Balbiani body （BB） is a large distinctive organelle aggregate uniquely present in developing oocytes of diverse animal species. BB is thought as a stage-specific structure that resembles germ plasm, the cytoplasmic organelle of germ cells. The role and function of BB have remained speculative because of a highly dynamic structure and a lack of genetic and molecular data. BB has been found to contain proteins and RNAs, none of them -- except the zebrafish foxH1 RNA, is or encodes a transcription factor. Here we report in the fish medaka （Oryzias latipes） that RNAs encoding microphthalmia-associated transcription factor （Mitf） are prominent components of the BB. By fluorescence in situ hybridization on ovarian section, we revealed that the transcripts of both mitfl and mitf2 genes concentrated in the BB, in which they co-localized with the dazl RNA, a definitive BB marker highly conserved in vertebrates. Therefore, the mitfproduct may play dual roles in germ gene transcription and BB formation and/or function in this organism. Our data provide the second evidence that the RNA of a transcription factor can be a prominent component of the BB in a vertebrate.

Magnetoreception is a hallmark ability of animals for orientation and migration via sensing and utilizing geomagnetic ***(MagR) and cryptochromes(Cry) have recently been identified as the basis for magnetoreception in ***,it has remained unknown whether MagR and Cry have conserved roles in diverse *** we report the identification and expression of magr and cry genes in the fish medaka(Oryzias latipes).Cloning and sequencing identified a single magr gene,four cry genes and one cry-like gene in *** sequence alignment,chromosomal synteny and gene structure analysis,medaka cry2 and magr were found to be the orthologs of human Cry2 and Magr,with cry1 aa and crylab being coorthologs of human ***,magr and cry2 have remained as single copy genes,whereas cry1 has undergone two rounds of gene duplication in ***,magr and cry genes were detected in various stages throughout embryogenesis and displayed ubiquitous expression in adult organs rather than specific or preferential expression in neural organs such as brain and ***,magr knockdown by morpholino did not produce visible abnormality in developing embryos,pointing to the possibility of producing viable magr knockouts in medaka as a vertebrate model for magnet biology.

Magnetoreception is essential for magnetic orientation in animal migration. The molecular basis for magnetoreception has re- cently been elucidated in fruitfly as complexes between the magnetic receptor magnetoreceptor （MagR） and its ligand crypto- chrome （Cry）. MagR and Cry are present in the animal kingdom. However, it is unknown whether they perform a conserved role in diverse animals. Here we report the identification and expression of zebrafish MagR and Cry homologs towards under- standing their roles in lower vertebrates. A single rnagr gene and 7 cry genes are present in the zebrafish genome. Zebrafish has four cryl genes （crylaa, crylab, crylba and cry]bb） homologous to human CRY1 and a single ortholog of human CRY2 as well as 2 cry-like genes （cry4 and cryS）. By RT-PCR, magr exhibited a high level of ubiquitous RNA expression in embryos and adult organs, whereas cry genes displayed differential embryonic and adult expression. Importantly, magr depletion did not produce apparent abnormalities in organogenesis. Taken together, magr and cry2 exist as a single copy gene, whereas cryl exists as multiple gene duplicates in zebrafish. Our result suggests that magr may play a dispensable role in organogenesis and predicts a possibility to generate rnagr mutants for analyzing its role in zebrafish.

Stem cells are present in developing embryos and adult tissues of multicellular organisms. Owing to their unique features, stem cells provide excellent opportunities for experimental analyses of basic developmental processes such as pluripotency control and cell fate decision and for regenerative medicine by stem cell-based therapy. Stem cell cultures have been best studied in 3 vertebrate organisms. These are the mouse, human and a small laboratory fish called medaka. Specifically, medaka has given rise to the first embryonic stem (ES) cells besides the mouse, the first adult testis-derived male stem cells spermatogonia capable of test-tube sperm production, and most recently, even haploid ES cells capable of producing Holly, a semi-cloned fertile female medaka from a mosaic oocyte created by microinjecting a haploid ES cell nucleus directly into a normal oocyte. These breakthroughs make medaka a favoring vertebrate model for stem cell research, the topic of this review.

鱼类性别决定和性分化呈多元性,既有雌雄同体也有雌雄异体。性腺的雌雄性分化过程受遗传和环境因素(如温度、光照、激素和p H值等)影响,具有可逆可塑性。随着生物技术和基因组学的迅速发展,近年来脊椎动物性别决定和性分化的研究有了重大进展和显著突破。本文通过聚焦青鳉及其他硬骨鱼纲保守存在的dmrt1、gsdf、amh等雄性因子,探讨硬骨鱼普遍存在的雌雄性别可塑可逆信号通路,并介绍了新的基因组编辑和性控育种技术,为单性选育等水产养殖技术的研发提供参考。

Ribosome biogenesis in the nucleolus requires numerous nucleolar proteins and small non-coding *** them is ribosome biogenesis factor Bmsl,which is highly conserved from yeast to *** yeast,Bmsl initiates ribosome biogenesis through recruiting Rcll to ***,little is known about the biological function of Bmsl in *** we report that Bmsl plays an essential role in zebrafish liver *** identified a zebrafish bms1l^（sq163） mutant which carries a T to A mutation in the gene bmsl-like（bms1l）.This mutation results in L^（152） to Q^（152） substitution in a GTPase motif in ***,bmsll^（sq163） mutation confers hypoplasia specifically in the liver,exocrine pancreas and intestine after 3 days post-fertilization（dpf）.Consistent with the bmsll^（sq163） mutant phenotypes,whole-mount in situ hybridization（WISH） on wild type embryos showed that bmsll transcripts are abundant in the entire digestive tract and its accessory *** for phospho-Histone 3（P-H3） and TUNEL assay revealed that impairment of hepatoblast proliferation rather than cell apoptosis is one of the consequences of bms1l（sq163） giving rise to an under-developed ***,our findings demonstrate that Bmsll is necessary for zebrafish liver development.
