Malnutrition in early life increases the risk of osteoporosis,but the association of early-life undernutrition combined with adulthood obesity patterns with low-energy fracture remains unknown.This study included 5323 community-dwelling subjects aged⩾40 years from China.Early-life famine exposure was identified based on the participants’birth dates.General obesity was assessed using the body mass index(BMI),and abdominal obesity was evaluated with the waist-to-hip ratio(WHR).Low-energy fracture was defined as fracture occurring after the age of⩾40 typically caused by falls from standing height or lower.Compared to the nonexposed group,the group with fetal,childhood,and adolescence famine exposure was associated with an increased risk of fracture in women with odds ratios(ORs)and 95%confidence intervals(CIs)of 3.55(1.57–8.05),3.90(1.57–9.71),and 3.53(1.05–11.88),respectively,but not in men.Significant interactions were observed between fetal famine exposure and general obesity with fracture among women(P for interaction=0.0008).Furthermore,compared with the groups with normal BMI and WHR,the group of women who underwent fetal famine exposure and had both general and abdominal obesity had the highest risk of fracture(OR,95%CI:3.32,1.17–9.40).These results indicate that early-life famine exposure interacts with adulthood general obesity and significantly increases the risk of low-energy fracture later in life in women.

Genome-wide analysis of UGT gene family identified key gene for the biosynthesis of bioactive flavonol glycosides in Epimedium pubescens Maxim

Epimedium pubescens Maxim.is a well-known traditional Chinese medicinal herb with flavonol glycosides as the major pharmaceutically active compounds.UDP-glycosyltransferases(UGTs)are a group of enzymes responsible for the glycosylation of flavonoid glycosides.In this study,a genome-wide analysis was performed to identify UGT family genes in E.pubescens.As a result,a total of 339 putative UGT genes were identified,which represents the largest UGT gene family known thus far,implying a significant expansion of the UGT gene family in E.pubescens.All EpUGTs were unevenly distributed across six chromosomes,and they were classified into 17 major groups.The expression profiles showed that UGT genes were differentially expressed in roots,leaves,flowers,shoots and fruits.In particular,several EpUGTs were highly induced by high light intensity,which was consistent with the accumulation level of bioactive flavonoids in E.pubescens.Six UGT79 genes that were preferentially expressed in roots or leaves were successfully expressed in E.coli,and only the recombinant EpGT60 protein was found to be active toward 8-prenylkaempferol and icaritin to produce the key bioactive compounds baohuoside II and baohuoside I.The optimal temperature,pH,k_(m) and V_(max) were determined for the recombinant EpGT60 protein.In addition,expression of recombinant EpGT60 in E.coli cell culture led to successful production of baohuoside II when fed 8-prenylkaempferol.Our study provides a foundation for further functional characterization of UGT genes in E.pubescens and provides key candidate genes for bioengineering bioactive flavonoids in E.pubescens.

Interactions between H_(2)O_(2)and dissolved organic matter during granular activated carbon-based residual H_(2)O_(2)quenching from the upstream UV/H_(2)O_(2)process

Granular activated carbon(GAC)filtration can be employed to synchronously quench residual H_(2)O_(2)from the upstream UV/H_(2)O_(2)process and further degrade dissolved organicmatter(DOM).In this study,rapid small-scale column tests(RSSCTs)were performed to clarify the mechanisms underlying the interactions between H_(2)O_(2)and DOM during the GAC-based H_(2)O_(2)quenching process.It was observed that GAC can catalytically decompose H_(2)O_(2),with a long-lasting high efficiency(>80%for approximately 50,000 empty-bed volumes).DOM inhibited GAC-based H_(2)O_(2)quenching via a pore-blocking effect,especially at high concentrations(10 mg/L),with the adsorbed DOM molecules being oxidized by the continuously generated·OH;this further deteriorated the H_(2)O_(2)quenching efficiency.In batch experiments,H_(2)O_(2)could enhance DOM adsorption by GAC;however,in RSSCTs,it deteriorated DOM removal.This observation could be attributed to the different·OH exposure in these two systems.It was also observed that aging with H_(2)O_(2)and DOM altered the morphology,specific surface area,pore volume,and the surface functional groups of GAC,owing to the oxidation effect of H_(2)O_(2)and·OH on the GAC surface as well as the effect of DOM.Addi-tionally,the changes in the content of persistent free radicals in the GAC samples were insignificant following different aging processes.This work contributes to enhancing understanding regarding the UV/H_(2)O_(2)-GAC filtration scheme,and promoting the application in drinking water treatment.

目的系统评价老年抑郁与轻度认知障碍的相关性。方法计算机检索CNKI、VIP、WanFang Data,Cochrane Library,EMbase,Pubmed和Web of Science数据库,搜集有关老年抑郁与轻度认知障碍相关性的队列研究,检索时限均从建库至2023年7月6日。由2名研究者独立完成筛选文献、数据提取及质量评价后,采用Review Manager5.4软件进行Meta分析。结果最终共纳入10篇文献,总样本量为26813人。Meta分析结果显示:老年抑郁与轻度认知障碍具有相关性(OR=1.47,95%CI:1.25~1.72,P<0.001)。结论老年抑郁是轻度认知障碍的发病危险因素之一。上述结论受研究类型、质量及数量的限制,仍待更多高质量研究进行验证。

我国工业园区污水厂进水碳氮比(C/N)普遍较低,常需补充碳源以提高脱氮效果。酒糟富含蛋白质、碳水化合物等有机组分,可生物利用性好,但目前缺乏其作为缓释碳源的研究。本研究以工业园区酒糟固废为原材料,以聚乙烯醇(PVA)、海藻酸钠(SA)为骨架材料,利用低温冷冻化学交联法制备复合缓释碳源,并进行释碳性能和反硝化性能评估。结果表明:通过骨架材料配比以及乳化剂优化研究,缓释碳源的快速释放期可延长到3 d,此阶段释碳过程为骨架溶蚀机制,单位质量缓释碳源的累积释碳量(以化学需氧量,即COD计)可达到1089 mg·(g·L)^(-1)。在酒糟用量为10 g·L^(-1),骨架材料配比为PVA:SA=8:1,乳化剂为1.0%span80条件下制备的缓释碳源在投加量为0.19 g·L^(-1),初始硝态氮(NO_(3)^(-)-N)为(41.53±0.1)mg·L^(-1)时,反硝化出水溶解性有机物(DOM)的腐殖化程度最低、分子质量最小、芳香环取代基种类和取代基程度最低,总氮去除率为99.2%,反硝化速率达到4.08 mg·(L·h)^(-1),为最佳缓释碳源。本研究结果可为工业园区的污水-固废协同治理提供参考。

目的探讨左归丸含药血清对成骨-破骨细胞共培养体系中Hedgehog-GLi通路关键因子mRNA表达及骨吸收功能的影响。方法建立成骨细胞、破骨细胞、骨磨片共培养体系,实验分为7组:空白对照组、假手术组、OVX组、阳性对照组、左归丸组、激动剂组和激动剂+左归丸组。选用50只雌性SD大鼠,分别制备相对应组含药血清,实验各组分别加入相对应组大鼠血清。通过实时荧光定量PCR检测成骨细胞Ihh、Ptch、Smo、GLi1、OPG及RANKL的mRNA表达;并采用甲苯胺蓝染色方法检测破骨细胞骨吸收功能。结果与假手术组相比,OVX组Ihh、Ptch、Smo、GLi1mRNA表达显著上调,同时OPGmRNA表达水平显著降低,RANKL mRNA表达水平显著增加,RANKL/OPG比值明显增高。与OVX组相比,阳性对照组与左归丸组Ihh、Ptch、Smo、GLi1mRNA表达明显下调,同时OPG mRNA表达水平显著升高,RANKL mRNA表达水平显著降低,RANKL/OPG比值明显下降。OVX组加入激动剂Shh重组蛋白后,Hedgehog-GLi信号通路关键因子的mRNA表达较OVX组明显增加。与激动剂组比较,激动剂+左归丸组Ihh、Ptch、Smo、GLi1mRNA表达以及RANKL/OPG比值均明显下降。与假手术组相比,OVX组的骨吸收陷窝面积显著增加;与OVX组相比,阳性对照组和左归丸组的骨吸收陷窝面积显著下降。结论左归丸含药血清可一定程度上抑制Hedgehog-GLi通路关键因子Ihh、Ptch、Smo、GLi1的活性,进而抑制OVX所致的骨吸收增强。

