High-throughput metabarcoding of SAR11 assemblages from the southwest Atlantic shelf and arid Patagonia:richness and associated rank abundance distributions

Background:Massively parallel sequencing of environmental DNA allows microbiological studies to be performed in greater detail than was possible with first-generation *** example,it facilitates the use of approaches hitherto largely applied to flora and fauna,such as rank abundance distribution(RAD)***:Here,we set out to advance the knowledge on ***(SAR11)communities from southern South America using environmental sequences from the open ocean in the Argentine sea,the uncharted Engaño Bay,as well as a river and an oligohaline shallow lake from the Patagonian Steppe *** structures of the SAR11 assemblages present in these ecosystems were dissected by direct and rarefaction-based estimates of species richness,and evaluations of the corresponding abundance distributions(ADs),which was addressed by RAD ***:Microbial community composition analyses revealed that the studied SAR11 assemblages coexist with 27 bacterial ***11 richness was in general very high,but ADs turned out to be highly *** results were compatible with prior knowledge,and similar to that derived from point estimates of ***,our comprehensive dissection allowed for more detailed quantitative comparisons to be made between the environments surveyed,and revealed differences regarding both richness and the underlying ***:Despite SAR11 assemblages being extremely rich,their ADs are very *** and ADs can vary,not only between fresh and salt water,but also between oceanic and coastal marine *** obtained results provide insights on general topics such as adaptation and the contrast between marine and freshwater radiations.

Detailed bed topography and sediment load measurements for two stepdown flows in a laboratory flume

Streams and rivers, particularly smaller ones, often do not maintain steady flow rates for long enough to reach equilibrium conditions for sediment transport and bed topography. In particular, streams in small watersheds may be subject to rapidly changing hydrographs, and relict bedforms from previous high flows can cause further disequilibrium that complicates the prediction of sediment transport rates. In order to advance the understanding of how bedforms respond to rapid changes in flow rate, a series of flume experiments were performed where the flow was reduced rapidly from equilibrium *** transport rates and bed elevation data across the flume and over a 15-meter-long test section were collected during the experiments to allow detailed examination of evolving bedform dynamics. It was found that relict bedforms stopped moving completely after flow reductions, and the mode of sediment transport was shifted to small bedforms that arose rapidly over dune stoss sections throughout the test section. The changes in sediment transport with time as the sand bed adjusted to the new flow rate was found to agree with predictions based on the relations proposed in Wren et al.(2020). Wavelet analysis is used to visualize changes in length and amplitude scales during the bed transition process.

Impacts of timing,length,and intensity of behavioral interventions to COVID-19 dynamics:North Carolina countylevel examples

We sought to examine how the impact of revocable behavioral interventions,e.g.,shelterin-place,varies throughout an epidemic,as well as the role that the proportion of susceptible individuals had on an intervention's *** estimated the theoretical impacts of start day,length,and intensity of interventions on disease transmission and illustrated them on COVID-19 dynamics inWake County,North Carolina,to inform how interventions can be most *** used a Susceptible,Exposed,Infectious,and Recovered(SEIR)model to estimate epidemic curves with modifications to the disease transmission parameter(β).We designed modifications to simulate events likely to increase transmission(e.g.,long weekends,holiday seasons)or behavioral interventions likely to decrease it(e.g.,shelter-in-place,masking).We compared the resultant curves'shape,timing,and cumulative case count to baseline and across other modified *** led to changes in COVID-19 dynamics,including moving the peak's location,height,and *** proportion susceptible,at the start day,strongly influenced their *** interventions shifted the curve,while interventions near the peak modified shape and case *** some scenarios,in which the transmission parameter was decreased,the final cumulative count increased over *** showed that the timing of revocable interventions has a strong impact on their *** same intervention applied at different time points,corresponding to different proportions of susceptibility,resulted in qualitatively differential *** estimation of the proportion susceptible is critical for understanding an intervention's *** findings presented here provide evidence of the importance of estimating the proportion of the population that is susceptible when predicting the impact of behavioral infection control *** emphasis should be placed on the estimation of this epidemic component in intervention design and decision-makin

Hemolysis of PM_(10) on RBCs in vitro:An indoor air study in a coal-burning lung cancer epidemic area

