Recent updates of precision therapy for gastric cancer: Towards optimal tailored management

Signaling pathways of gastric carcinogenesis and gastric cancer progression are being avidly studied to seek optimal treatment of gastric cancer. Among them, hepatocyte growth factor (HGF)/c-MET, phosphoinositide 3-kinase (PI3K)/Akt/mammalian target of rapamycin (mTOR) and janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) pathways have been widely investigated. Their aberrant expression or mutation has been significantly associated with advanced stage or poor prognosis of gastric cancer. Recently, aberrations of immune checkpoints including programmed cell death-1/programmed cell death ligand-1 (PD-1/PD-L1) have been suggested as an important step in the formation of a microenvironment favorable for gastric cancer. Accomplishments in basic research have led to the development of novel agents targeting these signaling pathways. However, phase III studies of selective anti-HGF/c-MET antibodies and mTOR inhibitor failed to show significant benefits in terms of overall survival and progression-free survival. Few agents directly targeting STAT3 have been developed. However, this target is still critical issue in terms of chemoresistance, and SH2-containing protein tyrosine phosphatase 1 might be a significant link to effectively inhibit STAT3 activity. Inhibition of PD-1/PD-L1 showed durable efficacy in phase I studies, and phase III evaluation is warranted. Therapeutic strategy to concurrently inhibit multiple tyrosine kinases is a reasonable option, however, lapatinib needs to be further evaluated to identify good responders. Regorafenib has shown promising effectiveness in prolonging progression-free survival in a phase II study. In this topic highlight, we review the biologic roles and outcomes of clinical studies targeting these signaling pathways.

采用厌氧-好氧移动床生物膜反应器串联处理城市垃圾渗滤液．探讨了各种操作条件对垃圾渗滤液生物降解效率的影响，并对其影响机理作了分析．结果表明，水力停留时间和有机容积负荷对系统的处理效率影响较大，当系统进水的COD容积负荷在4．01～7．87kg／（m3·d）范围内，系统COD平均总去除率为94．2％，其中厌氧反应器对COD的去除率占总去除率的87．95％～92．76％；当系统进水的容积负荷高达10．23～16．14kg／（m^3·d）时，系统总COD平均去除率仍高达92．64％，其中厌氧反应器对COD的去除率占总去除率的79．05％～86．56％．当好氧段HRT大于1．25d，系统对氨氮的总去除率始终在97％以上．当HRT=0．75d时，系统对氨氮的总去除率仅在20％左右．该系统具有很强的抗冲击负荷能力，即使在24h内受到超过正常运行负荷4倍的冲击时，系统经过约3d可恢复正常．

Differential Anticancer Effect of an Apple Extract (Applephenon<sup>&reg</sup>), Polyphenols and Isoflavones on Normal Human Keratinocytes and Epidermoid Cancer Cells

Applephenon®, a purified extract prepared from green apples, was examined for its cytotoxicity and inhibitory effects on the proliferation of cultures of normal human keratinocytes and several epidermoid cancer cell lines. Our HPLC studies demonstrated a high content of phenolic compounds (>65%), including catechin, epicatechin, caffeic acid and phloretin as well as polyphenols such as proanthocyanidins. Applephenon® demonstrated a greater cytotoxic effect against HeLa, A431 cancer cell lines and HaCaT, an immortalized keratinocyte cell line than serum-free cultures of proliferating normal human keratinocytes (NHK). Proliferation of NHK was inhibited at concentrations above 0.0013% while concentrations above 0.005% were cytotoxic. By contrast, Applephenon® solutions above 0.00025% killed each of the cancer cell lines. Treated cells displayed increased intercellular separation and evidence of keratinizing stratification. We also tested the effect of epicatechin, and two isoflavonoids, genistein and daidzein, on cancer cell lines. Hela cells were more sensitive to epicatechin and genistein inhibition of cell growth and cytotoxicity than were NHK. Daidzein at these concentrations had little effect on cancer cells. These results indicate that Applephenon® and some of its phenolic components have selective anticancer activity.

An Efficient Viewport-dependent 360 VR System Based on Adaptive Tiled Streaming

Recent advances in 360 video streaming technologies have enhanced the immersive experience of video streaming ***,there is immense potential for the application of 360 video encoding formats to achieve highly immersive virtual reality(VR)***,360 video streaming requires considerable bandwidth,and its performance depends on several ***,the optimization of 360 video bitstreams according to viewport texture is ***,we propose an adaptive solution for VR systems using viewport-dependent tiled 360 video *** increase the degrees of freedom of users,the moving picture experts group(MPEG)recently.defined three degrees plus of freedom(3DoF+)and six degrees of freedom(6DoF)to support free user movement within camera-captured *** proposed method supports 6DoF to allow users to move their heads ***,we propose viewport-dependent tiled 360 video streaming based on users’head *** proposed system generates an adaptive bitstream using tile sets that are selected according to a parameter set of user’s viewport *** extracted bitstream is then transmitted to the user’s *** decoding,the user’s viewport is generated and rendered on VR head-mounted display(HMD).Furthermore,we introduce certain approaches to reduce the motion-to-photon *** experimental results demonstrated that,in contrast with non-tiled streaming,the proposed method achieved high-performance 360 video streaming for VR systems,with a 25.89%BD-rate saving for y-PSNR and 61.16%for decoding time.

