Causal relationship between circulating vitamin C and 25-hydroxyvitamin D concentrations and common mental disorders-a Mendelian randomization study

Mental disorders seriously affect people’s health and social stability.This Mendelian randomization(MR)study was designed to investigate the causal relationship between circulating vitamin C(VC)or 25-hydroxyvitamin D(25(OH)D)levels and mental disorders.The data used for the MR analysis were derived from the summary genome-wide association studies(GWAS)database for VC and 25(OH)D and from the Finn Gen consortium for fourteen mental disorders.Based on the inverse variance weighted(IVW)method,we found a potential causal association between circulating VC and anxiety disorders(IVW:OR=1.139,95%CI:1.023-1.269,P=0.018).However,no causal association was found between VC or 25(OH)D and other mental disorders(P>0.05).In the reverse MR analysis,individuals with Alzheimer’s disease was causally associated with higher concentrations of circulating VC(P=0.012),while individuals with anxiety disorders had a negative association between the concentrations of 25(OH)D(P=0.012).However,the current evidence does not support a causal relationship between VC or 25(OH)D and other mental disorders.In addition,there was no causal association between circulating VC and 25(OH)D(P>0.05).Future studies are needed to confirm these findings and to elucidate the mechanisms of potential causality.

Cellular metabolism is the fundamental process by which cells maintain growth and self-renewal.It produces energy,furnishes raw materials,and intermedi-ates for biomolecule synthesis,and modulates enzyme activity to sustain normal cellular functions.Cellular metabolism is the foundation of cellular life processes and plays a regulatory role in various biological functions,including pro-grammed cell death.Ferroptosis is a recently discovered form of iron-dependent programmed cell death.The inhibition of ferroptosis plays a crucial role in tumorigenesis and tumor progression.However,the role of cellular metabolism,particularly glucose and amino acid metabolism,in cancer ferroptosis is not well understood.Here,we reviewed glucose,lipid,amino acid,iron and sele-nium metabolism involvement in cancer cell ferroptosis to elucidate the impact of different metabolic pathways on this process.Additionally,we provided a detailed overview of agents used to induce cancer ferroptosis.We explained that the metabolism of tumor cells plays a crucial role in maintaining intracellu-lar redox homeostasis and that disrupting the normal metabolic processes in these cells renders them more susceptible to iron-induced cell death,resulting in enhanced tumor cell killing.The combination of ferroptosis inducers and cel-lular metabolism inhibitors may be a novel approach to future cancer therapy and an important strategy to advance the development of treatments.

Combining prognostic value of serum carbohydrate antigen 19-9 and tumor size reduction ratio in pancreatic ductal adenocarcinoma

BACKGROUND Pancreatic ductal adenocarcinoma(PDAC)is a common cancer with increasing morbidity and mortality due to changes of social environment.AIM To evaluate the significance of serum carbohydrate antigen 19-9(CA19-9)and tumor size changes pre-and post-neoadjuvant therapy(NAT).METHODS This retrospective study was conducted at the Chongqing Key Laboratory of Translational Research for Cancer Metastasis and Individualized Treatment,Chongqing University Cancer Hospital.This study specifically assessed CA19-9 levels and tumor size before and after NAT.RESULTS A total of 156 patients who completed NAT and subsequently underwent tumor resection were included in this study.The average age was 65.4±10.6 years and 72(46.2%)patients were female.Before survival analysis,we defined the post-NAT serum CA19-9 level/pre-NAT serum CA19-9 level as the CA19-9 ratio(CR).The patients were divided into three groups:CR0.5 and1.With regard to tumor size measured by both computed tomography and magnetic resonance imaging,we defined the post-NAT tumor size/pre-NAT tumor size as the tumor size ratio(TR).The patients were then divided into three groups:TR0.5 and1.Based on these groups divided according to CR and TR,we performed both overall survival(OS)and disease-free survival(DFS)analyses.Log-rank tests showed that both OS and DFS were significantly different among the groups according to CR and TR(P<0.05).CR and TR after NAT were associated with increased odds of achieving a complete or near-complete pathologic response.Moreover,CR(hazard ratio:1.721,95%CI:1.373-3.762;P=0.006),and TR(hazard ratio:1.435,95%CI:1.275-4.363;P=0.014)were identified as independent factors associated with OS.CONCLUSION This study demonstrated that post-NAT serum CA19-9 level/pre-NAT serum CA19-9 level and post-NAT tumor size/pre-NAT tumor size were independent factors associated with OS in patients with PDAC who received NAT and subsequent surgical resection.

