Vascular endothelium can perceive fluid shear stress(FSS)and cyclic circumferential stretch(CCS)caused by the pulsatile blood flow,and translate the hemodynamics into biochemical signals to regulate vascular ***,existing methods provide little information about the real-time biochemical responses of endothelium when exposed to dynamic FSS and ***,a vasculature-on-a-chip integrated with stretchable sensing is engineered for recapitulating the hemodynamic milieus and in-situ monitoring biochemical responses of endothelial *** integrated device is developed by sandwiching a robust stretchable electrode between an upper fluidic channel and a lower pneumatic *** fluidic and pneumatic channels enable the simultaneous recapitulation of both FSS and CCS,and the integrated sensor exhibits excellent cell-adhesive capacity and electrochemical sensing stability even after long-term hemodynamic *** allow real-time monitoring of hemodynamic form-and duration-dependent endothelium responses,and further efficacy investigation about a recommended drug for COVID-19,demonstrating the great potential in vascular disease and drug screening.

Background Osteosarcoma （OS） is the most common primary malignant tumor of bone. Mouse models of human OS can invariably provide greater insight into the complex mechanisms that underlie the development and pathogenesis of this aggressive tumor. Bioluminescence technology favored tracing cancer cells in vivo. In this study, an OS model was described and evaluated using human OS cell line, Saos2, labeled with luciferase （Saos2-1uc）. Methods Saos2 cells were infected by lentivirus loading a firefly luciferase gene. Luciferase expression of Saos2-1uc cells was characterized both in vitro and in vivo. Specific biologic and oncologic features of Saos2-1uc cells were analyzed. The OS was established as orthotopic xenografts in nude mice. Both orthotopic tumors and spontaneous lung metastasis were analyzed. Results Tumorigenesis and spontaneous lung metastasis in nude mice could be monitored in vivo through in vivo imaging system. The enhancement in proliferation, migration and invasion abilities and the attenuation in adhesion ability were observed in Saos2-1uc cells compared with Saos2 cells. Furthermore, there were the up-regulation of Osteocalcin, CCRIO, CXCR1 and ID1 and the down-regulation of ALP, collagen I, CCR1, CCR3, CXCR3, NID and N-cadherin in Saos2-1uc cells compare to Saos2 cells. The rate of spontaneous lung metastasis in Saos2-1uc cells was higher than that in Saos2 cells, although without significant difference. Conclusions Lentivirus transfection may cause alteration of gene expression profiles and further biological functions. This model can be used in the elucidation of molecular mechanisms of tumorigenesis and the screening of new therapeutic agents.

In this study,focusing on the geometry characteristics of spherical expanding flame,the turbulent premixed flames of natural gas/air mixtures were investigated in a fan-stirred turbulent *** effects of initial temperature(T=300-400 K),initial pressure(P=0.1-0.3 MPa),turbulence intensity(u'=1.0-2.7 m/s),oxygen volumetric percentage(φ(O_(2))=15%-21%)and carbon dioxide volumetric percentage(φ(CO_(2))=0-20%)were delved *** flame profile under the Cartesian coordinate system was derived from the schlieren images taken by the high-speed ***,from both macroscopic and microscopic perspectives,the influence of experimental conditions on the flame geometry characteristics was explored through flame front extraction,wavelet decomposition and network *** results demonstrate that for significant flame wrinkling,changes in species concentrations and turbulence intensity have more pronounced effects on the flame wrinkling *** wrinkling of the flame front maintains a certain degree of similarity,as evidenced by the locally concentrated distribution of the angles of the maximum fluctuation *** disturbance energy under large-scale(D6-D8)disturbances exhibits relatively high values with a similar trend,exerting a significant impact on the geometry characteristics of the flame *** peaks of correlation degree are scattered either with the decomposition scale or the development of flame radius,indicating no linear correlation between different detail ***,the probability distribution of node degrees in key wrinkled regions exhibits different trends with that of large-scale wrinkling and disturbance energy,especially with changes in initial *** occurs because the number of key wrinkles varies based on the perturbation's strength or the region's ***,an increase in the fluctuation frequency of the flame's local radius may not necessarily lead to an increase in the number of key folded regions.

Hematophagous female mosquitoes are important vectors of numerous devastating human diseases,posing a major public health *** prevention and control of mosquito-borne diseases rely considerably on progress in understanding the molecular mechanisms of various life activities,and accordingly,the molecules that regulate the various life activities of mosquitoes are potential targets for implementing future vector control *** long non-coding RNAs(lncRNAs)have been identified in mosquitoes and significant progress has been made in determining their ***,we present a comprehensive overview of the research advances on mosquito lncRNAs,including their molecular identification,function,and interaction with other non-coding RNAs,as well as their synergistic regulatory roles in mosquito life *** also highlight the potential roles of competitive endogenous RNAs in mosquito growth and development,as well as in insecticide resistance and virus–host *** into the biological functions and mechanisms of lncRNAs in mosquito life activities,viral replication,pathogenesis,and transmission will contribute to the development of novel drugs and safe vaccines.

Background:Computed tomography(CT)plays a great role in characterizing and quantifying changes in lung structure and function of chronic obstructive pulmonary disease(COPD).This study aimed to explore the performance of CT-based whole lung radiomic in discriminating COPD patients and non-COPD ***:This retrospective study was performed on 2785 patients who underwent pulmonary function examination in 5 hospitals and were divided into non-COPD group and COPD *** radiomic features of the whole lung volume were *** absolute shrinkage and selection operator(LASSO)logistic regression was applied for feature selection and radiomic signature construction.A radiomic nomogram was established by combining the radiomic score and clinical *** operating characteristic(ROC)curve analysis and decision curve analysis(DCA)were used to evaluate the predictive performance of the radiomic nomogram in the training,internal validation,and independent external validation ***:Eighteen radiomic features were collected from the whole lung volume to construct a radiomic *** area under the curve(AUC)of the radiomic model in the training,internal,and independent external validation cohorts were 0.888[95%confidence interval(CI)0.869–0.906],0.874(95%CI 0.844–0.904),and 0.846(95%CI 0.822–0.870),*** were higher than the clinical model(AUC were 0.732,0.714,and 0.777,respectively,P<0.001).DCA demonstrated that the nomogram constructed by combining radiomic score,age,sex,height,and smoking status was superior to the clinical factor ***:The intuitive nomogram constructed by CT-based whole-lung radiomic has shown good performance and high accuracy in identifying COPD in this multicenter study.

The silk gland of the silkworm Bombyx mori serves as a valuable model for investigating the morphological structure and physiological functions of *** studies have demonstrated the notable regulatory role of let-7 microRNA in the silk gland,but its specific molecular mechanism remains to be elucidated across different segments of this *** this study,we further investigated the functional mechanism of let-7 in the middle silk gland(MSG).The MSG of a let-7 knockout strain was analyzed using a combined proteomic and metabolomic technique,revealing the enrichment of differential proteins and metabolites in the DNA synthesis and energy metabolism *** was identified as a novel target gene of let-7 in the MSG,and its downregulation inhibited the proliferation of BmN4-SID1 cells,which is exactly opposite to the role of let-7 in these ***/Cas9 genome editing and transgenic technologies were employed to manipulate BmCentrin in the *** of BmCentrin led to severe MSG atrophy,whereas the overexpression of BmCentrin resulted in beaded *** measurements of these knockout or overexpression strains revealed significant changes in the expression levels of sericin protein genes,the weight of the cocoon and the mechanical properties of the *** the biological role of BmCentrin in the silk gland offers valuable insights for elucidating the molecular mechanisms by which let-7 controls silk gland development and silk protein synthesis in the silkworm.

Accumulation characteristics and fate modeling of phthalic acid esters in surface water from the Three Gorges Reservoir area,China

Phthalic acid esters(PAEs)are a group of compounds widespread in the *** investigate the occurrence and accumulation characteristics of PAEs,surface water samples were collected from the Three Gorges Reservoir area,*** total concentrations of∑_(11)analyzed PAEs(11PAEs)in the collected water samples ranging from 197.7 to 1,409.3 ng/L(mean±IQR:583.1±308.4 ng/L).While DEHP was the most frequently detected PAE,DnBP and DnNP were the most predominant PAEs in the analyzed water samples with a mean contribution of 63.3%of the∑_(11)*** concentrations of the∑_(11)PAEs in the water samples from the upper reaches of the Yangtze River were significantly higher than those from themiddle *** better understand the transport and fate of the PAEs,seven detected PAEs were modeled by Quantitative Water Air Sediment Interaction(QWASI).The simulated and measured values were close for most PAEs,and differences are within one order of magnitude even for the worst *** all simulated PAEs,water and particle inflow were main sources in the reservoir,whereas water outflow and degradation in water were important removal *** contribution ratios of different sources/losses varied fromPAEs,depending on their *** calculated risk quotients of DnNP in the Three Gorges Reservoir area whether based onmonitoring or simulating results were all far exceeded the safety threshold value,implying the occurrence of this PAE compound may cause potential adverse effects for the aquatic ecology of the Three Gorges Reservoir area.
