An anaerobic,Gram-stain-positive,spore-forming bacterium,designated DY30321 was isolated from a hydrothermal sulfides mixture sample collected during the cruise DY30 of R/V Da Yang Yi Hao.Cells of strain DY30321 were rod-shaped with rounded ends,and were not motile.Strain DY30321 grew optimally at pH 8.0,30 ℃,with salinity(sea salts) of 30-40 g.L.The principal fatty acids of strain DY30321 were C and summed Feature 1(comprising iso H-C/30H-C).The predominant polar lipids of strain DY30321 were diphosphatidylglycerol,phosphatidylcholine and phosphatidylethanolamine.No respiratory quinone was detected.The G+C content of the genomic DNA of strain DY30321 was 33.4 mol%.Phylogenetically,strain DY30321 branched within the family Peptostreptococcaceae,with(misclassified) Clostridium halophilum Ml being its closest phylogenetic relative(94.6%similarity),followed by(misclassified) C caminithermale DVird3(92.1%).They show very low16 S rRNA gene sequence similarity(< 84%) to the type strain of C butyricum ATCC 19398,the type species of the genus Clostridium sensu stricto.On the basis of its phenotypic,phylogenetic and chemotaxonomic characteristics,we propose strain DY30321(=KCTC 15549=MCCC1A01532) as a representative of a new genus and species in the family Peptostreptococcaceae,for which the name Wukongibacter baidiensis gen.nov.,sp.***.is proposed.We also propose the genus Maledivibacter gen.***.to accommodate Clostridium halophilum as Maledivibacter halophilus comb.nov.(type species of the genus),and the genus Paramaledivibacter gen.***.to accommodate Clostridium caminithermale as Paramaledivibacter caminithermalis comb.nov.(type species of the genus).

A bacterial strain,YYQ-30,isolated from a water sand sediment mixed sample collected from a terrestrial Spring located in Dunhuang,China,was characterized with respect to its morphology,physiology and taxonomy.The bacterium was found to be Gram stain-negative,aerobic,oxidase- and catalase-positive,non-flagellated,oval to rod-shaped(0.5-1 μm in width and 1.1-6.6 μm in length) and divided by binary fission.Growth was observed at 0-10.0%(w/v)NaCl with optimal growth at 0-3.0%.The pH and temperature ranges for growth are pH 6.0-9.0and 10-45℃(optimum growth at pH 7.0-8.5 and 30-37℃).The isolate could reduce nitrate to nitrite,hydrolyse aesculin and gelatine(weakly),but was unable to degrade Tween 80 or starch.Phylogenetic analysis based on 16 S rRNA gene sequences indicated that strain YYQ-30 belongs to the family Rhodobacteraceae and forms a distinct lineage with the type strain of Albimonas donghaensis and forms a branch within a cluster constituted by the type strains of the genera Albimonas,Rhodovulum,Albidovulum,Haematobacter,Tropicimonas;levels of 16 S rRNA gene sequence similarity between strain YYQ-30 and the members of related genera ranged from 94.1%to 89.7%.Strain YYQ-30 contains Q-10 as the predominant ubiquinone and summed feature 8(Cω7c/ω6c(70.0%),C(9.5%),summed feature 2(C 3OH/C iso Ⅰ and C aldehyde(6.9%),and Cω7c 11-methyl(6.0%) as the principal fatty acids.The polar lipids comprise diphosphatidylglycerol,phosphatidylethanolamine,phosphatidylglycerol,three unidentified phospholipids,two unidentifided aminolipids,and five unknown lipids.The pufLM gene was detected.The G+C content of the genomic DNA was determined to be 71.7 mol%.Based on the phylogenetic,chemotaxonomic and phenotypic data obtained in this study,strain YYQ-30 is considered to represent a novel species in a new genus within the family Rhodobacteraceae,for which the name Halovulum dunhuangense gen.***.sp.***.is proposed.The type strain of Halovulum dunhuangense is YYQ-30(= LMG 27418= MCCC 1A06483).