目的探讨左归丸含药血清对成骨细胞中Hedgehog-GLi通路关键蛋白表达及破骨细胞数量的影响。方法选用50只雌性SD大鼠,随机分为空白对照组、假手术组、造模组。造模组大鼠切除双侧卵巢,12周后随机分为卵巢切除组(ovariectomy,OVX)、阳性对照组和左归丸组。阳性对照组大鼠灌服戊酸雌二醇混悬液,左归丸组大鼠灌服左归丸水煎液,连续干预7 d。末次给药1.5 h后,腹主动脉取血制备各组相应含药血清。建立成骨细胞、破骨细胞、骨磨片共培养体系,分为空白对照血清组、假手术血清组、OVX血清组、阳性对照血清组、左归丸含药血清组、激动剂组和激动剂+左归丸含药血清组,分别添加各组对应血清。免疫荧光法检测成骨细胞印度刺猬蛋白(Indian hedgehog,Ihh)、碎片蛋白(patched,Ptch)、润滑蛋白(smoothened,Smo)及GLi1蛋白表达;免疫组化法检测成骨细胞骨保护蛋白(osteoprotegerin,OPG)、核因子受体激活因子-κB配体(receptor activator for nuclear factor-κB ligand,RANKL)蛋白表达;对破骨细胞进行TRAP染色,计算TRAP+细胞数;ELISA检测共培养体系上清液中TRAP的含量。结果与假手术血清组比较,OVX血清组Ihh、Ptch、Smo、GLi1蛋白表达水平显著上调(P<0.01),RANKL/OPG比值增高;TRAP+细胞数和细胞上清液中TRAP含量亦明显增加(P<0.05)。与OVX血清组比较,左归丸含药血清组Ihh、Ptch、Smo、GLi1的蛋白表达明显下调,RANKL/OPG比值下降;TRAP+细胞数和细胞上清液中TRAP含量明显降低(P<0.05)。与激动剂组比较,加入左归丸含药血清后,Ihh、Ptch、Smo、GLi1的蛋白表达及RANKL/OPG比值均明显下降,TRAP+细胞个数及TRAP含量亦明显降低(P<0.05)。结论左归丸含药血清可通过抑制异常活跃的Hedgehog-GLi信号通路,使Ihh、Ptch、Smo、GLi1的蛋白表达水平下调,降低RANKL/OPG比值,抑制破骨细胞的数量和活性,起到减少骨丢失作用。

Background and Aims:Continuous release and transmission of hepatitis B virus(HBV)is one of the main factors leading to chronic hepatitis B(CHB)infection.However,the mechanism of HBV-host interaction for optimal viral transport is unclear.Hence,we aimed to explore how HBV manipulates microtubule-associated protein 1S(MAP1S)and microtubule(MT)to facilitate its transport and release.Methods:The expression of MAP1S or acetylated MT was investigated by immunofluorescence,RT-PCR,immunoblotting,and plasmid transfection.MAP1S overexpression or knockdown was performed by lentiviral infection or shRNA transfection,respectively.HBV DNA was quantified using q-PCR.Results:Significantly higher level of MAP1S in HepG2215 cells compared with HepG2 cells was detected using RT-PCR(p<0.01)and immunoblotting(p<0.001).Notably,stronger MAP1S expression was observed in the liver tissues of patients with CHB than in healthy controls.MAP1S overexpression or knockdown demonstrated that MAP1S promoted MT acetylation and reduced the ratio of HBV DNA copies inside to outside cells.Further,transfection with the hepatitis B virus X protein(HBx)-expressing plasmids induced significantly higher level of MAP1S than that in controls(p<0.0001),whereas HBVX−mutant-encoding HBV proteins(surface antigen,core protein,and viral DNA polymerase)hardly affected its expression.Conclusions:These results demonstrate that HBx induces the forma tion of stable MTs to promote the release of HBV particles through upregulating MAP1S.Thus,our studies delineate a unique molecular pathway through which HBV manipulates the cytoskeleton to facilitate its own transportation,and indicate the possibility of targeting MAP1S pathway for treatment of patients with CHB.

Identification and fine mapping of a fertility restorer gene for wild abortive cytoplasmic male sterility in the elite indica rice non-restorer line 9311

The wild abortive(WA)-type cytoplasmic male sterility(CMS)derived from the wild rice species Oryza rufipogon Griff.is used widely in three-line indica hybrids.The identification and mapping of restorer of fertility(Rf)genes aided in the development of WA-type hybrids.Here we report that testcross F1 plants from the WA-type CMS line and 9311 exhibited stainable pollen grains with no seed set,indicating that 9311 carries minor-effect Rfs for WA-type CMS.We developed an advanced backcross population consisting of plants harboring small regions of donor chromosomal segments from 9311 in the WATianfeng A genetic background with moderate seed setting rates.Genetic analysis showed that the pollen fertility levels of the backcross individuals are governed by a single gene from 9311 that we named Rf19(t).By use of the RICE 40 K gene chip,three introduced segments were identified in the fertile lines,and a candidate region spanning 4.37–8.29 Mb on chromosome 1 was identified for Rf19(t).Finally,Rf19(t)was fine-mapped to a region of 90 kb between the DNA marker loci STS1-163 and STS1-183,in which eight ORFs were predicted.Also,using relative expression analyses,comparative sequence analyses and functional domain analyses,we identified LOC_Os01g10530 as the most likely candidate gene for Rf19(t).Furthermore,Rf19(t)was found to function in fertility restoration,most probably by regulating the degradation of m RNA transcribed from the mitochondrial gene WA352.These results increase our knowledge of fertility restoration in WA-type CMS lines and will facilitate the development of high-quality pairs of WAtype CMS and maintainer lines.
