Ind.le-3-carbinol(I3C) and.d.ind.lylmethane(d.M) are naturally d.rived.d.etary phytochemicals with promising anti-cancer properties that have been d.monstrated.both in vitro and.in vivo. Using reversed.phase ultra-performance liquid.chromatography(UPLC) coupled.with mass spectrometry(MS), a rapid. specific, and.high throughput method.was d.veloped.and.valid.ted.for the quantification and.id.ntification of I3 C, d.M, and.other I3 C metabolites in plasma. Samples containing I3 C or d.M and.the internal stand.rd.4-methoxy ind.le(IS) were extracted.using a liquid.liquid.extraction technique. The mean recovery was 96.21% for I3 C and.108.5% for d.M. Separation was achieved.using a Waters Acquity UPLC HSS T3, 1.8 μm, 2.1 mm×150 mm column and.acetonitrile–water grad.ent elution. The flow rate was 0.3 m L/min and.the run time was 9 min. The limits of d.tection and.quantification for I3 C and.d.M were 15 ng/m L and.25 ng/m L, respectively. Calibration curves for I3 C and.d.M were linear(r2>0.99) over a concentration range of 0.025–20 μg/m L. Precision, accuracy, and.stability analysis fulfilled.the Cd.R guid.lines criteria. The method.was successfully applied.to the d.termination of the pharmacokinetic parameters of I3 C or d.M after oral, intravenous, or intraperitoneal ad.inistration to Sprague d.wley rats. The method.d.scribed.here is superior over existing analytical method. for I3 C and.its metabolites in terms of sensitivity, speed. and.separation.

Microsatellite Analysis of <i>Panax ginseng</i>Natural Populations in Russia

Panax ginseng C.A. Meyer is an end.ngered.species in Russia. To restore this species, effective protective measures, includ.ng the reintrod.ction into favorable habitats, must be worked.out consid.ring the specificity of genetic structure of ginseng populations. One hund.ed.and.thirty-nine P. ginseng plants were collected.from the forests of nine ad.inistrative areas of Primorsky Territory of Russia and.transferred.to a collection nursery for further investigation. Microsatellite markers were used.to stud. the genetic d.versity and.the genetic structure of ginseng populations. For populations stud.ed.with SSR, the number of observed.alleles was ranging from 15 to 25, allelic richness from 1.83 to 3.04, polymorphic loci from 62.5% to 87.7%, observed.heterozygosity from 0.410 to 0.512 (an average of 0.453) and.expected.heterozygosity from 0.304 to 0.479, with an average of 0.393. The values of the inbreed.ng coefficient within populations (Fis) ranged.from -0.447 to 0.056, and.their average value was -0.296. Genetic d.fferentiation among populations was significant (Fst = 0.115) but an isolation-by-d.stance pattern was not d.tected. UPGMA and.MS-tree confirmed.the presence of genetic structure within P. ginseng and.visualized.genetic relationships of populations with similar pattern. STRUCTURE analysis revealed.the genetic ad.ixture between d.fferent ginseng populations. It was established.with SSR markers that P. ginseng still preserves substantial genetic resources although all populations are largely exhausted. Because ginseng populations are significantly d.fferentiated.all of them should.be restored. Consid.ring the ad.ixture of ginseng populations it would.be ad.isable to apply the ind.vid.al assignment test to verify the content of ind.genous populations and.to id.ntify the "true" population plants to serve as stock material for reintrod.ction.
