Reduction of chlorogermylene MsFluindtBu-GeCl 1 with potassium graphite(KC8)afforded putative germylyne radical MsFluindtBu-Ge 2 as confirmed by electron paramagnetic resonance(EPR)spectroscopy.However,it slowly decayed via C—H bond activation at the fluorenyl moiety to yield a bis(germylene)3 at room temperature.By using a Lewis base to stabilize the unoccupied p orbital at the GeⅠradical center,acyclic two-coordinate GeⅠradicals MsFluindtBu-Ge(IMe4)4(IMe4=1,3,4,5-tetramethyl-imidazolin-2-ylidene),MsFluindtBu-Ge(IiPr)5(IiPr=1,3-diisopropyl-4,5-dimethyl-imidazolin-2-ylidene),MsFluindtBu-Ge(PMe3)6 were isolated in crystalline forms.The unpaired electron in 4-6 mainly resides at the Ge 4p orbital as revealed by EPR spectroscopic studies and theoretical calculations.Interestingly,facile ligand exchange of PMe3 in 6 with IMe4 and IiPr was observed to afford 4 and 5,respectively.Moreover,phosphinogermylyne 6 reacted with PhEEPh(E=S,Se),4-tetrabutylphenylacetylene(Ar'CCH),[CpMo(CO)3]2 and nBu3SnH to furnish germylenes MsFluindtBu-GeEPh(E=S 7,Se 8),MsFluindtBu-GeCH=CHAr'9,a germylyne complex MsFluindtBu-Ge≡Mo(CO)2Cp 10 and a Ge(Ⅳ)compound MsFluindtBu-GeH2SnnBu311,respectively.The reactivity studies demonstrate that 6 can act as a synthon of one-coordinate germylyne radical attributing to labile coordination of trimethylphosphine.

Acute ischemic stroke is a clinical emergency and a condition with high morbidity,mortality,and disability.Accurate predictive,diagnostic,and prognostic biomarkers and effective therapeutic targets for acute ischemic stroke remain undetermined.With innovations in high-throughput gene sequencing analysis,many aberrantly expressed non-coding RNAs(ncRNAs)in the brain and peripheral blood after acute ischemic stroke have been found in clinical samples and experimental models.Differentially expressed ncRNAs in the post-stroke brain were demonstrated to play vital roles in pathological processes,leading to neuroprotection or deterioration,thus ncRNAs can serve as therapeutic targets in acute ischemic stroke.Moreover,distinctly expressed ncRNAs in the peripheral blood can be used as biomarkers for acute ischemic stroke prediction,diagnosis,and prognosis.In particular,ncRNAs in peripheral immune cells were recently shown to be involved in the peripheral and brain immune response after acute ischemic stroke.In this review,we consolidate the latest progress of research into the roles of ncRNAs(microRNAs,long ncRNAs,and circular RNAs)in the pathological processes of acute ischemic stroke–induced brain damage,as well as the potential of these ncRNAs to act as biomarkers for acute ischemic stroke prediction,diagnosis,and prognosis.Findings from this review will provide novel ideas for the clinical application of ncRNAs in acute ischemic stroke.

Long-term nonunion of bone defects has always been a major problem in orthopedic treatment.Artificial bone graft materials such as Poly(lactic-co-glycolic acid)/β-tricalcium phosphate(PLGA/β-TCP)scaffolds are expected to solve this problem due to their suitable degradation rate and good osteoconductivity.However,insufficient mechanical properties,lack of osteoinductivity and infections after implanted limit its large-scale clinical application.Hence,we proposed a novel bone repair bioscaffold by adding zinc submicron particles to PLGA/β-TCP using low temperature rapid prototyping 3D printing technology.We first screened the scaffolds with 1 wt%Zn that had good biocompatibility and could stably release a safe dose of zinc ions within 16 weeks to ensure long-term non-toxicity.As designed,the scaffold had a multi-level porous structure of biomimetic cancellous bone,and the Young’s modulus(63.41±1.89 MPa)and compressive strength(2.887±0.025 MPa)of the scaffold were close to those of cancellous bone.In addition,after a series of in vitro and in vivo experiments,the scaffolds proved to have no adverse effects on the viability of BMSCs and promoted their adhesion and osteogenic differentiation,as well as exhibiting higher osteogenic and anti-inflammatory properties than PLGA/β-TCP scaffold without zinc particles.We also found that this osteogenic and anti-inflammatory effect might be related to Wnt/β-catenin,P38 MAPK and NFkB pathways.This study lay a foundation for the follow-up study of bone regeneration mechanism of Zn-containing biomaterials.We envision that this scaffold may become a new strategy for clinical treatment of bone defects.

