Role of T-cell receptor V beta 8.3 peptide vaccine in the prevention of experimental autoimmune uveoretinitis

Background T-cell receptor （TCR） plays an important role in the development of autoimmune diseases. Recently, it was reported that immunization of animals with TCR peptide derived from the pathogenic cells could prevent autoimmune diseases. The aim of this study was to investigate whether vaccination with a synthetic peptide from the hypervariable region of TCR Vβ 8.3, an experimental autoimmune uveoretinitis （EAU）-associated gene, was able to prevent the disease. Methods EAU was induced in Lewis rats by immunization with IRBP R16 peptide emulsified in complete Freund＇s adjuvant （CFA）. The clinical and histological appearances were scored. Delayed type hypersensitivity （DTH） and lymphocyte proliferation were detected. Cytokine levels of aqueous humour, supernatants of cells from spleen and draining lymph nodes were measured by enzyme linked immunosorbent assay （ELISA）. Gene expression of TCR Vβ8.3 on CD4^＋ T cells was examined by real time quantitative polymerase chain reaction （PCR）. Results After vaccination, the intraocular inflammation was significantly mitigated, antigen specific DTH and lymphocyte proliferation responses were suppressed, interleukin （IL）-2 in aqueous humour, interferon （IFN）-γ and IL-2 produced by the spleen and draining lymph node cells were significantly decreased, whereas the production of IL-4 and IL-10 were increased. The response of draining lymph node cells to TCR Vβ 8.3 peptide was enhanced after vaccination. Inoculation with CFA alone did not affect the severity of EAU and the above parameters. The suppression of EAU was much stronger in the group of four fold inoculations than the group of two fold inoculations. The expression of TCR Vβ 8.3 gene was significantly reduced in the group of fourfold inoculations. Conclusion Vaccination with the synthetic TCR Vβ 8.3 peptide could remarkably inhibit the development of EAU.

Crossed renal ectopia with rectal cancer:A case report

BACKGROUND Crossed renal ectopia(CRE)occurs when one kidney crosses the midline from the primary side to the contralateral side while the ureter remains on the primary side.Rectal cancer,one of the most common malignant tumors of the digestive tract,refers to cancer from the dentate line to the rectosigmoid junction.The concurrent presentation of CRE alongside rectal cancer is an uncommon clinical observation.CASE SUMMARY Herein,we report a 69-year-old male patient with rectal cancer who was diagnosed with CRE via computed tomography during hospitalization.Following thorough preoperative evaluations,the patient underwent Dixon surgery.CONCLUSION We performed laparoscopic radical resection of rectal cancer and adequate lymph node removal in a patient with CRE with no postoperative discomfort.

Buffer overflow poses a serious threat to the memory security of modern operating systems.It overwrites the con-tents of other memory areas by breaking through the buffer capacity limit,destroys the system execution environ-ment,and provides implementation space for various system attacks such as program control flow hijacking.That makes it a wide range of harms.A variety of security technologies have been proposed to deal with system security problems including buffer overflow.For example,No eXecute(NX for short)is a memory management technology commonly used in Harvard architecture.It can refuse the execution of code which residing in a specific memory,and can effectively suppress the abnormal impact of buffer overflow on control flow.Therefore,in recent years,it has also been used in the field of system security,deriving a series of solutions based on NX technology,such as ExecShield,DEP,StackGuard,etc.However,these security solutions often rely too much on the processor archi-tecture so that the protection coverage is insufficient and the accuracy is limited.Especially in the emerging system architecture field represented by RiSC-V,there is still a lack of effective solutions for buffer overflow vulnerabilities.With the continuous rapid development of the system architecture,it is urgent to develop defense methods that are applicable to different system application environments and oriented to all executable memory spaces to meet the needs of system security development.Therefore,we propose BOP,A new system memory security design method based on RISC-V extended instructions,to build a RISC-V buffer overflow detection and defense system and deal with the buffer overflow threat in RIsC-V.According to this method,NX technology can be combined with program control flow analysis,and Nx bit mechanism can be used to manage the executability of memory space,so as to achieve a more granular detection and defense of buffer overflow attacks that may occur in RISC-V system environment.In addition,The memory management an

