Identification and validation of novel prognostic fatty acid metabolic gene signatures in colon adenocarcinoma through systematic approaches

Colorectal cancer(CRC)belongs to the class of significantly malignant tumors found in ***,dysregulated fatty acid metabolism(FAM)has been a topic of attention due to its modulation in cancer,specifically ***,the regulatory FAM pathways in CRC require comprehensive ***:The clinical and gene expression data of 175 fatty acid metabolic genes(FAMGs)linked with colon adenocarcinoma(COAD)and normal cornerstone genes were gathered through The Cancer Genome Atlas(TCGA)-COAD corroborating with the Molecular Signature Database v7.2(MSigDB).Initially,crucial prognostic genes were selected by uni-and multi-variate Cox proportional regression analyses;then,depending upon these identified signature genes and clinical variables,a nomogram was ***,to assess tumor immune characteristics,concomitant evaluation of tumor immune evasion/risk scoring were ***:A 8-gene signature,including ACBD4,ACOX1,CD36,CPT2,ELOVL3,ELOVL6,ENO3,and SUCLG2,was generated,and depending upon this,CRC patients were categorized within high-risk(H-R)and low-risk(L-R)***,risk and age-based nomograms indicated moderate discrimination and good *** data confirmed that the 8-gene model efficiently predicted CRC patients’***,according to the conjoint analysis of tumor immune evasion and the risk scorings,the H-R cohort had an immunosuppressive tumor microenvironment,which caused a substandard ***:This investigation established a FAMGs-based prognostic model with substantially high predictive value,providing the possibility for improved individualized treatment for CRC individuals.

Agricultural Vulnerability to Drought in China's Agro-pastoral Ecotone:A Case Study of Yulin City,Shaanxi Province

Agro-pastoral ecotone of northern China is the prominent area for agricultural production,but it is also the most typical ecological fragile area with frequent drought *** Yulin City at Shaanxi Province in China as the case area,the paper aims to investigate the spatio-temporal changes of agricultural vulnerability to drought in China’s agro-pastoral ecotone in the period 2000 *** results show that:1)the agricultural vulnerability to drought in Yulin City has shifted from high vulnerability in the period2000–2010 to low vulnerability in the period 2011–2020.2)There exist obvious spatio-temporal differences of the agricultural vulnerability to drought in Yulin City during the research period.3)Four sensitive events and 14 resilient events were identified in the research and the crops of Yulin had become more resilient to ***,the paper put forward with policy implications to make adaptive strategies of agriculture to climate change in China’s agro-pastoral ecotone in the future,e.g.,carrying out agricultural zoning based on agricultural production conditions,intensifying the construction of disaster prevention and relief system,and integrating with modern agricultural technology to develop new type agriculture.

痤疮是一种慢性炎症性皮肤表现,其影响因素复杂多样。《已使用化妆品原料目录(2021年版)》规定了祛痘植物原料在化妆品领域的可使用范围。基于此,笔者从痤疮发生机制入手,详细阐述痤疮相关生物学变化;在调研文献资料的基础上,对未收录其中的10种祛痘功效植物成分的作用机制进行了更新和综述;最后,对2002年至2022年已授权祛痘相关专利进行分析,探索祛痘产品的研究现状和发展趋势,为祛痘类产品的研发提供新思路。

目的研究安罗替尼联合培美曲塞对晚期非鳞非小细胞肺癌JAK/STAT3信号传导通路的影响。方法选取82例晚期非鳞非小细胞肺癌患者为此次试验对象,随机分成2组。对照组(41例)术后接受培美曲塞治疗,实验组(41例)在此基础上联合安罗替尼治疗。对比两组患者治疗前后Janus激酶(JAK)、转录激活因子3(STAT3)表达情况;对比两组患者临床疗效和化疗不良反应。结果治疗后两组患者JAK、STAT3表达均明显降低,且相比于对照组,实验组显著更低(P0.05)。结论对于晚期非鳞非小细胞肺癌患者,在培美曲塞基础上联合安罗替尼治疗,可有效抑制JAK/STAT3信号传导通路活性,临床疗效较高,并且不会增加不良反应的发生。