The repair of osteochondral defects is one of the major clinical challenges in orthopaedics.Well-established osteochondral tissue engineering methods have shown promising results for the early treatment of small defects.However,less success has been achieved for the regeneration of large defects,which is mainly due to the mechanical environment of the joint and the heterogeneous nature of the tissue.In this study,we developed a multi-layered osteochondral scaffold to match the heterogeneous nature of osteochondral tissue by harnessing additive manufacturing technologies and combining the established art laser sintering and material extrusion techniques.The developed scaffold is based on a titanium and polylactic acid matrix-reinforced collagen“sandwich”composite system.The microstructure and mechanical properties of the scaffold were examined,and its safety and efficacy in the repair of large osteochondral defects were tested in an ovine condyle model.The 12-week in vivo evaluation period revealed extensive and significantly higher bone in-growth in the multi-layered scaffold compared with the collagen–HAp scaffold,and the achieved stable mechanical fixation provided strong support to the healing of the overlying cartilage,as demonstrated by hyaline-like cartilage formation.The histological examination showed that the regenerated cartilage in the multi-layer scaffold group was superior to that formed in the control group.Chondrogenic genes such as aggrecan and collagen-II were upregulated in the scaffold and were higher than those in the control group.The findings showed the safety and efficacy of the cell-free“translation-ready”osteochondral scaffold,which has the potential to be used in a one-step surgical procedure for the treatment of large osteochondral defects.

The disruption and reconstruction of the TREM2^(+) tissue resident macrophage(TRM)barrier on the surface of synovial lining play a key role in the activation and"remission"of rheumatoid arthritis(RA),which engender the prediction of this immunologic barrier as a potential driver for the achievement of"cure"in RA.However,strategies to promote the reconstruction of this barrier have not been reported,and the effect of patching this barrier remains unidentified.On the other hand,appropriate piezoelectric stimulation can reprogram macrophages,which has never been exerted on this barrier TRM yet.Herein,we design piezoelectric tetragonal BaTiO_(3)(BTO)ultrasound-driven nanorobots(USNRs)by the solvothermal synthesis method,which demonstrates satisfactory electro-mechanical conversion effects,paving the way to generate controllable electrical stimulation under ultrasound to reprogram the barrier TRM by minimally invasive injection into joint cavity.It is demonstrated that the immunologic barrier could be patched by this USNR effectively,thereby eliminating the hyperplasia of vessels and nerves(HVN)and synovitis.Additionally,TREM2 deficiency serum-transfected arthritis(STA)mice models are applied and proved the indispensable role of TREM2 in RA curing mediated by USNR.In all,our work is an interesting and important exploration to expand the classical tetragonal BTO nanoparticles in the treatment of autoimmune diseases,providing a new idea and direction for the biomedical application of piezoelectric ceramics.

Racial Differences in Left Ventricular Filling Pressure Following Acute Aerobic Exercise Between Chinese and Caucasians

Background Left ventricular filling pressure(LVFP)is an important early indicator of heart failure that is more prevalent in Caucasians than Chinese.Exercise-induced change in LVFP may provide more incremental information to assess dias-tolic function.But it was unknown whether there was difference in LVFP following acute exercise between Caucasians and Chinese.Purpose The purpose of this study was to investigate the change of LVFP following an acute 45-min aerobic exercise in healthy Caucasian and Chinese individuals.Methods Sixty participants(30 Caucasians and 30 Chinese,half was male,respectively)performed an acute bout of aero-bic exercise at 70%of heart rate reserve.Hemodynamics,Left ventricle(LV)morphology and function parameters were measured at baseline,then at 30-min and 60-min post-exercise.Results There was a similar LV ejection fraction,LV fraction shorten,lateral E/eʹand lateral eʹbetween Chinese and Caucasians at baseline.There was a significant race-by-time interaction in lateral E/eʹand lateral eʹbetween Chinese and Caucasians from pre-exercise to 30 min and 60 min after acute aerobic exercise.TheΔE/eʹwas significant correlated with baseline systolic blood pressure.Conclusion The change of LVFP was different between Chinese and Caucasians following acute aerobic exercise.The racial differences may be primarily caused by the changes of LV relaxation following exercise,baseline systolic blood pressure may also contribute to the differences.

Betaine-homocysteine methyltransferase(BHMT) regulates protein methylation and is correlated with tumorigenesis; however, the effects and regulation of BHMT in hepatocarcinogenesis remain largely unexplored. Here, we determined the clinical significance of BHMT in the occurrence and progression of hepatocellular carcinoma(HCC) using tissue samples from 198 patients. BHMT was to be frequently found(86.6%) expressed at relatively low levels in HCC tissues and was positively correlated with the overall survival of patients with HCC. Bhmt overexpression effectively suppressed several malignant phenotypes in hepatoma cells in vitro and in vivo, whereas complete knockout of Bhmt(Bhmt-/-) produced the opposite effect. We combined proteomics, metabolomics, and molecular biological strategies and detected that Bhmt-/-promoted hepatocarcinogenesis and tumor progression by enhancing the activity of glucose-6-phosphate dehydrogenase(G6PD)and PPP metabolism in DEN-induced HCC mouse and subcutaneous tumor-bearing models. In contrast, restoration of Bhmt with an AAV8-Bhmt injection or pharmacological inhibition of G6PD attenuated hepatocarcinogenesis. Additionally, coimmunoprecipitation identified monomethylated modifications of the G6PD, and BHMT regulated the methylation of G6PD. Protein sequence analysis, generation and application of specific antibodies, and site-directed mutagenesis indicated G6PD methylation at the arginine residue 246. Furthermore, we established bidirectionally regulated BHMT cellular models combined with methylation-deficient G6PD mutants to demonstrate that BHMT potentiated arginine methylation of G6PD, thereby inhibiting G6PD activity, which in turn suppressed hepatocarcinogenesis. Taken together, this study reveals a new methylation-regulatory mechanism in hepatocarcinogenesis owing to BHMT deficiency, suggesting a potential therapeutic strategy for HCC treatment.

Purpose–The smoothness of the high-speed railway(HSR)on the bridge may exceed the allowable standard when an earthquake causes vibrations for HSR bridges,which may threaten the safety of running trains.Indeed,few studies have evaluated the exceeding probability of rail displacement exceeding the allowable standard.The purposes of this article are to provide a method for investigating the exceeding probability of the rail displacement of HSRs under seismic excitation and to calculate the exceeding probability.Design/methodology/approach–In order to investigate the exceeding probability of the rail displacement under different seismic excitations,the workflow of analyzing the smoothness of the rail based on incremental dynamic analysis(IDA)is proposed,and the intensity measure and limit state for the exceeding probability analysis of HSRs are defined.Then a finite element model(FEM)of an assumed HSR track-bridge system is constructed,which comprises a five-span simply-supported girder bridge supporting a finite length CRTS II ballastless track.Under different seismic excitations,the seismic displacement response of the rail is calculated;the character of the rail displacement is analyzed;and the exceeding probability of the rail vertical displacement exceeding the allowable standard(2mm)is investigated.Findings–The results show that:(1)The bridge-abutment joint position may form a step-like under seismic excitation,threatening the running safety of high-speed trains under seismic excitations,and the rail displacements at mid-span positions are bigger than that at other positions on the bridge.(2)The exceeding probability of rail displacement is up to about 44%when PGA 50.01g,which is the level-five risk probability and can be described as’very likely to happen’.(3)The exceeding probability of the rail at the mid-span positions is bigger than that above other positions of the bridge,and the mid-span positions of the track-bridge system above the bridge may be the most hazardous area for the running safety of trains

In tissue engineering field,it is important to develop a suitable numerical model to evaluate scaffold geometry design.The experimental evaluation of the effect of each specific scaffold parameter on tissue regeneration requires large cost and long time expend.Dynamic cell culture is commonly used for generating tissues which could replace damaged tissues.A perfusion bioreactor model is developed which is able to simulate dynamic cell culture,to evaluate scaffold quality.The wall-film model is used to simulate cell attachment with the assumption that cells could be seen as liquid drops.In the process of cell attachment,the cells could impinge to a solid surface and form a liquid film which were considered as cell attached on the scaffold surface.Two types of cell-scaffold interactions were involved in numerical models including trap model and Stanton-Rutland(Cell impinge model—CIM)model.For trap model,all cells impinged the scaffold are seen as attached.For Stanton-Rutland model,four regimes of cell-scaffold interaction are involved in the cell attachment,including stick,rebound,spread,and splash,and only stick and spread are seen as attached.By comparison with two different numerical methods,the results showed that CIM model result is more related to the experimental results than trap model,which indicated that four regimes of cell-scaffold interaction occurred in cell attachment process.By evaluating two different geometry scaffold's cells seeding by these two models,the results further indicated that this model are able to use for assessing the scaffold design.
