Toward a Learnable Climate Model in the Artificial Intelligence Era

Artificial intelligence(AI)models have significantly impacted various areas of the atmospheric sciences,reshaping our approach to climate-related challenges.Amid this AI-driven transformation,the foundational role of physics in climate science has occasionally been overlooked.Our perspective suggests that the future of climate modeling involves a synergistic partnership between AI and physics,rather than an“either/or”scenario.Scrutinizing controversies around current physical inconsistencies in large AI models,we stress the critical need for detailed dynamic diagnostics and physical constraints.Furthermore,we provide illustrative examples to guide future assessments and constraints for AI models.Regarding AI integration with numerical models,we argue that offline AI parameterization schemes may fall short of achieving global optimality,emphasizing the importance of constructing online schemes.Additionally,we highlight the significance of fostering a community culture and propose the OCR(Open,Comparable,Reproducible)principles.Through a better community culture and a deep integration of physics and AI,we contend that developing a learnable climate model,balancing AI and physics,is an achievable goal.

A method for stereoselective construction of Z-monofluoroalkenes by nickel-catalyzed defluorinative coupling of gem‑difluoroalkenes in mild conditions was described.The combination of lithium organoborate and ZnBr_(2) generated in situ lithium aryl zincates,which facilitates the transmetalation step of the nickel-catalyzed cross coupling reaction.

Correcting Climate Model Sea Surface Temperature Simulations with Generative Adversarial Networks:Climatology,Interannual Variability,and Extremes

Climate models are vital for understanding and projecting global climate change and its associated impacts.However,these models suffer from biases that limit their accuracy in historical simulations and the trustworthiness of future projections.Addressing these challenges requires addressing internal variability,hindering the direct alignment between model simulations and observations,and thwarting conventional supervised learning methods.Here,we employ an unsupervised Cycle-consistent Generative Adversarial Network(CycleGAN),to correct daily Sea Surface Temperature(SST)simulations from the Community Earth System Model 2(CESM2).Our results reveal that the CycleGAN not only corrects climatological biases but also improves the simulation of major dynamic modes including the El Niño-Southern Oscillation(ENSO)and the Indian Ocean Dipole mode,as well as SST extremes.Notably,it substantially corrects climatological SST biases,decreasing the globally averaged Root-Mean-Square Error(RMSE)by 58%.Intriguingly,the CycleGAN effectively addresses the well-known excessive westward bias in ENSO SST anomalies,a common issue in climate models that traditional methods,like quantile mapping,struggle to rectify.Additionally,it substantially improves the simulation of SST extremes,raising the pattern correlation coefficient(PCC)from 0.56 to 0.88 and lowering the RMSE from 0.5 to 0.32.This enhancement is attributed to better representations of interannual,intraseasonal,and synoptic scales variabilities.Our study offers a novel approach to correct global SST simulations and underscores its effectiveness across different time scales and primary dynamical modes.

m^(6)A demethylase FTO regulate CTNNB1 to promote adipogenesis of chicken preadipocyte

Background:N6-methyladenosine(m^(6)A)is an abundant post-transcriptional RNA modification that affects various biological processes.The fat mass and obesity-associated(FTO)protein,a demethylase encoded by the FTO gene,has been found to regulate adipocyte development in an m^(6)A-dependent manner in multiple species.However,the effects of the m^(6)A methylation and FTO demethylation functions on chicken adipogenesis remain unclear.This study aims to explore the association between m^(6)A modification and chicken adipogenesis and the underlying mechanism by which FTO affects chicken preadipocyte development.Results:The association between m^(6)A modification and chicken lipogenesis was assessed by treating chicken pread-ipocytes with different doses of methyl donor betaine and methylation inhibitor cycloleucine.The results showed that betaine significantly increased methylation levels and inhibited lipogenesis,and the inverse effect was found in preadipocytes after cycloleucine treatment.Overexpression of FTO significantly inhibited m^(6)A levels and promoted proliferation and differentiation of chicken preadipocytes.Silencing FTO showed opposite results.Mechanistically,FTO overexpression increased the expression of catenin beta 1(CTNNB1)by improving RNA stability in an m^(6)A-dependent manner,and we proved that FTO could directly target CTNNB1.Furthermore,CTNNB1 may be a positive regulator of adipogenesis in chicken preadipocytes.Conclusions:m^(6)A methylation of RNA was negatively associated with adipogenesis of chicken preadipocytes.FTO could regulate CTNNB1 expression in a demethylation manner to promote lipogenesis.

Poaceae(the grasses)includes rice,maize,wheat,and other crops,and is the most economically important angiosperm family.Poaceae is also one of the largest plant families,consisting of over 11000 species with a global distribution that contributes to diverse ecosystems.Poaceae species are classified into 12 subfamilies,with generally strong phylogenetic support for their monophyly.However,many relationships within subfamilies,among tribes and/or subtribes,remain uncertain.To better resolve the Poaceae phylogeny,we generated 342 transcriptomic and seven genomic datasets;these were combined with other genomic and transcriptomic datasets to provide sequences for 357 Poaceae species in 231 genera,representing 45 tribes and all 12 subfamilies.Over 1200 low-copy nuclear genes were retrieved from these datasets,with several subsets obtained using additional criteria,and used for coalescent analyses to reconstruct a Poaceae phylogeny.Our results strongly support the monophyly of 11 subfamilies;however,the subfamily Puelioideae was separated into two non-sister clades,one for each of the two previously defined tribes,supporting a hypothesis that places each tribe in a separate subfamily.Molecular clock analyses estimated the crown age of Poaceae to be101 million years old.Ancestral character reconstruction of C3/C4 photosynthesis supports the hypothesis of multiple independent origins of C4 photosynthesis.These origins are further supported by phylogenetic analysis of the ppc gene family that encodes the phosphoenolpyruvate carboxylase,which suggests that members of three paralogous subclades(ppc-aL1a,ppc-aL1b,and ppcB2)were recruited as functional C4 ppc genes.This study provides valuable resources and a robust phylogenetic framework for evolutionary analyses of the grass family.

Associations of daily sedentary behavior,physical activity,and sleep with irritable bowel syndrome:A prospective analysis of 362,193 participants

Background:Irritable bowel syndrome(IBS)substantially affects quality of life and requires early prevention.This study aimed to elucidate the relationships between IBS and daily behaviors,including sedentary behavior(SB),physical activity(PA),and sleep.In particular,it seeks to identify healthy behaviors to reduce IBS risk,which previous studies have rarely addressed.Methods:Daily behaviors were retrieved from self-reported data of 362,193 eligible UK Biobank participants.Incident cases were determined by self-report or health care data according to RomeⅣcriteria.Results:A total of 345,388 participants were IBS-free at baseline,during a median follow-up of 8.45 years,19,885 incident IBS cases were recorded.When examined individually,SB and shorter(≤7 h/day)or longer(>7 h/day)sleep duration were each positively associated with increased IBS risk,and PA was associated with lower IBS risk.The isotemporal substitution model suggested that replacing SB with other activities could provide further protective effects against IBS risk.Among people sleeping≤7 h/day,replacing 1 h of SB with equivalent light PA,vigorous PA,or sleep was associated with 8.1%(95%confidence interval(95%CI):0.901-0.937),5.8%(95%CI:0.896-0.991),and 9.2%(95%CI:0.885-0.932)reduced IBS risk,respectively.For people sleeping>7 h/day,light and vigorous PA were associated with a 4.8%(95%CI:0.926-0.978)and a 12.0%(95%CI:0.815-0.949)lower IBS risk,respectively.These benefits were mostly independent of genetic risk for IBS.Conclusion:SB and unhealthy sleep duration are risk factors for IBS.A promising way to mitigate IBS risk for individuals sleeping≤7 h/day and for those sleeping>7 h/day appears to be by replacing SB with adequate sleep or vigorous PA,respectively,regardless of the genetic predisposition of IBS.