Hereditary hearing loss(HHL),a genetic disorder that impairs auditory function,significantly affects quality of life and incurs substantial economic losses for society.To investigate the underlying causes of HHL and evaluate therapeutic outcomes,appropriate animal models are necessary.Pigs have been extensively used as valuable large animal models in biomedical research.In this review,we highlight the advantages of pig models in terms of ear anatomy,inner ear morphology,and electrophysiological characteristics,as well as recent advancements in the development of distinct genetically modified porcine models of hearing loss.Additionally,we discuss the prospects,challenges,and recommendations regarding the use pig models in HHL research.Overall,this review provides insights and perspectives for future studies on HHL using porcine models.

Improvement for creep strength of a second-generation single crystal superalloy by design of heat treatments

Design of heat treatments is related to the key technology for development of nickel-based single crystal superalloys(Ni-SXs). Based on the full understanding of the solidification characteristics, this work applies optimization design of heat treatments for a second-generation Ni-SX. Microstructure evolution and creep properties are compared in the material under conventional/standard(Std.) and optimized(Opt.) treatments. For the Std. sample,strong dendritic segregations determine inconsistent microstructure evolution in the dendritic(D) and interdendritic region(ID), while the latter serves as weak area to have the prior microcrack initiation, damaging overall performance of the alloy. The Opt. treatment applies higher homogenization temperature, leading to overall reduced segregations, while not inducing incipient melting. A lower temperature of first-step ageing is used to lower the size ofγ'particles. These help to form the more uniform microstructure in dendritic and interdendritic region and relieve the inconsistent microstructure evolution. The balanced local strength makes ID no longer as the weak area,thus restricting microcrack initiation. Great improvement of high temperature and low stress property is obtained by this progress, leading to the pronounced increase of creep rupture life under 1100 °C/140 MPa.

Rationally designing electrolyte additives for highly improving cyclability of LiNi_(0.5)Mn_(1.5)O_(4)/Graphite cells

High voltage is necessary for high energy lithium-ion batteries but difficult to achieve because of the highly deteriorated cyclability of the batteries.A novel strategy is developed to extend cyclability of a high voltage lithium-ion battery,LiNi_(0.5)Mn_(1.5)O_(4)/Graphite(LNMO/Graphite)cell,which emphasizes a rational design of an electrolyte additive that can effectively construct protective interphases on anode and cathode and highly eliminate the effect of hydrogen fluoride(HF).5-Trifluoromethylpyridine-trime thyl lithium borate(LTFMP-TMB),is synthesized,featuring with multi-functionalities.Its anion TFMPTMB-tends to be enriched on cathode and can be preferentially oxidized yielding TMB and radical TFMP-.Both TMB and radical TFMP can combine HF and thus eliminate the detrimental effect of HF on cathode,while the TMB dragged on cathode thus takes a preferential oxidation and constructs a protective cathode interphase.On the other hand,LTFMP-TMB is preferentially reduced on anode and constructs a protective anode interphase.Consequently,a small amount of LTFMP-TMB(0.2%)in 1.0 M LiPF6in EC/DEC/EMC(3/2/5,wt%)results in a highly improved cyclability of LNMO/Graphite cell,with the capacity retention enhanced from 52%to 80%after 150 cycles at 0.5 C between 3.5 and 4.8 V.The as-developed strategy provides a model of designing electrolyte additives for improving cyclability of high voltage batteries.

