tumor antigens,and therefore is an attractive target for cancer immunotherapy.Current strategies to activate the STING pathway,including use of pharmacological agonists,have made substantial advancements,particularly when combined with immune checkpoint inhibitors.These approaches have shown promise in preclinical and clinical settings,by enhancing patient survival rates.This review describes the evolving understanding of the c GAS-STING pathway's involvement in tumor biology and therapy.Moreover,this review explores classical and non-classical STING agonists,providing insights into their mechanisms of action and potential for optimizing immunotherapy strategies.Despite challenges and complexities,the c GAS-STING pathway,a promising avenue for enhancing cancer treatment efficacy,has the potential to revolutionize patient outcomes.

Construction and properties of venlafaxine hydrochloride sustained release system based on hollow mesoporous silica microspheres

The aim of this work is to develop a venlafaxine hydrochloride sustained release system based on hollow mesoporous silica microspheres(HMSMs).HMSMs were innovatively prepared with tetraethyl silicate(TEOS)as the precursor and volatile n-heptane as a soft template.The obtained HMSMs show a well-defined hollow structure with an average size of 967 nm and pore volume of 0.85 cm^(3)/g,implying it is a potential drug carrier.Subsequently,venlafaxine hydrochloride(VF)was absorbed in the HMSMs with a content of 37.67% or so.The sustained release effect is further measured by the dissolution in-strument at 37℃ and 50 rpm in ultrapure water.The results showed that the HMSMs/VF system shows good sustained release properties compared with sustained release tablets with hydroxypropyl meth-ylcellulose as the main component.This HMSMs sustained release system appears to be a promising candidate for a sustained drug release.

Aqueous sodium-ion batteries(ASIBs)and aqueous potassium-ion batteries(APIBs)present significant potential for large-scale energy storage due to their cost-effectiveness,safety,and environmental compatibility.Nonetheless,the intricate energy storage mechanisms in aqueous electrolytes place stringent require-ments on the host materials.Prussian blue analogs(PBAs),with their open three-dimensional framework and facile synthesis,stand out as leading candidates for aqueous energy storage.However,PBAs possess a swift capacity fade and limited cycle longevity,for their structural integrity is compromised by the pronounced dis-solution of transition metal(TM)ions in the aqueous milieu.This manuscript provides an exhaustive review of the recent advancements concerning PBAs in ASIBs and APIBs.The dissolution mechanisms of TM ions in PBAs,informed by their structural attributes and redox processes,are thoroughly examined.Moreover,this study delves into innovative design tactics to alleviate the dissolution issue of TM ions.In conclusion,the paper consolidates various strategies for suppressing the dissolution of TM ions in PBAs and posits avenues for prospective exploration of high-safety aqueous sodium-/potassium-ion batteries.

Cu-substituted nickel hexacyanoferrate with tunable reaction potentials for superior ammonium ion storage

In this work,a variety of Cu_(x)Ni_(2-x)Fe(CN)_(6)(x=0,0.4,0.8,1.2,1.6,2)cathodes for ammonium ion batteries are prepared and their electrochemical performances are investigated.During the introduction of copper in nickel hexacyanoferrate,the electrochemical performance varies without changing the structure of nickel hexacyanoferrate.The increase of Cu content in nickel hexacyanoferrate leads to the enhancement of reaction potential and capacity.Electrochemical results suggest that the substitution of Cu for Ni has a positive effect on improving the cycling stability and rate capacity of nickel hexacyanoferrate when x in Cu_(x)Ni_(2-x)Fe(CN)_(6)is less than 0.4.Therefore,Cu_(0.4)Ni_(1.6)Fe(CN)_(6)exhibits the best cycling per-formance(capacity retention of 97.54%at 0.3 C)and the highest rate capacity(41.4 mAh g^(-1)at 10 C)in Cu_(x)Ni_(2-x)Fe(CN)_(6).Additionally,the X-ray diffraction(XRD)and X-ray photoelectron spectroscopy(XPS)tests also reveal that the structural evolution of Cu_(0.4)Ni_(1.6)Fe(CN)_(6)is highly reversible upon NH_(4)^(+)storage.Therefore,this work proposes a candidate material for ammoniumion batteries and offers a novel avenue for adjusting the operating potential of the material.

Influence of Anteroposterior Symmetrical Aero-Wings on the Aerodynamic Performance of High-Speed Train

The running stability of high-speed train is largely constrained by the wheel-rail coupling relationship,and the continuous wear between the wheel and rail surfaces will profoundly affect the dynamic performance of the train.In recent years,under the background of increasing train speed,some scientific researchers have proposed a new idea of using the lift force generated by the aerodynamic wings(aero-wing)installed on the roof to reduce the sprung load of the carriage in order to alleviate the wear and tear of the wheel and rail.Based on the bidirectional running characteristics of high-speed train,this paper proposes a scheme to apply aero-wings with anteroposterior symmetrical cross-sections on the roof of the train.After the verification of the wind tunnel experimental data,the relatively better airfoil section and extension formof anteroposterior symmetrical aero-wing is selected respectively in this paper,and the aero-wings are fixedly connected to the roof of the train through the mounting column to conduct aerodynamic simulation analysis.The research shows that:compared with the circular-arc and oval crosssections,this paper believes that the crescent cross-section can form greater aerodynamic lift force in a limited space.Considering factors such as aerodynamic parameters,ground effect,and manufacturing process,this paper proposes to adopt aero-wings with arc type extension form and connect them to the roof of the train through mounting columns with shuttle cross-section.When the roof of the train is covered with aero-wings and runs at high speed,the sprung load of the carriages can be effectively reduced.However,there are certain hidden dangers in the tail carriage due to the large amount of lift force,so,the intervention of the aero-wing lifting mechanism is required.At the same time,it is necessary to optimize the overall aerodynamic drag force reduction in the followup work.

CuInS_(2)quantum-dot-modified g-C_(3)N_(4)S-scheme heterojunction photocatalyst for hydrogen production and tetracycline degradation

CuInS_(2)quantum-dot(CIS QD)-modified g-C_(3)N_(4)(CN)catalysts(CIS/CN)were prepared with the aid of an in-situ growth process.The as-obtained photocatalysts were explored by measuring their crystallinity,surface morphology,binding energy and light absorption activity.The photocatalytic efficiency of the pho-tocatalysts was evaluated through photocatalytic water splitting for hydrogen production and tetracycline(TC)antibiotic degradation under the simulated solar light and visible light respectively.The optimized sample(10CIS/CN)showed the best photocatalytic activity:producing 102.4μmol g^(-1)h^(-1)of hydrogen in 1 h,or degrading 52.16%of TC in 120 min,which were respectively 48 or 3.4 times higher than the photocatalytic activity of CN itself.The enhancement in the efficiency of the composite system was prin-cipally accredited to the enlargement of light absorption,the more effective in charge transfer and the dropping of the charge carrier pair recombination through a formed S-scheme heterojunctional interface.This work is an effort to adjust CN-based polysulfide QD for speedy photocatalysis.The enriched photo-catalytic activity grants a new sense for adjusting the optical properties of CN.
