This research has been undertaken to screen availability as a chemosensitizer resource of 14 local citrus spp. cultivating in Geju Island, Korea. We report that methanol extracts from fruits and flower have a chemosensitizing effect that can reverse P-glycoprotein(Pgp)-mediated multidrug-resistance(MDR). MDR reversing activity of citrus extracts using drug sensitive AML-2/WT and resistant AML-2/D100 of human acute myelogenous leukemia cells was more strong immature fruits than mature fruits and flower. In the several immature fruits with high MDR reversing activity, treatment of drug-resistant AML-2/D100 cells with its extracts increased remarkably their cytotoxicity to vincristine, resultantly causing high chemosensitizing index(CI). Immature Inchangkyool(C. ichangiensis) showed the most high CI of 14 citrus extracts. Also, Hongkyool(C. tachibana), Byungkyool(C. platymamma), Cheongkyool(C. nippokoreana), Jinkyool(C. sunki) of immature fruits strongly potentiated vincristine cytotoxicity in AML-2/D100 cells. In addition, peel extracts increased about 2 to 10 fold chemosensitizing effect as compared with whole fruit extract in mature Hongkyool(C. tachibana), Byungkyool(C. platymamma), Inchangkyool(C. ichangiensis). In the case of flower extracts, the mean IC50 value for extracts in AML-2/WT and AML-2/D100 without vincristine was up to 200 μg/ml whereas the range of its mean IC50 value in AML-2/D100 in the presence of vincristine was 18-200μg/ml. CI of flower extracts displayed higher than mature extracts but not immature fruits. This result indicates that immature citrus fruit of C. ichangiensis would contian the active principles which do not exert their activity solely by cytotoxicity. In particular, Inchangkyool(C. ichangiensis), Byungkyool(C. platymamma), and Cheongkyool(C. nippokoreana of 14 local citrus suggest that possibility as effective resources for chemosensitizing compounds isolation by further investigation.

Cloning,expression in Escherichia coli,and enzymatic properties of laccase from Aeromonas hydrophila WL-11

A strain WL-11 with high laccase activity was isolated from activated sludge collected from the effluent treatment plant of a textile and dyeing industry. It was identified as Aeromonas hydrophila by physiological test and 16S rDNA sequence analysis. A gene encoding of laccase from a newly isolated Aeromonas hydrophila WL-11 was cloned and characterized. Nucleotide sequence analysis showed an open reading frame of 1605 bp encoding a polypeptide comprised of 534 amino acids. The primary structure of the enzyme predicted the structural features characteristic of other laccases, including the conserved regions of four histidine-rich copper-binding sites. The predicted amino acid sequence showed a high homology （more than 60%） with bacterial laccases in the genome and protein databases and the highest degree of similarity （61% identity） was observed with the multicopper oxidase of KlebsieUa sp. 601. When expressed in Escherichia coli, the recombinant enzyme was overproduced in the cytoplasm as soluble and active form. The purified enzyme had an optimum pH of 2.6 and 8.0 for ABTS （2,2＇-azino-bis（3-ethylbenzthiazolinesulfonic acid） and DMP （2,6-dimethoxyphenol）, respectively. The kinetic study on ABTS revealed a higher affinity of this enzyme to this substrate than DMP.

Analysis of interval constants in calendars affiliated with the Shoushili

We study interval constants that are related to motions of the Sun and Moon, i.e., the Qi, Intercalation, Revolution and Crossing interval, in calendars affiliated with the Shoushi calendar （Shoushili）, such as Datongli and Chiljeongsannaepyeon. It is known that these interval constants were newly introduced in the Shoushili calendar and revised afterward, except for the Qi interval constant, and the revised values were adopted in later calendars affiliated with the Shoushili. We first investigate the accu- racy of these interval constants and then the accuracy of calendars affiliated with the Shoushili in terms of these constants by comparing times for the new moon and the maximum solar eclipse calculated by each calendar with modem methods of calcula- tion. During our study, we found that the Qi and Intercalation interval constants used in the early Shoushili were well determined, whereas the Revolution and Crossing interval constants were relatively poorly measured. We also found that the interval constants used by the early Shoushili were better than those of the later one, and hence better than those of Datongli and Chiljeongsannaepyeon. On the other hand, we found that the early Shoushili is, in general, a worse calendar than Datongli for use in China but a better one than Chiljeongsannaepyeon for use in Korea in terms of times for the new moon and when a solar eclipse occurs, at least for the period 1281 - 1644. Finally, we verified that the times for sunrise and sunset in the Shoushili-Li-Cheng and Mingshi are those at Beijing and Nanjing, respectively.