Generalized load graphical forecasting method based on modal decomposition

In a“low-carbon”context,the power load is affected by the coupling of multiple factors,which gradually evolves from the traditional“pure load”to the generalized load with the dual characteristics of“load+power supply.”Traditional time-series forecasting methods are no longer suitable owing to the complexity and uncertainty associated with generalized loads.From the perspective of image processing,this study proposes a graphical short-term prediction method for generalized loads based on modal decomposition.First,the datasets are normalized and feature-filtered by comparing the results of Xtreme gradient boosting,gradient boosted decision tree,and random forest algorithms.Subsequently,the generalized load data are decomposed into three sets of modalities by modal decomposition,and red,green,and blue(RGB)images are generated using them as the pixel values of the R,G,and B channels.The generated images are diversified,and an optimized DenseNet neural network was used for training and prediction.Finally,the base load,wind power,and photovoltaic power generation data are selected,and the characteristic curves of the generalized load scenarios under different permeabilities of wind power and photovoltaic power generation are obtained using the density-based spatial clustering of applications with noise algorithm.Based on the proposed graphical forecasting method,the feasibility of the generalized load graphical forecasting method is verified by comparing it with the traditional time-series forecasting method.

Organofluorine compounds are widely used in the realm of drug discovery and material science.Herein,we developed palladium catalyzed intermolecular aminofluorination and oxy-aminofluorination of gem‑difluoroalkenes with N-fluorobenzenesulfonimide(NFSI),in which NFSI was used as the nitrogen source and oxidant.The reaction provides an efficient and straightforward synthesis route of a series ofα-trifluoromethyl benzylic amines.Notably,three/four components oxy-aminofluorination processes were realized to giveα-trifluoromethyl benzylic ether with a terminal amino group,which proceed through C(sp^(3))-O bond cleavage of easily available ether and simultaneous introduced a fluorine,an amino and an oxy substituent in one pot with excellent regioselectivity.The divergent reactivity not only included the incorporation of one ether molecular,but also much more challenged two ether insertion with excellent selectivity through succession C(sp^(3))-O bonds cleavage.This protocol allows for concise synthesis of high value amines with fluoroalkyl-substituents and selectively transformation of easily available ethers by high-valent palladium catalysis.

Macrophages,a heterogeneous population of innate immune cells,exhibit remarkable plasticity and play pivotal roles in coordinating immune responses and maintaining tissue homeostasis within the context of metabolic diseases.The activation of inflammatory macrophages in obese adipose tissue leads to detrimental effects,inducing insulin resistance through increased inflammation,impaired thermogenesis,and adipose tissue fibrosis.Meanwhile,adipose tissue macrophages also play a beneficial role in maintaining adipose tissue homeostasis by regulating angiogenesis,facilitating the clearance of dead adipocytes,and promoting mitochondrial transfer.Exploring the heterogeneity of macrophages in obese adipose tissue is crucial for unraveling the pathogenesis of obesity and holds significant potential for targeted therapeutic interventions.Recently,the dual effects and some potential regulatory mechanisms of macrophages in adipose tissue have been elucidated using single-cell technology.In this review,we present a comprehensive overview of the intricate activation mechanisms and diverse functions of macrophages in adipose tissue during obesity,as well as explore the potential of drug delivery systems targeting macrophages,aiming to enhance the understanding of current regulatory mechanisms that may be potentially targeted for treating obesity or metabolic diseases.

Geometry and formation mechanism of tension gashes and their implication on the hydrocarbon accumulation in the deep-seated strata of sedimentary basin:A case from Shunnan area of Tarim Basin

With the theoretical and technological developments related to cratonic strike-slip faults,the Shuntuoguole Low Uplift in the Tarim Basin has attracted considerable attention recently.Affected by multi-stage tectonic movements,the strike-slip faults have controlled the distribution of hydrocarbon resources owing to the special fault characteristics and fault-related structures.In contrast,the kinematics and formation mechanism of strike-slip faults in buried sedimentary basins are difficult to investigate,limiting the discussion of these faults and hydrocarbon accumulation.In this study,we identified the characteristics of massive sigmoidal tension gashes(STGs)that formed in the Shunnan area of the Tarim Basin.High-resolution three-dimensional seismic data and attribute analyses were used to investigate their geometric and kinematic characteristics.Then,the stress state of each point of the STGs was calculated using seismic curvature attributes.Finally,the formation mechanism of the STGs and their roles in controlling hydrocarbon migration and accumulation were discussed.The results suggest that:(1)the STGs developed in the Shunnan area have a wide distribution,with a tensile fault arranged in an enéchelon pattern,showing an S-shaped bending.These STGs formed in multiple stages,and differential rotation occurred along the direction of strike-slip stress during formation.(2)Near the principal displacement zone of the strike-slip faults,the stress value of the STGs was higher,gradually decreasing at both ends.The shallow layer deformation was greater than the deep layer deformation.(3)STGs are critical for connecting source rocks,migrating oil and gas,sealing horizontally,and developing efficient reservoirs.This study not only provides seismic evidence for the formation and evolution of super large STGs,but also provides certain guidance for oil and gas exploration in this area.