A novel strain 22II-Sll-z3 was isolated from the deep sea sediment of the Atlantic Ocean.The bacterium was aerobic,Gram-negative,oxidase positive and catalase negative,ovalto rod-shaped,and non-motile.Growth was observed at salinities of 1-9%and at temperatures of10-45℃.The isolate was capable of aesculin hydrolysis and Tween 20,40 and 80 degradation,but unable to reduce nitrate to nitrite or hydrolyze gelatin.Phylogenetic analysis based on 16 S rRNA gene sequences indicated that strain 22II-S11-z3 belonged to the genus Aestuariivita,with highest sequence similarity to Aestuariivita boseongensis BS-B2(97.47%).The ANI value and digital DNA-DNA hybridization estimate values between strain 22II-Sll-z3 and A.boseongensis BS-B2 are 71.49-71.50%and 20.00±2.31%,respectively.The G+C content of the chromosomal DNA was 65.5 mol%.The principal fatty acids(>5%) were Sum In Feature 8(Cω7c/ω6c)(35.24%),Ccycloω8c(20.85%),C(11.76%),Cω7c 11-methyl(11.42%)and C 3OH(9.44%).The respiratory quinone was determined to be Q-10.Diphosphatidylglycerol,phosphatidylcholine,phosphatidylglycerol,nine unidentified phospholipids,one unidentified aminolipid and two unidentified lipids were present.The combined genotypic and phenotypic data show that strain 22II-S11-z3 represents a novel species of the genus Aestuariivita,for which the name Aestuariivita atlantica sp.***.is proposed,with the type strain 22II-S11-z3(= KCTC 42276 = MCCC 1A09432).

A new aerobic,Gram stain-negative,oxidase- and catalase- positive,ovoid or rod-shaped bacterial strain,designated HTCJW17,was isolated from the sediment collected from the South China Sea and subjected to a polyphasic taxonomic characterization.The isolate forms small,creamy-white,opaque and circular colonies on agar plates.Growth occurs between 15 and45℃,1-7%(w/v) NaCl and pH 6-9.Phylogenetic analysis based on 16 S rRNA gene sequences showed that strain HTCJW17 belongs to the family Kordiimonadaceae of the order Kordiimonadales,with highest sequence similarity to Kordiimonas gwangyangensis GW14-5(91.1%).The major fatty acids were identified to be summed feature 8(Cω7c/ω6c;56.6%),C(8.5%),C2-OH(8.4%),summed feature 3(Cω7c/Cω6c;6.2%),and C(5.2%).The polar lipids were determined to be phosphatidylcholine,phosphatidylethanolamine,psphatidylglycerol,six unidentified glycolipids and two undentified phospholipids.The respiratory quinone was identified as ubiquinone Q-10.The genomic DNA G+C content was determined to be 56.3 mol%.On the basis of genotypic,chemotaxonomic and phenotypic distinctness,we propose a novel genus,Emcibacter gen.nov.,with Emcibacter nanhaiensis sp.***.as the type species.The type strain is HTCJW17(= CGMCC 1.12471= LMG 27419=MCCC 1A06723).