Organic solar cells for indoor power generation

During last decades,organic solar cells(OSCs)have achieved remarkable advancements via extensive studies on the optimization of photovoltaic donor/acceptor(D/A)materials,device fabrication,and the D/A bulk heterojunction *** conversion efficiencies(PCEs)up to〜16%for single junction cells and over 17%PCEs were successfully demonstrated for tandem OSC devices[1,2].OSCs may find real industrial applications soon,including building integrated photovoltaics,portable power sources and indoor power generation for low power consumption electronic devices(i.e.,internet of things(IOT)sensors).Among them,indoor applications have great potentials because OSCs can convert indoor lights into electricity more efficiently compared to inorganic counterparts[3].

p73 G4C14 to A4T14 polymorphism is associated with colorectal cancer risk and survival

AIM:To analyze the association between the p73 G4C14-to-A4T14 polymorphism(a.k.a.,the GC/AT variation) and colorectal cancer risk and survival in the Korean population,and to evaluate the relationships between p73 polymorphism and the p73 protein expression or clinicopathological characteristics of colorectal ***:Three hundred and eighty-three histologically confirmed cases and 469 healthy controls,recruited at one teaching hospital in Pusan,Korea from 2001 and 2007,were genotyped for p73 G4C14-to-A4T14 by PCR with confronting two-pair primers(PCR-CTPP) and the expression profile of p73 in cancer tissues(n=383) was analyzed by ***:Odds ratios(ORs) and 95% confidence intervals(CIs) were calculated by unconditional logistic regression model adjusted for age and *** with the GC/GC genotypes,the GC/AT and AT/AT genotypes were significantly associated with colorectal cancer risk(GC/AT vs GC/GC:OR = 1.46,95% CI:1.10-1.94;AT/AT vs GC/GC:1.72,0.98-3.03;Ptrend=0.01).When stratified by age and gender,the association was restricted to those less than 60 years of age(GC/AT or AT/AT vs GC/GC:2.22,1.39-3.55) and male(GC/AT or AT/AT vs GC/GC:1.91,1.31-2.77).The expression of p73 was associated with invasion depth(P = 0.003) and advanced Duke's stage(P = 0.06) of colorectal *** patients with the GC/GC genotype were associated with worse survival compared with those with the other genotypes(P = 0.02).However,no signif icant relationship was observed between the p73 G4C14-to-A4T14 polymorphism and p73 protein expression in cancer ***:Our results suggest that the p73 GC/AT polymorphism is associated with an increased colorectal cancer risk and survival in the Korean population.

Isolation and characterization of Pseudomonas otitidis WL-13 and its capacity to decolorize triphenylmethane dyes

Pseudomonas otitidis WL-13, which has a high capacity to decolorize triphenylmethane dyes, was isolated from activated sludge obtained from a wastewater treatment plant of a dyeing industry. This strain exhibited a remarkable color-removal capability when tested against several triphenylmethane dyes under both shaking and static conditions at high concentrations of dyes. More than 95% of Malachite Green and Brilliant Green was removed within 12 h at 500 μmol/L dye concentration under shaking conditions. Crystal Violet lost about 13% of its color under the same conditions tested. The rate of decolorization increased when the M9 medium was supplemented with yeast extract. The optimum pH and temperature for color removal were 7-9 and 35-40℃, respectively. The observed changes in the visible spectra and the inspection of bacterial growth indicated the color-removal by the adsorption of dye to the cells during incubation with strains.

Collision tumors are thought to arise from the accidental meeting and interpenetration of two independent tumors. We report here a highly unusual case of a 61-year old man who had a unique tumor that was composed of a metastatic adenocarcinoma from the stomach to the rectum, which harbored a collision tumor of primary rectal adenocarcinoma. The clonalities of the two histologically distinct lesions of the rectal mass were confirmed by immunohistochemical and molecular analysis. Although histologic examination is the cornerstone in pathology, immunohistochemical and molecular analysis can provide evidence regarding whether tumors originate from the same clone or different clones. To the best of our knowledge, this is the fi rst reported case of such an occurrence.

Interleukin-24 is correlated with differentiation and lymph node numbers in rectal cancer

AIM:To assess the significance of interleukin(IL)-24 and vascular endothelial growth factor(VEGF)expression in lymph-node-positive rectal cancer. METHODS:Between 1998 and 2005,90 rectal adenocarcinoma patients with lymph node involvement were *** patients received radical surgery and postoperative pelvic chemoradiotherapy of 50.4-54.0 *** of 5-fluorouracil and leucovorin or levamisole was given intravenously during the first and last week of radiotherapy,and then monthly for about 6 *** of IL-24 and VEGF was evaluated by immunohistochemical staining of surgical specimens, and their relations with patient characteristics and survival were *** median follow-up of surviving patients was 73 mo(range:52-122 mo). RESULTS:IL-24 expression was found in 81 out of 90 patients;31 showed weak intensity and 50 showedstrong *** expression was found in 64 out of 90 *** and weak intensities of IL-24 expression were classified as negative expression for ***-24 expression was significantly reduced in poorly differentiated tumors in comparison with well or moderately differentiated tumors(P=0.004),N2b to earlier N stages(P=0.016),and stageⅢc to stageⅢ a orⅢb(P=0.028).The number of involved lymph nodes was also significantly reduced in IL-24-positive patients in comparison with IL-24-negative ones. There was no correlation between VEGF expression and patient *** of IL-24 and VEGF was not correlated with survival,but N stage and stages were significantly correlated with survival. CONCLUSION:IL-24 expression was significantly correlated with histological differentiation,and inversely correlated with the degree of lymph node involvement in stageⅢrectal cancer.