胃肠间质瘤(gastrointestinal stromal tumor,GIST)是消化道最常见的间叶源性肿瘤。随着伊马替尼的应用,晚期GIST的疗效得到了显著改善。尽管多数患者接受伊马替尼治疗后获益,但仍有部分患者会发生继发性耐药。本例GIST患者接受伊马替尼治疗15个月后出现了耐药及肿瘤进展,镜下示部分肿瘤细胞出现明显的多形性,免疫组织化学染色示伴横纹肌细胞分化。在进展的病灶中未发现c-Kit、PDGFRA基因的继发突变。由于GIST横纹肌肉瘤去分化罕见,本病例的报道将有助于伊马替尼治疗后GIST的诊断,避免误诊及漏诊。

Advances in pharmacological effects of tanshinone ⅡA on pain

Pain is one of the most serious problems plaguing human health today. Drug therapy is one of the main ways to treat pain in clinic. The analgesic drugs commonly used in clinical treatment of pain are often accompanied by many side effects, the analgesic effect is still not ideal. Salvia miltiorrhiza is a traditional medicinal material with the same origin as food and medicine. It has the functions of promoting blood circulation and removing blood stasis, relieving pain through menstrual circulation, and contains many effective ingredients such as tanshinone and salvianolic acid. Tanshinone is a kind of rosin diterpenoid compound, which mainly consists of o-quinone type and p-quinone type parent nucleus, and tanshinone Ⅱ A is the representative compound. The pharmacological mechanism of tanshinone Ⅱ A in labor pain mainly includes:(1) Regulate inflammatory *** cytokines played an important role in the occurrence and progression of pain. It was found that the analgesic effect of tanshinone ⅡA was related to the antiinflammatory effect. Tanshinone Ⅱ A showed anti-injurious activity in various pain models, such as bone cancer pain and sciatic nerve ligation, and related studies found that tanshinone Ⅱ A could inhibit the expression of inflammatory factors TNF-α, IL-1β and IL-6 in the spinal cord of model rats. In the spinal nerve ligation model,tanshinone ⅡA also promoted the release of anti-inflammatory cytokine IL-10 in the spinal cord of rats.(2) Regulate signal pathways related to regulating spinal cord oxidation and apoptosis. Apoptosis and oxidation played an important role in the process of pain. When nerve injury was caused by stimulation, oxidative stress and apoptosis of nerve cells were involved in the mechanism of hyperalgesia. Tanshinone Ⅱ A sodium sulfonate could relieve pain by regulating apoptosis-related pathways. In neuralgia model, tanshinone ⅡA could reduce the apoptosis of spinal cord neurons by inhibiting oxidative stress response

Amino-functionalized UiO-66-doped mixed matrix membranes with high permeation performance and fouling resistance

For the reduction of bovine serum proteins from wastewater,a novel mixed matrix membrane was prepared by functionalizing the substrate material polyaryletherketone(PAEK),followed by carboxyl groups(C-SPAEKS),and then adding amino-functionalized UiO-66-NH_(2)(Am-UiO-66-NH_(2)).Aminofunctionalization of UiO-66 was accomplished by melamine,followed by an amidation reaction to immobilize Am-UiO-66-NH_(2),which was immobilized on the surface of the membrane as well as in the pore channels,which enhanced the hydrophilicity of the membrane surface while increasing the negative potential of the membrane *** nanoparticle-loaded ultrafiltration membrane has good permeation performance,with a pure water flux of up to 482.3 L·m^(-2)·h^(-1) for C-SPAEKS/AmUiO-66-NH_(2) and a retention rate of up to 98.7%for bovine serum albumin(BSA)-contaminated ***,after several hydrophilic modifications,the flux recovery of BSA contaminants by this series of membranes increased from 56.2%to 80.55%of pure *** results of ultra-filtration flux time tests performed at room temperature showed that the series of ultrafiltration membranes remained relatively stable over a test time of 300 ***,the newly developed mixed matrix membrane showed potential for high efficiency and stability in wastewater treatment containing bovine serum proteins.