Cell Consistency Evaluation Method Based on Multiple Unsupervised Learning Algorithms

Unsupervised learning algorithms can effectively solve sample imbalance.To address battery consistency anomalies in new energy vehicles,we adopt a variety of unsupervised learning algorithms to evaluate and predict the battery consistency of three vehicles using charging fragment data from actual operating conditions.We extract battery-related features,such as the mean of maximum difference,standard deviation,and entropy of batteries and then apply principal component analysis to reduce the dimensionality and record the amount of preserved information.We then build models through a collection of unsupervised learning algorithms for the anomaly detection of cell consistency faults.We also determine whether unsupervised and supervised learning algorithms can address the battery consistency problem and document the parameter tuning process.In addition,we compare the prediction effectiveness of charging and discharging features modeled individually and in combination,determine the choice of charging and discharging features to be modeled in combination,and visualize the multidimensional data for fault detection.Experimental results show that the unsupervised learning algorithm is effective in visualizing and predicting vehicle core conformance faults,and can accurately predict faults in real time.The“distance+boxplot”algorithm shows the best performance with a prediction accuracy of 80%,a recall rate of 100%,and an F1 of 0.89.The proposed approach can be applied to monitor battery consistency faults in real time and reduce the possibility of disasters arising from consistency faults.

BACKGROUND Atherosclerosis(AS),a chronic inflammatory disease of blood vessels,is a major contributor to cardiovascular disease.Dental pulp stem cells(DPSCs)are capable of exerting immunomodulatory and anti-inflammatory effects by secreting cytokines and exosomes and are widely used to treat autoimmune and inflam-mation-related diseases.Hepatocyte growth factor(HGF)is a pleiotropic cytokine that plays a key role in many inflammatory and autoimmune diseases.AIM To modify DPSCs with HGF(DPSC-HGF)and evaluate the therapeutic effect of DPSC-HGF on AS using an apolipoprotein E-knockout(ApoE-/-)mouse model and an in vitro cellular model.METHODS ApoE-/-mice were fed with a high-fat diet(HFD)for 12 wk and injected with DPSC-HGF or Ad-Null modified DPSCs(DPSC-Null)through tail vein at weeks 4,7,and 11,respectively,and the therapeutic efficacy and mechanisms were analyzed by histopathology,flow cytometry,lipid and glucose measurements,real-time reverse transcription polymerase chain reaction(RT-PCR),and enzyme-linked immunosorbent assay at the different time points of the experiment.An in vitro inflammatory cell model was established by using RAW264.7 cells and human aortic endothelial cells(HAOECs),and indirect co-cultured with supernatant of DPSC-Null(DPSC-Null-CM)or DPSC-HGF-CM,and the effect and mechanisms were analyzed by flow cytometry,RT-PCR and western blot.Nuclear factor-κB(NF-κB)activators and inhibitors were also used to validate the related signaling pathways.RESULTS DPSC-Null and DPSC-HGF treatments decreased the area of atherosclerotic plaques and reduced the expression of inflammatory factors,and the percentage of macrophages in the aorta,and DPSC-HGF treatment had more pronounced effects.DPSCs treatment had no effect on serum lipoprotein levels.The FACS results showed that DPSCs treatment reduced the percentages of monocytes,neutrophils,and M1 macrophages in the peripheral blood and spleen.DPSC-Null-CM and DPSC-HGF-CM reduced adhesion molecule expression in tumor necrosis factor-αstimulated HAOECs and regul

以铜绿微囊藻和三氯酚(TCP)为研究对象,探究了紫外/过硫酸盐(UV/PS)预氧化强化混凝技术同步除藻和TCP的效果。结果表明,经过UV/PS预氧化强化混凝沉淀以后,对OD680、Chl-a、浊度和TCP的去除率分别提高了66.2%、67.4%、51.7%和95.6%;UV/PS预氧化阶段最佳PS投加量为100 mg/L,最佳预氧化时间为10 min。机理分析发现,UV/PS预氧化可以破坏细胞的完整性,使藻细胞释放胞内有机物(IOM),并且能够进一步破坏核酸;同时,UV/PS预氧化还可以降低藻细胞表面的Zeta电位、改变细胞粒径,进而强化混凝除藻。
