Taxol(Paclitaxel),an important anticancer drug,is derived at very low yields from Taxus(yew)species that grow very *** the present study,thirteen genes that encode enzymes involved in Taxol biosynthesis in Taxus *** analyzed with bioinformatics methods,and their expression levels in different tissues and after cold and hormone treatments were also *** results indicated that many cis-elements related to abiotic stresses and hormones were found in the promoter sequences of the 8 genes involved in Taxol ***,the 13 enzymes encoded by the target genes were located in different organelles and had many phosphorylation sites in the response *** 13 genes were expressed highly either in roots or in stems,with lower transcripts in needles,and they were highly expressed after treatment with cold,gibberellin,methyl jasmonate or coronatine,consistent with predictions based on the bioinformatics *** results suggest that the factors such as hormones and abiotic stresses stimulate taxane biosynthesis in yews,providing an important way to sustainably generate taxanes from yew trees or their cell cultures to improve Taxol yields.

Effect of degradation of PLGA and PLGA/β-TCP scaffolds on the growth of osteoblasts

Osteoblasts were cultured on porous scaffolds of poly(L-lactide-co-glycolide) (PLGA) and PLGA/β-tricalcium phosphate (β-TCP) to evaluate their *** proliferation of the cells on both scaffolds was examined before and after in vitro degradation for 4,8 and 12 weeks under static (shaking water bath) and dynamic (cyclic loading) *** indicate that porous PLGA and PLGA/β-TCP scaffolds have good biocompatibility and can be used as effective templates for guiding the growth of *** degradation of the scaffolds affects the proliferation of osteoblasts and the cell viability decreased with the degradation time.

Dynamical Simulation of Cornea Deformation in Laser Surgery

A dynamical simulation method is presented to model the cornea deformation in surgery of laser thermokeratoplasty. The virtual cornea is constructed as a mass-spring system. The corneal surface tension is simulated by damping spring stretch between mass points on the cornea model. The aqueous humor in the eyeball is modeled as ideal gas, and the intraocular pressure is simulated by gas pressure. The coagulation force is exerted on each photocoagulation spot to demonstrate its collapse caused by the condensation of corneal soft tissue irradiated by laser. An extra viscous drag force is added to each mass point to weaken the mass point oscillation. The use of the effective time-corrected Verlet integral method brings about flowing and stable dynamic simulation procedures. The simulation results show that, comparing to the undeformed model, the curvature of the region between the optical center and photocoagulation spot increases obviously. Moreover, the shape of the deformed virtual cornea is much similar to that of the real cornea after surgery.

OBJECTIVE: To investigate how compound Sophorae decoction(CSD) works on rats' models of ulcerative colitis(UC) induced by 2,4,6-trinitrobenzenesulfonic acid solution(TNBS) by metabolomics studies of colon, liver, and kidney tissue ***: Rats with UC induced by TNBS enema were used as models in this study. Metabolic profiles of the three tissues were analyzed and pathway analysis of biomarkers was performed after CSD administration and further integration of metabolic ***: Thirteen biomarkers were screened from colon, liver, and kidney tissue extracts, and the levels of these substances were up-or down-regulated in the model group, but their levels were reversed after CSD administration. These biomarkers were mainly related to Phenylalanine, tyrosine and tryptophan biosynthesis, Phenylalanine metabolism, Glutathione metabolism, Arachidonic acid metabolism, Nicotinate and nicotinamide metabolism,Alanine, aspartate and glutamate ***: CSD could significantly ameliorate the symptoms of UC by regulating multiple metabolic pathways.

Structural Characterization of Glucan Produced by Lactobacillus sake L-7 from Sausage

A linear glucan was produced by Lactobacillus sake L-7 isolated from homemade sausage. Cultivation of the strain in Man–Rogosa–Sharpe(MRS) medium containing 50 g/L sucrose yielded 5.3 g/L of purified exopolysaccharide(EPS). The EPS was characterized by gas chromatography(GC), Fourier-transform infrared(FT-IR) spectroscopy, high-performance sizeexclusion chromatography(HPSEC), nuclear magnetic resonance(NMR) spectroscopy, and scanning electron microscopy(SEM). The monosaccharide composition of the EPS was glucose, and its molecular weight was 1 × 10~7 Da. The FT-IR and NMR spectra revealed that the L-7 EPS was a linear glucan with α-(1 → 6) glucosidic bonds. SEM images of the dried EPS revealed a hollow tubular structure. The water solubility index and water holding capacity of L-7 EPS were 96 and 272%, respectively. The results of hydrolysis indicated that L-7 EPS was not susceptible to hydrolysis by physiological barriers and can be used as a soluble dietary fiber with health benefits. All these characteristics suggest that L-7 EPS might have potential applications in the food, cosmetic, and pharmaceutical industries.

Isolation, Purification, and Characterization of Exopolysaccharide Produced by Leuconostoc Citreum N21 from Dried Milk Cake

A strain with high production of exopolysaccharide(EPS) was isolated from dried milk cake(a traditional fermented food from Inner Mongolia). The strain was called N21 and later identified as Leuconostoc citreum( Leu. citreum). The strain was cultured in Man-Rogosa-Sharpe medium containing 50 g/L of sucrose for 48 h at 30 °C and the EPS purified, with a yield of 24.5 g/L. An average molecular weight of 6.07 × 10~6 g/mol was determined by high-performance size-exclusion chromatography. The structure of the purified EPS was investigated through gas chromatography, ~1H and ^(13)C nuclear magnetic resonance spectroscopy, and Fourier transform infrared spectroscopy. The results demonstrated a polysaccharide composed of D-glucopyranose units in a linear chain with consecutive α(1 → 6) linkages. No branching was found in the structure of the exopolysaccharide. The purified EPS showed high water solubility and emulsibility. Based on the thermogravimetric curve, the degradation temperature of the EPS was 308.47 °C, which suggested that the dextran in the study exhibited high thermal stability. The results indicated that Leu. citreum N21 could be widely used to produce linear EPS and that the EPS has potential applications in food science and processing as a food additive.

Tumor-targeting intravenous lipid emulsion of paclitaxel:Characteristics,stability,toxicity,and toxicokinetics

Lipid nanoemulsions are promising nanodrug delivery carriers that can improve the efficacy and safety of paclitaxel(PTX).However,no intravenous lipid emulsion of PTX has been approved for clinical treatment,and systemic safety profiles have not yet been *** we outline the development of a PTXloaded tumor-targeting intravenous lipid emulsion(PTX Emul)and describe its characteristics,colloidal stability,and systemic safety profiles in terms of acute toxicity,long-term toxicity,and *** also compare PTX Emul with conventional PTX *** showed that PTX Emul exhibited an ideal average particle size(approximately 160 nm)with narrow size distribution and robust colloidal stability under different *** reaction and hemolysis tests revealed that PTX Emul did not induce hypersensitivity reactions and had no hemolytic *** addition,where the alleviated systemic toxicity of PTX Emul may be attributed to the altered toxicokinetic characteristics in beagle dogs,including the decreased AUC and increased plasma clearance and volume of distribution,PTX Emul alleviated acute and long-term toxicity as evidenced by the enhanced the median lethal dose and approximate lethal dose,moderate body weight change,decreased bone marrow suppression and organ toxicity compared with those under PTX injection at the same dose.A fundamental understanding of the systemic safety profiles,high tumor-targeting efficiency,and superior antitumor activity in vivo of PTX Emul can provide powerful evidence of its therapeutic potential as a future treatment for breast cancer.

Efficient mucosal delivery remains a major challenge for the reason of the respiratory tract mucus act as a formidable barrier to nanocarriers by trapping and clearing foreign particulates. The surface property of nanoparticles determines their retention and penetration ability within the respiratory tract mucus. However, the interaction between nanoparticles and mucus, and how these interactions impact distribution has not been extensively investigated. In this study, polymeric nanoparticles loaded with a baicalein-phospholipid complex were modified with two kinds of polymers, mucoadhesive and mucus-penetrative polymer. Systematic investigations on the physicochemical property, mucus penetration, transepithelial transport, and tissue distribution were performed to evaluate the interaction of nanoparticles with the respiratory tract. Both nanoparticles had a similar particle size and good biocompatibility, exhibited a sustained-release profile, but showed a considerable difference in zeta potential. Interestingly, mucus-penetrative nanoparticles exhibited a higher diffusion rate in mucus, deeper penetration across the mucus layer, enhanced in vitro cellular uptake, increased drug distribution in airways, and superior local distribution and bioavailability as compared to mucoadhesive *** results indicate the potential of mucus-penetrative nanoparticles in design of a rational delivery system to improve the efficiency of inhaled therapy by promoting mucus penetration and increasing local distribution and bioavailability.