A taxonomic study was carried out on strain 2211-Sllg,which was isolated from the surface seawater of the Atlantic Ocean.The bacterium was found to be Gram-negative,rod shaped without flagellum,oxidase positive and weakly catalase positive.Growth was observed at NaCl concentrations of 0.5-9%and at temperatures of 10-41℃.The isolate was incapable of gelatin hydrolysis and unable to reduce nitrate to nitrite,degrade aesculin and Tween 80.On the basis of16 S rRNA gene sequence similarity,strain 2211-S11g was found to be most closely related to Oceanicola batsensis HTCC2597(97.26%),followed by Oceanicola nitratireducens JLT1210(96.39%),whilst other species of genus Oceanicola shared 94.00-96.34%sequence similarity.However,it showed low similarity to Oceanicola granulosus HTCC2516(94.79%),the type species of the genus Oceanicola.Phylogenetic analysis showed that strain 2211-S11 g formed a clade with six species currently classified in the genus Oceanicola,but strain O.granulosus HTCC2516 and strain O.litoreus M-M22 clustered with two other genera respectively.The ANI values between strain 2211-S11g and two type strains(O.batsensis HTCC2597 and O.granulosus HTCC2516) are 91.86%and 91.81%respectively.The digital DNA-DNA hybridization estimate values between strain 2211-S11g and two type strains(O.batsensis HTCC2597 and O.granulosus HTCC2516) are 23.4±2.4%and 20.0±2.3%,respectively.The principal fatty acids were identified as summed feature 8(Cω7c/ω6c),C,Cω7c11-methyl and C3OH.The G+C content determined from the draft genome sequence is 64.1mol%.The respiratory quinone was determined to be Q-10(100%).Phosphatidylethanolamine,phosphatidylglycerol,an aminolipid,phosphatidylcholine,a phospholipid and three lipids were identified in the polar lipids.The combined genotypic and phenotypic data also show that strain2211-S11g should not be assigned to the genus Oceanicola;consequently strain 2211-S11g is concluded to represent a novel species of a novel genus in the family Rhodobacteraceae,for which the name Pseudooceanicola atl

A taxonomic study was carried out on strain 22II14-10F7 T, which was isolated from the deep sea water of the Atlantic Ocean with oil-degrading enrichment. The bacterium was Gram-negative, oxidase- and catalase-positive, rod-shape. Growth was observed at salinities from 0.5 to 15 % and at temperatures from 4 to 37 °C, it was unable to hydrolyze Tween 40, 80 or gelatin. Phylogenetic analysis based on 16 S r RNA gene sequences indicated that strain 22II14-10F7 T belonged to the genus Zunongwangia, with highest sequence similarity of 97.3 % to Zunongwangia profunda SM-A87 T, while the others were all below 94.0 %. The DDH estimate value between strain 22II14-10F7 T and Z. profunda SM-A87 T was 27.20 %±2.43% according to their genome sequences. The principal fatty acids were iso-C15:0(22.42 %), C15:0 anteiso(8.90 %), iso-C15:1G(8.50 %), iso-C17:0 3OH(12.78 %), Sum In Feature 3(C16:1ω7c /ω6c)(8.09 %) and Sum In Feature 9(iso-C17:1ω9c or C16:0 10-methyl)(13.54 %). The G+C content of the chromosomal DNA was 35.5 mol%. The major respiratory quinone was determined to be MK-6. Phosphatidylethanolamine(PE), two aminolipids(AL1 and AL2) and five unknown lipids(L1-L5) were present. The combined genotypic and phenotypic data show that strain 22II14-10F7 T represents a novel species of the genus Zunongwangia, for which the name Zunongwangia atlantica sp. nov. is proposed, with the type strain 22II14-10F7T(=CGMCC1.12470 T =LMG 27421T=MCCC 1A06481T).

A taxonomic study was carried out on strain R12B,which was isolated from seawater of the Atlantic Ocean.The bacterium was Gram-negative,oxidase positive and catalase weakly positive,rod shaped.Growth was observed at salinities of 0.5-15%and at temperatures of 4-45℃.The isolate was incapable of gelatine,Tween 80,aesculin hydrolysis or nitrate reduction.Phylogenetic analysis based on 16 S rRNA gene sequences indicated that strain R12B belonged to the genus Roseovarius,with highest sequence similarity to Roseovarius indicus B108(97.14%),followed by Roseovarius halotolerans HJ50(96.78%),other species of genus Roseovarius shared94.02-96.14%sequence similarity.The DNA-DNA hybridization estimate values between strain R12B and Roseovarius indicus B108 was 23.2%.The ANI value between strain R12B and Roseovarius indicus B108 was 77.08%.The principal fatty acids were Sum In Feature 8(Cω7c/ω6c)(62.28%) and C(15.70%).The G+C content of the chromosomal DNA was63.6 mol%.The respiratory quinone was determined to be Q-10.Diphosphatidylglycerol(DPG),phosphatidylglycerol(PG),phosphatidylcholine(PC),phosphatidylethanolamine(PE),two aminolipids(ALs),two phospholipids(PLs) were present.The combined genotypic and phenotypic data show that strain R12B represents a novel species within the genus Roseovarius,for which the name Roseovarius atlanticus sp.***.is proposed,with the type strain R12B(=MCCC 1A09786= KCTC 42506).