The Arabidopsis bZIP1 Transcription Factor Is Involved in Sugar Signaling, Protein Networking, and DNA Binding

Sugar signaling is a mechanism that plants use to integrate various internal and external cues to achieve nutrient homeostasis, mediate developmental programs, and articulate stress responses. Many bZlP transcription factors are known to be involved in nutrient and/or stress signaling. An Arabidopsis Sl-group bZlP gene, AtbZIP1, was identified as a sugar-sensitive gene in a previous gene expression profiling study （Plant Cell. 16, 2128-2150）. In this report, we show that the expression of AtbZIP1 is repressed by sugars in a fast, sensitive, and reversible way. The sugar repression of Atb- ZIP1 is affected by a conserved sugar signaling component, hexokinase. Besides being a sugar-regulated gene, AtbZIP1 can mediate sugar signaling and affect gene expression, plant growth, and development. When carbon nutrients are limited, gain or loss of function of AtbZlP1 causes changes in the rates of early seedling establishment. Results of phenotypic analyses indicate that AtbZlP1 acts as a negative regulator of early seedling growth. Using gain- and loss-of-function plants in a microarray analysis, two sets of putative AtbZIP1-regulated genes have been identified. Among them, sugar-responsive genes are highly over-represented, implicating a role of AtbZlP1 in sugar-mediated gene expression. Using yeast two-hybrid （y-2-H） screens and bimolecular fluorescence complementation （BiFC） analyses, we are able to recapitulate extensive C/S1 AtbZlP protein interacting network in living cells. Finally, we show that AtbZIP1 can bind ACGT-based motifs in vitro and that the binding characteristics appear to be affected by the heterodimerization between AtbZlP1 and the C-group AtbZIPs, including AtbZlP10 and AtbZlP63.

Artemisia capillaris Thunberg is a medicinal plant used as a traditional medicine in many cultures. It is an effective remedy for liver problems including hepatitis. Recent pharmacological reports have indicated that Artemisia species can exert various neurological effects. Previously, we reported a memory-enhancing effect of Artemisia species. However, the mechanisms underlying the neuroprotective effect of A. capillaris(AC) are still unknown. In the present study, we investigated the effect of an ethanol extract of AC on ischemic brain injury in a mouse model of transient forebrain ischemia. The mice were treated with AC for seven days, beginning one day before induction of transient forebrain ischemia. Behavioral deficits were investigated using the y-maze. Nissl and Fluoro-jade B staining were used to indicate the site of injury. To determine the underlying mechanisms for the drug, we measured acetylcholinesterase activity. AC(200 mg·kg-1) treatment reduced transient forebrain ischemia-induced neuronal cell death in the hippocampal CA1 region. The AC-treated group also showed significant amelioration in the spontaneous alternation of the y-maze test performance, compared to that in the untreated transient forebrain ischemia group. Moreover, AC treatment showed a concentration-dependent inhibitory effect on acetylcholinesterase activity in vitro. Finally, the effect of AC on forebrain ischemia was blocked by mecamylamine, a nonselective nicotinic acetylcholine receptor antagonist. Our results suggested that in a model of forebrain ischemia, AC protected against neuronal death through the activation of nicotinic acetylcholine receptors.

The Efficacy of Psychosocial Interventions for Adults in Contexts of Ongoing Man-Made Violence—A Systematic Review

Compared to psychosocial programs implemented in post-conflict settings those executed in areas of ongoing conflicts may have different effects. Their evidence of efficacy has never been systematically reviewed. We searched PubMed, PsychInfo and the Dutch Tropical Institute Literature Portal from inception to31 January 2013 to identify studies on community-oriented psychosocial and psychiatric/clinical services for adults during ongoing manmade conflict or its direct aftermath. Of 6358 articles screened, 16 met our inclusion criteria. The interventions varied from psycho-educational to psychotropic drugs. The review is presented using outcome indicators such as PTSD, anxiety, depression, physical health, functioning and well being. A substantial improvement of some outcome was found though the small number of studies and their heterogeneity did not justify strong conclusions. PTSD symptoms improved significantly by treatments that included exposure (such as narrative exposure therapy). A number of studies (eight) showed notable improvement of the client’s functioning through counseling interventions. depression and anxiety both improved considerably using that culturally adapted interventions (two studies), whereas non-culturalized interventions did not. We found a notable lack of studies on the efficacy of medication and on preferred western, evidence-based interventions for PTSD such Eye Movement desensitization. To measure outcomes only two studies applied locally.developed diagnostic labels and validated instruments. Future research should encourage the use of robust research methods that are culturally valid, including mixed-methods research to combine measurable outputs with qualitative research aimed at improved understanding from the client’s perspective.