Assembled Photosensitizers Applied for Enhanced Photodynamic Therapy

Photodynamic therapy(PDT)has emerged as an efficient method for cancer therapy.However,traditional photosensitizers(PSs)always have low bioavailability.For example,hydrophobic PSs tend to aggregate in cells and lead to aggregation-induced quenching;while hydrophilic PSs that have good solubility in water systems can hardly penetrate into cells whose membranes are lipophilic.To overcome these drawbacks,suitable PSs that meet the requirements of PDT are needed.Numerous investigations have been introduced,especially the molecular-assembly technique that can increase the bioavailability of PSs during the tumor therapy process.Besides,increasing the quantum yield of reactive oxygen species(ROS)by adjusting the PS triplet state lifetime as well as developing aggregation-induced emission(AIE)agents can also improve the PDT effect.This review summarizes the molecular-assembly technique to obtain intelligent PSs to achieve high PDT efficiency.First,increasing the quantum yield of ROS by decreasing the energy gap between S_(1)and T_(1)states or increasing the spin–orbit coupling Hamiltonian are introduced.Second,we present the bioavailability of traditional PSs by improving the amphiphilicity of the PSs or using intelligent nanostructures.Then,the AIE PSs that can form ROS in the aggregated state under irradiation are described.Finally,the perspective and challenges of PDT are discussed.

RNA N6-methyladenosine(m^(6)A),as the most abundant modification of messenger RNA,can modulate insect behaviors,but its specific roles in aggregation behaviors remain unexplored.Here,we conducted a comprehensive molecular and physiological characterization of the individual components of the methyltransferase and demethylase in the migratory locust Locusta migratoria.Our results demonstrated that METTL3,METTL14 and ALKBH5 were dominantly expressed in the brain and exhibited remarkable responses to crowding or isolation.The individual knockdown of methyltransferases(i.e.,METTL3 and METTL14)promoted locust movement and conspecific attraction,whereas ALKBH5 knockdown induced a behavioral shift toward the solitary phase.Furthermore,global transcriptome profiles revealed that m^(6)A modification could regulate the orchestration of gene expression to fine tune the behavioral aggregation of locusts.In summary,our in vivo characterization of the m^(6)A functions in migratory locusts clearly demonstrated the crucial roles of the m^(6)A pathway in effectively modulating aggregation behaviors.

Hand,foot,and mouth disease(HFMD)is a common pediatric illness mainly caused by enteroviruses,which are important human pathogens.Currently,there are no available antiviral agents for the therapy of enterovirus infection.In this study,an excellent high-content antiviral screening system utilizing the EV-A71-eGFP reporter virus was developed.Using this screening system,we screened a drug library containing 1042 natural compounds to identify potential EV-A71 inhibitors.Fangchinoline(FAN),a bis-benzylisoquinoline alkaloid,exhibits potential inhibitory effects against various enteroviruses that cause HFMD,such as EV-A71,CV-A10,CV-B3 and CV-A16.Further investigations revealed that FAN targets the early stage of the enterovirus life cycle.Through the selection of FAN-resistant EV-A71 viruses,we demonstrated that the VP1 protein could be a potential target of FAN,as two mutations in VP1(E145G and V258I)resulted in viral resistance to FAN.Our research suggests that FAN is an efficient inhibitor of EV-A71 and has the potential to be a broad-spectrum antiviral drug against human enteroviruses.

Simultaneous high-performance detection and removal of tetracycline with a polyoxomolybdate-based coordination polymer

The excessive residues of tetracycline(TC) have been found to cause persistent adverse effects on both human beings and the environment. In this study, a polyoxomolybdate-based coordination polymer(BUC-14) with negative surface and functional group(-NH2) was selected as a multifunctional platform for the detection and removal of TC. The fluorescence emission of BUC-14 at 468 nm was observed to be quenched by TC in a wide concentration range of 7–107 μM attributed to the inner filter effect(IFE) and photo-induced electron transfer(PET) with a detection limit(LOD) of 38.7 n M. The adsorption properties for TC were thoroughly studied through kinetic and equilibrium experiments. And the excellent adsorption performance significantly can promote the preconcentration of TC, thereby enhancing the sensing performance. Possible mechanisms of adsorption including electrostatic attraction, hydrogen bonding and π-π interactions were studied. Furthermore, the practicality of BUC-14 was confirmed by successfully sensing TC in real water samples with satisfactory recovery from 90.61% to103.53%, and a relative standard deviation(RSD) ≤ 5.12%.
