This work aims to investigate the effects of dosing regiments on drug delivery in solid tumors and to validate them with experiments on rats. The lumped parameter models of pharmacokinetics and of drug delivery in tumor were developed to simulate time courses of average drug concentration （Ct） of tumor interstitium in two types of dosing regiments （i.e., single-shot and triple-shot ones）. The two regiments were performed via antitumor drug, hydroxycamptothecin （HCPT）, on rats, to measure the drug concentration in the tumor. The simulations of the drug concentration in the tumor of the two dosing regiments were conducted and compared with the experimental data on rats. The coefficients in the models were investigated. It is concluded that the triple-shot method is more effective than that of single-shot injection. The present lumped-parameter model is quantitatively competent for drug delivery in solid tumor.

目的分析臭灵丹提取物调控转化生长因子-β/骨形成蛋白(TGF-β/BMPs)信号通路对股骨粗隆间骨折固定术后感染大鼠的影响.方法选取SPF级Wistar大鼠44只,10只大鼠作为空白组,剩余34只建立股骨粗隆间骨折固定术后感染模型,共有30只大鼠建模成功,将其分为模型组、药物对照组、臭灵丹提取物组3组,每组10只.药物对照组给予阿莫西林,臭灵丹提取物组给予臭灵丹提取物,空白组、模型组不做处理,干预7 d后观察大鼠变化.结果与空白组相比,模型组、药物对照组、臭灵丹提取物组肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、Ⅰ型胶原羧基端肽β特殊序列(β-CTX)水平上升,IL-10、Ⅰ型原胶原N-端前肽(PⅠNP)、骨碱性磷酸酶(BALP)水平、转化生长因子-β(TGF-β)mRNA、骨形态发生蛋白-2(BMP-2)mRNA表达量及相对表达量下降(P<0.05);与模型组相比,药物对照组、臭灵丹提取物组TNF-α、IL-6、β-CTX水平下降,IL-10、PⅠNP、BALP、TGF-βmRNA、BMP-2 mRNA表达量及相对表达量上升(P<0.05);与药物对照组相比,臭灵丹提取物组TNF-α、IL-6、β-CTX水平下降,IL-10、PⅠNP、BALP水平、TGF-βmRNA、BMP-2 mRNA表达量及相对表达量上升(P<0.05).结论股骨粗隆间骨折固定术后感染大鼠经臭灵丹提取物干预后炎性反应减轻,骨代谢改善,其机制可能与TGF-β/BMPs信号通路激活有关.

Numerical simulation of hemodynamics under the combined effects of both the host blood vessel and the microvascular network, which is based on a 2-D tumor inside and outside vascular network generated from a discrete mathematical model of tumor-induced angiogenesis, is performed systemically. And a ＂microvascular network-- transport across microvascular network--flow in interstitium＂ model is developed to study the flow in solid tumor. Simulations are carried out to examine the effects of the variations of the inlet Reynolds number in the host blood vessel, the hydraulic conductivity of the microvascular wall, and interstitial hydraulic conductivity coefficient on the fluid flow in tumor microcirculation. The results are consistent with data obtained in terms of physiology. These results may provide some theoretical references and the bases for further clinical experimental research.

Microtubule Structure and Male Sterility in a Gene-Cytoplasmic Male Sterile Line of Rice, Zhen Shan 97A

Histological changes that occur during microsporogenesis are documented in a gene-cytoplasmic male sterile rice ( Oryza saliva L.) line, Zhen Shan 97A, its maintainer line, Zhen Shan 97B, and the restorer line, Ce64 of a Mine hybrid rice production system. In the restorer line, Ce64, the developing microsporocytes have dense cytoplasm and a distinct set of circumferential microtubules around the nucleus. Successive cytokinesis results in the formation of tetrads. The microtubules within the cells of tetrads and microspores radiate from the surface of the nucleus towards the outer edge of the cytoplasm. Subsequent pollen development is normal. During the course of microspore formation tubulin speckles can be found in the cytoplasm. The general pattern of development and microtubule organization in the maintainer lined Zhen Shan 97B, is similar to Ce64, except that a few more tubulin speckles appear during microspore formation. In the case of the mate sterile line, Zhen Shan 97A, a number of abnormalities can be discerned during early microsporogenesis. These include vacuoles forming within the developing microsporocyte and faintly stained microtubules with no defined distribution pattern. Prominent tubulin speckles are common within the cytoplasm. For those microsporocytes that undergo meiosis, no defined organizational patterns of microtubules can be found within the tetrad. All microspores abort soon after. Abnormalities and defects in microtubule organization observed in Zhen Shan 97A showed that complex interactions between the cytoplasm and the nucleus began at very early stage of microsporocyte development.

Double Toeplitz（shortly DT） codes are introduced here as a generalization of double circulant codes. The authors show that such a code is isodual, hence formally self-dual（FSD）. FSD codes form a far-reaching generalization of self-dual codes, the most important class of codes of rate one-half. Self-dual DT codes are characterized as double circulant or double negacirculant. Likewise, even binary DT codes are characterized as double circulant. Numerical examples obtained by exhaustive search show that the codes constructed have best-known minimum distance, up to one unit, amongst formally self-dual codes, and sometimes improve on the known values. For q = 2, the authors find four improvements on the best-known values of the minimum distance of FSD codes. Over F4an explicit construction of DT codes, based on quadratic residues in a prime field, performs equally well. The authors show that DT codes are asymptotically good over Fq. Specifically, the authors construct DT codes arbitrarily close to the asymptotic Varshamov-Gilbert bound for codes of rate one half.

In this paper a mathematical model is developed for the description of the pulsatile blood flow through an axisymmetric artery of mild varying cross-section, taking into account the tethering effect of the sorrounding *** on the linear Navier-Stokes equation and boundary conditions,a set of differential equa- tions for the modulus of phase velocity,blood velocities,pressure and deformation of the vessel wall are deduced and the numerical solutions are found by utilizing the Runge-Kutta *** axial velocity profiles and the peak wall shear stresses are given for both mild tapering tube and mild stenosed *** is shown that the peak wall shear stresses in the tethered tube are higher than those in the free *** is also shown that in a series of previously research work on peak wall shear stresses of varying cross-section vessels the free vessel assumption has the tendency of under assessing the peak wall shear stresses and vice versa for the rigid vessel assumption.

Actin Visualization in Living Immature Pollen of Rice Using a GFP-Mouse Talin Fusion Protein

Green fluorescent protein (GFP) fused to the F_actin binding domain of mouse talin labels the actin cytoskeleton in the immature pollen of stable transformed rice (Oryza sativa L.) plants. Actin microfilaments could be visualized only in the late_developmental stage of the immature pollen. During this developmental stage, microfilaments, initially composed of very short fibrils, develop into a very complex and novel network that sometimes totally and sometimes partially encloses the vegetative nucleus and the spherical shaped generative cell in the central cytoplasm of the immature pollen. The behavior of the actin microfilamentous structure throughout the late_developmental stage of the immature pollen is extremely dynamic, and the likelihood of this structure in generating forces for vegetative nucleus and generative cell movement in the immature pollen has been discussed. No actin filaments were visualized in the spherical generative cells.
