Autophagy plays a pro-apoptotic role in arsenic trioxide-induced cell death of liver cancer

Objective: The effects of arsenic trioxide(As2O3) on hepatocellular carcinoma have been documented widely. Autophagy plays dual roles in the survival and death of cancer cells. Therefore, we investigated the exact role of autophagy in As2O3-induced apoptosis in liver cancer ***: The viability of hepatoma cells was determined using the MTT assay with or without fetal bovine serum. The rate of apoptosis in liver cancer cells treated with As2O3was evaluated using flow cytometry, Hoechst 33258 staining, and TUNEL assays. The rate of autophagy among liver cancer cells treated with As2O3was detected using immunofluorescence, Western blot assay and transmission electron ***: Upon treatment with As2O3, the viability of HepG2 and SMMC-7721 cells was decreased in a time-and dose-dependent manner. The apoptosis rates of both liver cancer cell lines increased with the concentration of As2O3, as shown by flow cytometry. Apoptosis in liver cancer cells treated with As2O3was also shown by the activation of the caspase cascade and the regulation of Bcl-2/Bax ***, As2O3treatment induced autophagy in liver cancer cells;this finding was supported by Western blot, immunofluorescence of LC3-II and beclin 1, and transmission electron microscopy. In liver cancer cells, As2O3inhibited the phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR) signal pathway that plays a vital role in both apoptosis and autophagy. The PI3K activator SC-79 partially reversed As2O3-induced autophagy and apoptosis. Furthermore, inhibiting autophagy with 3-methyladenine partially reversed the negative effects of As2O3on cell *** starvation increased autophagy and amplified the effect of As2O3on cell ***: As2O3induces apoptosis and autophagy in liver cancer cells. Deterministic and Robust Volt-var Control Methods of Power System Based on Convex Deep Learning

Volt-var control(VVC)is essentially a non-convex optimization problem due to the non-convexity of power flow(PF)constraints,resulting in the difficulty in obtaining the optimum without convexity *** existing second-order cone method for the convexity conversion often leads to a sharp increase in PF constraints and optimization variables,which in turn increases the optimization difficulty or even leads to optimization *** paper first proposes a deterministic VVC method based on convex deep learning power flow(DLPF).This method uses the input convex neural network(ICNN)to establish a single convex mapping between state parameters and node voltage to complete the convexity conversion while the optimization variables only correspond to reactive power equipment,which can ensure the global optimum with extremely fast computation *** cope with the impact brought by the uncertainty of distributed energy and omit the additional worst scenario search of traditional robust VVC,this paper proposes robust VVC method based on convex deep learning interval power flow(DLIPF),which continues to adopt ICNN to establish another convex mapping between state parameters and node voltage *** DLIPF with DLPF,this method decreases the modeling and optimization difficulty of robust VVC *** results on 30-bus,118-bus,and 200-bus systems prove the correctness and rapidity of the proposed methods.

Comparing short-term outcomes of robot-assisted and conventional laparoscopic total mesorectal excision surgery for rectal cancer in elderly patients

Genome-wide identification of the pectate lyase(PEL)gene family members in Malvaceae,and their contribution to cotton fiber quality

Pectin is a major constituent of the plant cell *** lyase(PEL,EC 4.***.2.2)uses anti-β-elimination chemistry to cleave theα-1,4 glycosidic linkage in the homogalacturonan region of ***,limited information is available on the comprehensive and evolutionary analysis of PELs in the *** this study,we identified 597PEL genes from 10 Malvaceae *** and motif analyses revealed that these PELs are classified into six subfamilies:Clades I,II,III,IV,Va,and *** two largest subfamilies,Clades I and II,contained 237 and222 PEL members,*** members of Clades Va and Vb only contained four or five motifs,far fewer than the other *** duplication analysis showed that segmental duplication played a crucial role in the expansion of the PEL gene family in Gossypium *** PELs from Clades I,IV,Va,and Vb were expressed during the fiber elongation stage,but nearly all PEL genes from Clades II and III showed no expression in any of the investigated fiber developmental *** further performed single-gene haplotype association analysis in 2,*** accessions and 229 *** ***,14 PELs were significantly associated with fiber length and strength traits in *** with superior fiber quality,while only eight GhPEL genes were found to be significantly associated with fiber quality traits in *** findings provide important information for further evolutionary and functional research on the PEL gene family members and their potential use for fiber quality improvement in cotton.

Objective:To evaluate the efficacy and safety of Huashi Baidu Granules(HSBD)in treating patients with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron ***:A single-center retrospective cohort study was conducted during COVID-19 Omicron epidemic in the Mobile Cabin Hospital of Shanghai New International Expo Center from April 1st to May 23rd,*** COVID-19 patients with asymptomatic or mild infection were assigned to the treatment group(HSBD users)and the control group(non-HSBD users).After propensity score matching in a 1:1 ratio,496 HSBD users of treatment group were matched by propensity score to 496 non-HSBD *** in the treatment group were administrated HSBD(5 g/bag)orally for 1 bag twice a day for 7 consecutive *** in the control group received standard care and routine *** primary outcomes were the negative conversion time of nucleic acid and negative conversion rate at day *** outcomes included the hospitalized days,the time of the first nucleic acid negative conversion,and new-onset symptoms in asymptomatic *** events(AEs)that occurred during the study were *** subgroup analysis was conducted in vaccinated(378 HSBD users and 390 non-HSBD users)and unvaccinated patients(118 HSBD users and 106 non-HSBD users).Results:The median negative conversion time of nucleic acid in the treatment group was significantly shortened than the control group[3 days(IQR:2-5 days)vs.5 days(IQR:4-6 days);P<0.01].The negative conversion rate of nucleic acid in the treatment group were significantly higher than those in the control group at day 7(91.73%vs.86.90%,P=0.014).Compared with the control group,the hospitalized days in the treatment group were significantly reduced[10 days(IQR:8-11 days)vs.11 days(IQR:10.25-12 days);P<0.01].The time of the first nucleic acid negative conversion had significant differences between the treatment and control groups[3 days(IQR:2-4 days)vs.5 days(IQR:
