Recent Advances and Progress for the Fabrication and Surface Modification of AIE-active Organic-inorganic Luminescent Composites

Organic dyes based hybrid organic-inorganic luminescent nanomaterials with high quantum efficiency, good physical or chemical stability, and favorable biocompatibility, have attracted growing attention recently because of their important applications in the areas of biomedical imaging, chemical sensors, and light-emitting diodes(LEDs). Nevertheless, conventional fluorescence molecules suffer from aggregation-caused quenching(ACQ) when they are doped into inorganic nanomaterials. Aggregation-induced emission(AIE) is an abnormal and intriguing fluorescent phenomenon that has aroused increasing interest for various applications especially in biomedical fields. Compared with conventional organic dyes, the AIE-active molecules will emit more intense fluorescence in their aggregates or solid states. It provides an elegant route to overcome the drawbacks of conventional organic molecules. Over the past few decades, the fabrication and surface modification of various organic-inorganic luminescent composites doped with AIE-active molecules have been reported. Therefore, it is highly desirable to summarize these advances. In this review, recent advances and progress in constructing various AIEgens-doped organic-inorganic hybrid nanocomposites and their subsequent surface modification were summarized. We hope this review could further promote the research of AIE-active functional materials.

Preparation of ^(99m)Tc-HYNIC-Annexin V

99mTc-HYNIC-Annexin V was prepared for apoptosis radionuclide imaging in vivo. The expression plas-mid of human Annexin V was constructed and expressed in E. coli.. HYNIC was prepared and used as bifunctional chelating agent for 99mTc labeling. The radiochemical purity, radiolabeling yield and stability of 99mTc-HYNIC-Annexin V were studied. The coding sequence of human Annexin V gene was successfully cloned into the expression vector, and highly expressed in E. coli. A protein with a molecular weight of about 36kD could be in-duced and confirmed by Western Blotting and SDS-PAGE. Annexin V could be labeled with 99mTc using HYNIC,and showed high radiochemical purity of more than 90%. The radiolabeling yield could reach more than 90%. The reac-tion condition was moderate. It suggested that 99mTc-HYNIC-Annexin V may be a promising agent for apoptosis im-aging and clinical application.

Facile Fabrication of Aggregation-induced Emission Based Red Fluorescent Organic Nanoparticles for Cell Imaging

A cyano-substituted diarylethlene derivative aggregation-induced emission （ALE） dye with two amino end-groups and 4,4＇-（hexafluoroisopropylidene）diphthalic anhydride were facilely incorporated into red fluorescent organic nanoparticles （FONs） via room temperature anhydride ring-opening polymerization under an air atmosphere. These obtained RO-HFDA FONs were characterized by a series of techniques including gel permeation chromatography, Fourier transform infrared spectroscopy, size distribution and zeta potential measurements, UV-Vis absorption spectrum, fluorescent spectroscopy and transmission electron microscopy. Biocompatibility evaluation and cell uptake behavior of RO-HFDA FONs were further investigated to explore their potential biomedical application. We demonstrated that such FONs showed high water dispersibility, stable uniform spherical morphology （150-200 nm）, broad excitation band （350-605 nm）, intense red fluorescence （627 nm） and excellent biocompatibility, making them promising for cell imaging applications.

Tetraphenylethene End-capped Polyethylenimine Fluorescent Nanoparticles for Cell Imaging

Water soluble tetraphenylethene-based（TPE） aggregation-induced emission fluorescent organic nanoparticles（FONs） were facilely prepared via Schiff base condensation with polyethylenimine（PEI） and subsequent reduction. The obtained TPE-PEI FONs were characterized by a series of techniques including 1H-NMR, 13C-NMR, gel permeation chromatography, UV absorption spectra, fluorescence spectra, Fourier transform infrared spectroscopy, size distribution and zeta potential measurement, and transmission electron microscopy. Biocompatibility evaluation and cell imaging of TPE-PEI FONs were further explored. We demonstrated that such FONs showed intense fluorescence, spherical morphology and excellent biocompatibility, making them very suitable for cell imaging application.

Forkhead box protein 3 mRNA expression in the peripheral blood of kidney-transplant recipients with acute rejection

Background Regulatory T cells （Tregs） are immunologically and clinically interesting not least because of the important role they play in allograft rejection. Likewise, expression of the transcription factor forkhead box protein 3 （FOXP3）, detected in transplant biopsies, is also of intere.（;t because of its role in the development of regulatory T cells. In this study, we investigated the relationship between FoxP3 mRNA expression and acute organ rejection in kidney-transplant recipients. Methods In this prospective study, FoxP3 mRNA expression levels in peripheral blood samples from 10 recipients of living relative-donor kidney transplants were measured before transplantation as well as at the 14th and 90th days post-transplantation. In addition, 46 first-time kidney-transplant recipients participated in a cross-sectional study, with 28 patients classified as having acute organ rejection; whilst the remaining 18 patients had functionally stable allografts. FoxP3 mRNA expression levels in peripheral blood samples were compared between these two different groups. Results Before transplantation mean FoxP3 mRNA levels vs. GADPH mRNA levels （Ig（FoxP3 mRNA/GADPH mRNA）） in the 10 recipients were 1.11±0.67. The mean FoxP3 mRNA expression levels measured at 14th and 90th days post-transplantation were significantly higher than before transplantation （1.69±0.38, P=0.03; 1.44±0.21, P=0.04, respectively）. Additionally, the mean FoxP3 mRNA levels vs. GADPH mRNA expression levels （Ig（FoxP3 mRNA/GADPH mRNA）） were significantly higher in recipients suffering acute rejection compared with those with stable allografts （1.77±0.61 and 1.43±0.27, respectively, P=0.03）. Conclusions After kidney transplantntion, FoxP3 mRNA levels were found to increase in the peripheral blood of all recipients. Considerably higher FoxP3 rnRNA levels were observed in recipients suffering acute rejection. These results suggest that FoxP3 mRNA levels in peripheral blood samples can be used as a diagnostic t

Microsatellite instability in gastric cancer and pre-cancerous lesions

AIM: To investigate the microsatellite instability (MSI) in cancer and pre-cancerous lesions of the stomach and its mechanisms underlying the development of gastric ***: Thirty-six gastric cancer samples were obtained from patients undergoing surgery. Forty-one gastric mucosa samples with dysplasia and 51 with intestinal metaplasia (IM) were obtained from patients with chronic gastritis undergoing gastro-endoscopy. Genomic DNA was extracted from the samples. Silver staining single strand conformation polymorphis-polymerize chain reaction (SSCP-PCR) was used to screen MSI markers at 5 loci (Bat-25, Bat-26, D5S346, D17S250, and D2S123)in fresh tissues and formalin-fixed, paraffin-embedded samples and their corresponding normal gastric ***: The abnormal shifting of the single-strand DNA (MSI) was identified in 21 out of 36 (58.3%) gastric *** cases showed high-level MSI (two or more loci altered) and 14 showed low-level MSI (one locus altered).Gastric cancer with MSI had a tendency to be located in the distal stomach. MSI was also detected in 11 out of 41(26.8%) dysplasia samples and in 9 of 51 (17.6%) IM samples respectively. Three cases of dysplasia and one case of IM showed high-level MSI. Eight cases of dysplasia and 8 cases of IM displayed low-level MSI. MIS in IM was found only in moderate or severe-grade IM. No association was detected between MSI and dysplasia ***: Accumulation of MSI in dysplasia and intestinal metaplasia of gastric mucosa may be an early molecular event during gastric carcinogenesis and may contribute to the acquisition of transformed cell phenotype and the development of gastric cancer.

Rapamycin instead of mycophenolate mofetil or azathioprine in treatment of post-renal transplantation urothelial carcinoma

Background Malignant tumor is the most common complication occurred in transplant recipients. It is widely recognized that immunosuppressive treatments increase the risk of cancer in transplant recipients. The efficacy and safety of rapamycin （RPM） in combination with low-dose calcineurin inhibitor （CNI） in treating 15 renal allograft recipients which developed urothelial carcinoma were observed.Methods Immunosuppressive regimen in all recipients was altered with rapamycin to replace mycophenolate mofetil （MMF） or azathioprine （Aza）. The initial loading dosage was 2 mg/d, and the next dosage was 1 mg/d. The dosage of rapamycin was carefully adjusted according to the blood drug level and concentration of the drug was maintained at 4-6 μg/L. In all the 15 patients, the calcineurin inhibitor was reduced down to one third of the original dosage after the rapamycin blood concentration became stable. Surgical treatment and intravesical instillation chemotherapy were carried out in all patients. Recurrence of the tumor was monitored throughout the study. Post-transplant renal function and side effects were also closely monitored.Results Among the 15 patients, 9 had no tumor recurrence in 2 years, 2 had tumor recurrences twice, and 4 had once. There was no acute rejection observed during RPM treatment. Post-transplant renal function in 11 patients was improved with a decreased creatinine level. Hyperlipoidemia and thrombocytopenia were the most frequent adverse events which responded well to corresponding treatments.Conclusion Among the renal allograft recipients with urothelial carcinoma, combination of rapamycin and low dose calcineurin inhibitor treatment is effective and safe.

Effect of laser shock peening on combined low- and high-cycle fatigue life of casting and forging turbine blades

Laser shock peening （LSP） is a novel effective surface treatment method to improve the fatigue performance of turbine blades. To study the effect of LSP on combined low- and high-cycle fatigue （CCF） life of turbine blades, the CCF tests were conducted at elevated temperatures on two types of full-scale turbine blades, which were made of K403 by casting and GH4133B by forging. Probabilistic analysis was conducted to find out the effect of LSP on fatigue life of those two kinds of blades. The results indicated that LSP extended the CCF life of both casting blades and forging blades obviously, and the effect of LSP on casting blades was more evident; besides, a threshold vibration stress existed for both casting blades and forging blades, and the CCF life tended to be extended by LSP only when the vibration stress was below the threshold vibra- tion stress. Further study of fractography was also conducted, indicating that due to the presence of compressive residual stress and refined grains induced by LSP, the crack initiation sources in LSP blades were obviously less, and the life of LSP blades was also longer; since the compressive residual stress was released by plastic deformation, LSP had no effect or adverse effect on CCF life of blade when the vibration stress of blade was above the threshold vibration stress.

Friction stir welding(FSW)was used to prepare Q235 low-carbon steel joint,and the microstructure of different zones of the joint was *** electrochemical corrosion behavior of different macroscopic zones of the joint was evaluated in 3.5 wt.%NaCl *** results showed that the retreated-side heat-affected zone(HAZRS)and the advanced-side heat-affected zone(HAZAS)did not undergo phase transformation during FSW,and their microstructures were similar to those of the base material(BM),which was mainly composed of blocky ferrite and *** retreated-side thermo-mechanical affected zone(TMAZRS),the stirring zone(SZ),and the advanced-side thermo-mechanical affected zone(TMAZAS)underwent phase transformation,and the microstructure was mainly composed of proeutectoid ferrite and *** order of the corrosion resistance of different micro-zones from high to low was:HAZRS>BM>HAZAS>TMAZRS>SZ>*** corrosion mechanism for BM,HAZRS,and HAZAS was mainly the dissolution of *** contrast,the corrosion mechanism for TMAZRS,SZ,and TMAZAS was mainly galvanic corrosion between proeutectoid ferrite and pearlite.

Factors affecting the long-term renal allograft survival

Background In the past decades, the one-year graft survival of cadaveric renal allografts has been markedly improved,but their long-term survival has not kept pace. The attrition rate of renal allografts surviving after one year remains almost unchanged. The causes for late graft loss are multiple. The aim of this study was to analyze the predictive factors that impact long-term survival of grafts after kidney *** We retrospectively analyzed 524 kidney transplantation patients who were treated in our hospital between January 1991 and January 2000, including 254 patients who had lived more than 10 years with normal graft function （long survival group）, and 270 cases whose renal graft had survived less than 10 years （control group）. Specifically, we analyzed 10 factors that may potentially affect graft survival by both univariate and Logistic model multivariate analyses to pinpoint the independent risk *** Univariate analyses showed that no significant differences existed in the age or gender of recipients, dialysis time, lymphotoxin levels, or cold ischemia time between the two groups. However, the ratio of delayed graft function and acute rejection, and the uric acid levels of patients in the long survival group were significantly lower than those in the control group （P ＜0.01）. Furthermore, we found that the concentration of cyclosporin A at one year after transplantation and the histocompatibility antigen match of donor-recipients for patients within the long survival group were significantly higher than those in the control group （P ＜0.01 ）. Furthermore, multivariate analyses showed that these four factors were independent risk factors that impact patient *** The ratios of delayed graft function and acute rejection, the concentration of cyclosporin A at one year after transplantation, and serum uric acid levels are very important factors that affect the long-term survival of renal grafts.
