Parquetina nigrescens leaves： chemical profile and influence on the physical and biochemical indices of sexual activity of male Wistar rats

OBJECTIVE： The leaves of Parquetina nigrescens have been claimed in folk medicine to be useful for managing sexual dysfunction, but there is inadequate scientific evidence for this claim. This investigation was conducted to assess the effects of aqueous leaf extract of Parquetina nigrescens （AEPN） in rats induced with sexual dysfunction. METHODS： Male rats were allocated into various groups after being induced into sexual dysfunction with paroxetine hydrochloride. The groups were treated with distilled water, PowMaxM （reference drug）, and the AEPN at 20, 40 and 80 mg/kg body weight （BW） respectively for 7 d. Frequencies of mount （MF）, intromission （IF）, ejaculation （EF）, as well as latencies of mount （ML）, intromission （IL） and post - ejaculatory interval （PEI） were progressively monitored with receptive female rats on days 1, 3 and 7. The precentage of mounted, intromitted, and ejaculated rats, intromission ratio （IR）, copulatory efficiency （CE） and intercopulatory interval （ICI） were also computed. Standard methods were employed to determine the levels of serum luteinizing and follicle stimulating hormones （LH and FSH）, testosterone, nitric oxide （NO） and the activity of phosphodiesterase V （PDE5）. RESULTS： The plant contained alkaloids, saponins, flavonoids, cardiac glycosides, steroids, tannins, phlobatannins, cardenolides, phenolics, anthraquinones and triterpenes with alkaloids （2.32 mg/g） occurring in greatest quantity while flavonoids, anthraquinones and cardenolides （0.01 mg/g, each） were the least. All the 20 standard amino acids were detected in the plant leaf, with the lowest concentration being 0.30 mg/g for methionine and the highest being 2.12 mg/g for cysteine. Furthermore, P. nigrescens leaves contained Na, K, Fe, Mg, Zn, Ca, Cu, Mn, P, Pb, Cd, Ni, Cr, and Co, with Fe present in the highest and Co the lowest concentrations. Objective:To investigate the loxic implications of ethanolic stem bark extract of Azadirachta indica(A-indica)at 50,100,200 and 300 mg/kg body weight in Wistar ***:Fifty male rats of Wistar strains were randomly grouped into five(A-F.)of ten animals *** in Group A(control)were orally administered 1 mL of distilled water on daily basis for 21 days while those in Groups B-E received same volume of the extract corresponding to 50,100,200 and 300mg/kg body ***:The extract did not significantly(P>0.05)alter the levels of albumin,total protein,red blood cells and factors relating to it whereas the white blood cell,platelets,serum triacylglycerol and high-density lipoprotein cholesterol decreased significantly(P<0.05).In contrast,the final body weights,absolute weights of the liver,kidney,lungs and heart as well as their organ-body weight ratios,serum globulins,total and conjugated bilirubin,serum cholesterol,low-density lipoprotein cholesterol and computed atherogenic index increased *** spleen-body weight ratio,alkaline phosphatase,alanine and aspartate transaminases,sodium,potassium,calcium,feed and water intake were altered at specific ***:Overall,the alterations in the biochemical parameters of toxicity have consequential effects on the normal functioning of the organs of the ***,the ethanolic extract of *** stem bark at the doses of 50,100,200 and 300 mg/kg body weight may not be completely safe as an oral remedy and should be taken with caution if absolutely necessary.

This study conducts a systematic literature review(SLR)of blockchain consensus mechanisms,an essential protocols that maintain the integrity,reliability,and decentralization of distributed ledger *** aim is to comprehensively investigate prominent mechanisms’security features and vulnerabilities,emphasizing their security considerations,applications,challenges,and future *** existing literature offers valuable insights into various consensus mechanisms’strengths,limitations,and security vulnerabilities and their real-world ***,there remains a gap in synthesizing and analyzing this knowledge *** this gap would facilitate a structured approach to understanding consensus mechanisms’security and vulnerabilities *** study adheres to Preferred Reporting Items for Systematic Reviews and Meta-Analyses(PRISMA)guidelines and computer science standards and reviewed 3749 research papers from 2016 to 2024,excluding grey literature,resulting in 290 articles for descriptive *** research highlights an increased focus on blockchain consensus security,energy efficiency,and hybrid mechanisms within 60%of research papers post-2019,identifying gaps in scalability,privacy,and interoperability for future *** synthesizing the existing research and identifying the key trends,this SLR contributes to advancing the understanding of blockchain consensus mechanisms’security and guiding future research and structured innovation in blockchain systems and applications.

A numerical study of partial slip boundary condition is investigated. The stagnation-point flow problem involving some physio-chemical parameters has been elucidated. The process involves developing a multivariate mathematical model for the flow and transforming it into a coupled univariate equation. Key parameters of interest in the study are the buoyancy force, the surface stretching, the unsteadiness, the radiation, the dissipation effects, the slip effects, the species reaction and the magnetic field parameters. It is concluded that the impact of physio-chemical factors significantly alters the kinematics of the flow in order to optimally achieve desired product characteristics.

Fusarium graminearum is an important plant pathogenic fungus that causes disease and yield reduction in many cereal crops, such as wheat and barley. Gyp8 stimulates GTP hydrolysis on Ypt1 in yeast. However, the functions of Gyp8 in plant pathogenic fungi are still unknown. In this study, we investigated the roles of Fg Gyp8 in F. graminearum by genetic and pathological analyses. Through gene knockout and phenotypic analyses, we found that Fg Gyp8 is required for vegetative growth in F. graminearum. The conidiation, conidial size and number of septa per conidium of ΔFggyp8 mutant are significantly reduced when compared to the wild type PH-1. Furthermore, Fg Gyp8 is crucial for pathogenicity on wheat coleoptiles and wheat heads. Fg Gyp8 contains a conserved TBC domain. Domain deletion analysis showed that the TBC domain, C-and N-terminal regions of Fg Gyp8 are all important for its biological functions in F. graminearum. Moreover, we showed that Fg Gyp8 catalyzes the hydrolysis of the GTP on Fg Rab1 to GDP in vitro, indicating that Fg Gyp8 is a GTPase-activating protein(GAP) for Fg Rab1. In addition, we demonstrated that Fg Gyp8 is required for Fg Snc1-mediated fusion of secretory vesicles with the plasma membrane in F. graminearum. Finally, we showed that Fg Gyp8 has functional redundancy with another Fg Rab1 GAP, Fg Gyp1, in F. graminearum. Taken together, we conclude that Fg Gyp8 is required for vegetative growth, conidiogenesis, pathogenicity and acts as a GAP for Fg Rab1 in F. graminearum.

Evaluating the Role of Shelterbelt in Vegetation Development in a Semi Arid Zone of Yobe State, Nigeria

This study evaluates the role of shelterbelts on the development and dynamics of vegetation in relation to livelihoods in the study area using geo-information techniques. The study covered the period between 1972 and 2007. The results show that in 1972 vegetation cover in the study area extended over an area of 6955 hectares (i.e.?shrubs, grasses and trees) which represents 65% of the area;however there was a decrease in 1986 to an area of 5779 hectares (54%). Despite the establishment of shelterbelt in the area in 1987, the trend did not change. This is obvious in the reduction of vegetation cover between 1986 and 2000 to a total area of 3893 hectares (37%). The vegetation cover further decreased to 2791 hectares (26%) in 2005 and 1659 hectares (16%) in 2007 respectively. The study concludes that the establishment of shelterbelts alone cannot guarantee the success of vegetation cover development in the area without a sustainable plan which recognizes the local communities as critical stakeholders. Thus, variables like population growth and GDP have a role to play in vegetation cover depletion. The study recommends that efforts geared towards enlightenment of the local communities on the need for conservation should be encouraged by Government and Non-Governmental Organisations.