Spectral purification improves monitoring accuracy of the comprehensive growth evaluation index for film-mulched winter wheat

In order to further improve the utility of unmanned aerial vehicle(UAV)remote-sensing for quickly and accurately monitoring the growth of winter wheat under film mulching, this study examined the treatments of ridge mulching,ridge–furrow full mulching, and flat cropping full mulching in winter *** on the fuzzy comprehensive evaluation (FCE) method, four agronomic parameters (leaf area index, above-ground biomass, plant height, and leaf chlorophyll content) were used to calculate the comprehensive growth evaluation index (CGEI) of the winter wheat, and 14 visible and near-infrared spectral indices were calculated using spectral purification technology to process the remote-sensing image data of winter wheat obtained by multispectral *** machine learning algorithms, partial least squares, support vector machines, random forests, and artificial neural network networks(ANN), were used to build the winter wheat growth monitoring model under film mulching, and accuracy evaluation and mapping of the spatial and temporal distribution of winter wheat growth status were carried *** results showed that the CGEI of winter wheat under film mulching constructed using the FCE method could objectively and comprehensively evaluate the crop growth *** accuracy of remote-sensing inversion of the CGEI based on the ANN model was higher than for the individual agronomic parameters, with a coefficient of determination of 0.75,a root mean square error of 8.40, and a mean absolute value error of *** purification could eliminate the interference of background effects caused by mulching and soil, effectively improving the accuracy of the remotesensing inversion of winter wheat under film mulching, with the best inversion effect achieved on the ridge–furrow full mulching area after spectral *** results of this study provide a theoretical reference for the use of UAV remote-sensing to monitor the growth status of winter wheat with film mulching.

Gapless Genome Assembly of ZH8015 and Preliminary Multi-Omics Analysis to Investigate ZH8015's Responses Against Brown Planthopper Infestation

Accurate genomic information is essential for advancing genetic breeding research in specific rice *** study presented a gapless genome assembly of the indica rice cultivar Zhonghui 8015(ZH8015)using Pac Bio HiFi,Hi-C,and ONT(Oxford Nanopore Technologies)ultra-long sequencing technologies,annotating 43037 gene ***,utilizing this genome along with transcriptomic and metabolomic techniques,we explored ZH8015's response to brown planthopper(BPH)*** transcriptomic sampling indicated significant changes in gene expression levels around 48 h after BPH *** analysis revealed particularly significant alterations in genes related to reactive oxygen species scavenging and cell wall *** results demonstrated marked increases in levels of several monosaccharides,which are components of the cell wall and dramatic changes in flavonoid *** association analysis identified differentially expressed genes associated with key metabolites,shedding light on ZH8015's response to BPH *** summary,this study constructed a reliable genome sequence resource for ZH8015,and the preliminary multi-omics results will guide future insect-resistant breeding research.

Achieving high fouling resistance and permeability using membrane separation technology in water treatment processes remains a *** this work,a novel mixed-matrix membrane(MMM)(poly(arylene ether ketone)[PAEK]-containing carboxyl groups[PAEK-COOH]/UiO-66-NH_(2)@graphene oxide[GO])with superb fouling resistance and high permeability was prepared by the nonsolvent-induced phase separation method,by in-situ growth of UiO-66-NH_(2) on the GO layer,and by preparing hydrophilic *** the basis of the structure and performance analysis of the MMM,the maximum water flux reached 591.25 L·m^(-2)·h^(-1) for PAEK-COOH/UiO-66-NH_(2)@GO,whereas the retention rate for bovine serum albumin increased from 85.40%to 94.87%.As the loading gradually increased,the hydrophilicity of the MMMs increased,significantly enhancing their fouling *** strongest anti-fouling ability observed was 94.74%,which was 2.02 times greater than that of the pure *** the same time,the MMMs contained internal amide and hydrogen bonds during the preparation process,forming a cross-linked structure,which further enhanced the mechanical strength and chemical *** summary,the MMMs with high retention rate,strong permeability,and anti-fouling ability were successfully prepared.
