Bone-conditioned medium contributes to initiation and progression of osteogenesis by exhibiting synergistic TGF-β1/BMP-2 activity

Guided bone regeneration (GBR) often utilizes a combination of autologous bone grafts, deproteinized bovine bone mineral(DBBM), and collagen membranes. DBBM and collagen membranes pre-coated with bone-conditioned medium (BCM) extracted from locally harvested autologous bone chips have shown great regenerative potential in GBR. However, the underlying molecular mechanism remains largely unknown. Here, we investigated the composition of BCM and its activity on the osteogenic potential of mesenchymal stromal cells. We detected a fast and significant (P <0.001) release of transforming growth factor-β1 (TGF-β1) from autologous bone within 10 min versus a delayed bone morphogenetic protein-2 (BMP-2) release from 40 min onwards. BCMs harvested within short time periods (10, 20, or 40 min), corresponding to the time of a typical surgical procedure, significantly increased the proliferative activity and collagen matrix production of BCM-treated cells. Long-term (1, 3, or 6 days)-extracted BCMs promoted the later stages of osteoblast differentiation and maturation. Short-term-extracted BCMs, in which TGF-β1 but no BMP-2was detected, reduced the expression of the late differentiation marker osteocalcin. However, when both growth factors were present simultaneously in the BCM, no inhibitory effects on osteoblast differentiation were observed, suggesting a synergistic TGF-β1/BMP-2 activity. Consequently, in cells that were co-stimulated with recombinant TGF-β1 and BMP-2, we showed a significant stimulatory and dose-dependent effect of TGF-β1 on BMP-2-induced osteoblast differentiation due to prolonged BMP signaling and reduced expression of the BMP-2 antagonist noggin. Altogether, our data provide new insights into the molecular mechanisms underlying the favorable outcome from GBR procedures using BCM, derived from autologous bone grafts.

Sub-chronic effects of a Phthirusa pyrifolia aqueous extract on reproductive function and comparative hormone levels in male rats

Objective: To explore the effects of aqueous extract from Phthirusa pyrifolia leaves(67 mg/kg body weight for 12 days) on the reproductive function of male Wistar rats through oral ***: Animals(n = 30), aged 13 weeks and weighing(378.5 ± 5.0) g, were housed in a vivarium under controlled environmental conditions [photoperiod of 12 h light/dark,temperature of(23 ± 1)C] and were fed standard rations ad libitum. The experiment ran for 12 days, wherein animals were divided into three groups: negative control(n = 6)received water, positive control(n = 12) with finasteride at a concentration of 1.0 mg/kg;and a test group(n = 12) submitted to aqueous extract. At the end of the experiment, the animals were sacrificed and submitted to ***: The morphological results of the testes showed that the aqueous extract induced significant changes in the diameter and cross-sectional area of the seminiferous tubules as well as the thickness of the seminiferous epithelium. Furthermore, the extract was able to abruptly decrease testosterone concentrations by about 81.88% in the treated group when compared with the negative control,(47.0 ± 4.8) ng/d L and(255.0 ± 2.0) ng/d L,respectively, and 76.8%,(211.0 ± 8.7) ng/d L, when compared with finasteride. However,the extract causes neither liver damage nor impairment of renal ***: These results suggest that the high amounts of flavonoids shown to be in the extract may be responsible for its hepato-protective effects and suggest a possible decrease in the libido and reproduction of rats.

本系列病例的目的,是将一种新型可吸收胶原基质(collagen matrix,CMX)应用于美学区种植同期行引导骨再生(guided bone ***)术中.评估其在8周愈合期内增强组织厚度的安全性和可行性。使用超声波装置测定种植位点和邻牙的软组织厚度。通过连续数字表面模型叠加牙周参数评估整体组织轮廓变化。牙周参数和患者相关结果显示无显著变化。GBR联合新型CMX使得植入部位的软组织厚度在8周后显著增加156mm.且从4周到8周软组织没有显著减少。整体组织轮廓在距粘膜边缘5mm处增加最为显著.与种植体肩部区域的组织增加相对应。8周后邻牙未见受到影响。

Oomycete,a group of filamentous plant pathogens,is responsible for most notorious crop diseases such as potato late blight that threaten the global food *** has been known that many small cysteine rich（SCR） effectorgenes are highly inducedduring oomycete infection,however,the molecular mode of actions of SCR effectors remain largely unknown.A preliminary bioinformatics analysis combined with high-throughputin planta screening assay resulted in the identification of a Phytophthora infestans cysteine rich effector SCR2,which could trigger significant reactive oxygen burst,defence related gene induction and hypersensitive response in Nicotiana *** many other SCR effectors,SCR2 orthologs are wildly presented in oomycete plant pathogens,including species from Phytophthora,Hyaloperonospora,Pythium and *** induced gene silencing（VIGS） found that SCR2 induced host immunity required immune hub regulator BAK1 and akinase MAPKKKξ.Furthermore,chemical inhibition assay suggested that Serine proteases are also involved in SCR2 *** summary,we identified a novel SCR effector with priming host immunity *** biochemical functions of SCR2 and the mechanisms of recognition deserve further investigations.

大洋底部多数为沉积物所覆盖,因此沉积物就构成了地球空间覆盖中最大的一个单一生态系统。虽然生活在沉积物表面和内部的微型、小型和大型海洋有机体中只有一小部分被描述过,而且很少有人试图对物种总数和生物地理分布进行统计,但对若干物种的比较全面的了解足以说明底栖有机体对生态系统主要过程有显著的影响。底栖有机体参与碳、氮、硫的循环,水体的演变,污染物的传播、积累、二次污染和迁移,以及沉积物的稳定性等过程。虽然对群体之间的联系和功能上的冗余水平知之甚少,但在群体内部还是可能存在大量冗余的。说明生物多样性对于海底系统参与生态系统功能来说是必要的证据很少,但鉴于它们之间联系了解很少,预测性知识也仅限于几个物种,所以目前还无法准确推断物种的减少如何影响这些功能和生态系统的健康。因此,"做最坏的打算"作为一种保险的方法是可行的,而且要尽量避免由于捕渔、污染、栖息地的破坏,非本地(外来)物种的引入和全球变暖所造成的物种和基因多样性的减少。同时,科学家们应充分利用当前蓬勃发展的科学技术和对生物多样性的高昂的兴趣,以提供关于底栖生物和生态系统过程的具体而全面的信息。