Fast Blood Oxygenation through Hemocompatible Asymmetric Polymer of Intrinsic Microporosity Membranes

Membrane technology has attracted considerable attention for chemical and medical applications,among others.Artificial organs play important roles in medical science.A membrane oxygenator,also known as artificial lung,can replenish O_(2) and remove CO_(2) of blood to maintain the metabolism of patients with cardiopulmonary failure.However,the membrane,a key component,is subjected to inferior gas transport property,leakage propensity,and insufficient hemocompatibility.In this study,we report efficient blood oxygenation by using an asymmetric nanoporous membrane that is fabricated using the classic nonsolvent-induced phase separation method for polymer of intrinsic microporosity-1.The intrinsic superhydrophobic nanopores and asymmetric configuration endow the membrane with water impermeability and gas ultrapermeability,up to 3,500 and 1,100 gas permeation units for CO_(2) and O_(2),respectively.Moreover,the rational hydrophobic–hydrophilic nature,electronegativity,and smoothness of the surface enable the substantially restricted protein adsorption,platelet adhesion and activation,hemolysis,and thrombosis for the membrane.Importantly,during blood oxygenation,the asymmetric nanoporous membrane shows no thrombus formation and plasma leakage and exhibits fast O_(2) and CO_(2) transport processes with exchange rates of 20 to 60 and 100 to 350 ml m^(−2) min^(−1),respectively,which are 2 to 6 times higher than those of conventional membranes.The concepts reported here offer an alternative route to fabricate high-performance membranes and expand the possibilities of nanoporous materials for membrane-based artificial organs.

A thermodynamic perspective on electrode poisoning in solid oxide fuel cells

A critical challenge to the commercialization of clean and high-efficiency solid oxide fuel cell(SOFC)technology is the insuf-ficient stack lifespan caused by a variety of degradation mechanisms,which are associated with cell components and chemical feedstocks.Cell components related degradation refers to thermal/chemical/electrochemical deterioration of cell materials under operating conditions,whereas the latter regards impurities in feedstocks of oxidant(air)and reductant(fuel).This article provides a thermodynamic perspective on the understanding of the impurities-induced degradation mechanisms in SOFCs.The discussion focuses on using thermodynamic ana-lysis to elucidate poisoning mechanisms in cathodes by impurity species such as Cr,CO_(2),H_(2)O,and SO_(2) and in the anode by species such as S(or H_(2)S),SiO_(2),and P_(2)(or PH_(3)).The author hopes the presented fundamental insights can provide a theoretical foundation for search-ing for better technical solutions to address the critical degradation challenges.

In situ direct reprogramming of astrocytes to neurons via polypyrimidine tract-binding protein 1 knockdown in a mouse model of ischemic stroke

In situ direct reprogramming technology can directly convert endogenous glial cells into functional neurons in vivo for central nervous system repair. Polypyrimidine tract-binding protein 1(PTB) knockdown has been shown to reprogram astrocytes to functional neurons in situ. In this study, we used AAV-PHP.e B-GFAP-sh PTB to knockdown PTB in a mouse model of ischemic stroke induced by endothelin-1, and investigated the effects of GFAP-sh PTB-mediated direct reprogramming to neurons. Our results showed that in the mouse model of ischemic stroke, PTB knockdown effectively reprogrammed GFAP-positive cells to neurons in ischemic foci, restored neural tissue structure, reduced inflammatory response, and improved behavioral function. These findings validate the effectiveness of in situ transdifferentiation of astrocytes, and suggest that the approach may be a promising strategy for stroke treatment.

Malignant hyperthermia(MH)is a genetic disorder of skeletal muscle cells that affects muscle cytoplasmic calcium homeostasis,with high mortality and low morbidity.Generally,it presents with non-specific signs of a hypermetabolic response,including high fever,tachycardia,and elevated end-tidal carbon dioxide(ETCO_(2)).The successful treatment lies in the timely recognition and early use of dantrolene.[1]As an inhibitor of Ca2+release through ryanodine receptor(RYR)channels,the skeletal muscle relaxant dantrolene has proven to be both a valuable experimental probe of intracellular Ca2+signaling and a lifesaving treatment for MH.[2]Dominant mutations in the skeletal muscle RYR1 gene are well-recognized causes of both malignant hyperthermia susceptibility(MHS)and central core disease(CCD).