【目的】为了从南沙海域中分离获得微生物菌种资源,【方法】本文通过沉积物采样、可培养菌分离及16S rRNA鉴定,【结果】从22个站点的沉积物样品中获得349株细菌,分属于87个种。发现产芽孢细菌分布最广,并在10个站点的分离株中占多数;它们是Bacillus,Halobacillus,Brevibacillus,Paenibacillus,Pontibacillus和Thalassobacillus。其中芽孢杆菌(Bacillus)无论在数量上还是种类上都最多,分别属于34种,其中有8个可能的新种。此外,γ-Proteobacteria是分离率较高的另一亚群;其中,假单胞菌(Pseudomonas),海杆菌(Marinobacter),食烷菌(Alcanivorax)属的细菌最多。统计还发现,在深度750～2000 m之间,低GC含量的细菌最丰富,而深度2000 m以下,分离株则全部为γ-Proteobacteria。【结论】南沙沉积物可培养微生物中产芽孢细菌及γ-Proteobacteria比较丰富;其中,产芽孢细菌的多样性最高,具有进一步研究开发价值。

为研究污水厂/养殖池中好氧反硝化微生物的多样性及菌株反硝化能力,本研究采集了位于福建省厦门市和漳州市的污水处理厂、排污口、污水池、对虾养殖池的污水和污泥样品进行好氧反硝化微生物的富集、分离、鉴定和功能筛选。分别以NaNO3、... 详细信息

为研究污水厂/养殖池中好氧反硝化微生物的多样性及菌株反硝化能力,本研究采集了位于福建省厦门市和漳州市的污水处理厂、排污口、污水池、对虾养殖池的污水和污泥样品进行好氧反硝化微生物的富集、分离、鉴定和功能筛选。分别以NaNO3、NaNO2作为唯一氮源共分离纯化获得128株单菌。其中以NaNO3为唯一氮源分离得到63株,以NaNO2为唯一氮源分离得到65株。16SrRNA基因序列分析表明,128株单菌分属于γ-变形菌纲(Gammaproteobacteria,58.6%)、芽胞杆菌纲(Bacilli,6.4%)、放线菌纲(Actinobacteria,11.7%)、α-变形菌纲(Alphaproteobacteria,8.6%)、纤维菌纲(Cytophagia,2.3%)、鞘脂杆菌纲(Sphingobacteria,0.8%)和黄杆菌纲(Flavobacteria,1.6%)7个纲中的38个属。其中盐单胞菌属(Halomonas,29.7%)和芽胞杆菌属(Bacillus,12.5%)为优势菌属,并且广泛存在于各个样品中。反硝化功能初筛结果表明,35株菌能在72h内将20mg·L-1 NO-3-N/NO-2-N完全去除;复筛结果表明,21株菌能在72h内将100 mg·L-1 NO-3-N/NO-2-N完全去除,并且盐单胞菌属、卓贝尔氏菌属(Zobellella)、斯塔普氏菌属(Stappia)及节杆菌属(Arthrobactor)反硝化效果较好,其中斯塔普氏属是首次报道具有好氧反硝化功能。本研究结果表明,污水场/养殖池等环境中可培养反硝化细菌多样性丰富,同时高效反硝化菌的获得也为含氮废水的生物处理提供了良好的菌种资源。