Epidemiological studies have suggested that inhalation exposure to indoor ambient air from coal-burning environments is causally associated with respiratory health *** order to explore the toxicological mechanisms behind the adverse health effects,the hemolytic activity of PM_(10)(particulate matter with an aerodynamic diameter of 10 um or less)samples collected from homes burning coal in the recognized China"cancer village"Xuanwei were evaluated and matched against their trace elemental *** results demonstrated that the hemolytic activity of indoor PM_(10) in coal-burning environments ranged from 4.28%to 5.24%,with a clear positive dose-response *** low dose samples exhibited a reduced hemolytic activity,PM_(10) could have a toxic effect upon people in a coal-burning indoor environment for extended time *** concentrations of analyzed trace elements in PM_(10) samples ranged from 6966 to 12,958 *** the analyzed elements,Zn,Ti,Ni,Cu,Pb,Ba,Mn,Cr and V were found at higher concentrations and accounted for over 95%of the total *** concentrations of total analyzed elements in the PM_(10) samples revealed a significant positive correlation with PM_(10) hemolytic *** the analyzed elements,Zn,Pb and Cs positively correlated with hemolysis,while Li,U and V negatively correlated with the hemolysis of human red blood cells(RBCs).Therefore,the heavy metal elements could be one of the main factors responsible for the hemolytic capacity of indoor PM_(10) in coal-burning environments.

Antigen unmasking does not improve the visualization of phospholipase C zeta in human spermatozoa

Phospholipase C zeta(PLCζ)is a sperm-specific protein that triggers oocyte *** analysis of PLCζexpression in human spermatozoa can be used as a diagnostic marker for oocyte activation *** laboratory has previously optimized a standard"in-house"assay to determine PLCζexpression in human ***,one study has suggested that an antigen unmasking method(AUM)would be more efficient in visualizing PLCζin human *** study aimed to compare our established assay and AUM(involving HCl,acidic Tyrode's solution[AT],and heat).The mean relative fluorescence(RF)intensity of PLCζin frozen-thawed spermatozoa from fourteen fertile donors stained with the in-house method was significantly higher than three other AUM groups(in-house[mean±standard error of mean]:18.87±2.39 arbitrary units[a.u.]vs non-AUM:11.44±1.61 a.u.,AT-AUM:12.38±1.89 a.u.,and HCl-AUM:12.51±2.16 a.u.,P<0.05,one-way analysis of variance).The mean RF intensity of PLCζin AT-and HCl-treated spermatozoa from 12 infertile males was not significantly different from that of the non-AUM ***,the in-house method resulted in the highest RF intensity(12.11±1.36 a.u.,P<0.01).Furthermore,specificity testing of antibody-antigen binding indicated that the in-house method showed more specific binding than spermatozoa treated by the *** conclusion,our in-house method showed superior visualization and reliability than the AUM,thus supporting the continued use of our in-house assay for clinical research screening.

Social relationship satisfaction and accumulation of chronic conditions and multimorbidity:a national cohort of Australian women

Intensities of human disturbance dictate the floral diversity in tropical forest:the case of a secondary forest in Benguet, Philippines

This study inventoried the floral diversity of a secondary forest in Tuba,Benguet,Philippines as affected by different intensities of human *** study is unique from other land use studies in Southeast Asia in having conventional farms as the most disturbed land use while other sites have oil palm plantations.A total of 267 species belonging to 222 genera and 78 families was *** showed that the level of human disturbance is the major factor affecting the floral diversity while the effect of elevation is *** level of human disturbance is significantly associated with species richness,species composition,dominant species and biodiversity *** human disturbance,as in the case of conventional farms,results in lower species richness,predominantly weed composition,dominant weed species and lower biodiversity *** the other hand,lesser human disturbance(as in the case of secondary forest)leads to higher species richness,greater native species composition,dominance of indigenous species,and higher diversity *** most disturbed land use experienced the greatest loss of species richness,change in floristic composition and vegetation structure,as well as a shift from native to alien-dominated plant *** results showed that the effect of land use change is not limited to species richness but also to species composition,dominant species and biodiversity ***,the study is an important baseline data for biodiversity conservation of the Marcos Highway Watershed Forest Reserve.

Influence of biological sex and exercise on murine cardiac metabolism

Although the structural and functional effects of exercise on the heart are well established,the metabolic changes that occur in the heart during and after exercise remain *** this study,we used metabolomics to assess time-dependent changes in the murine cardiac metabolome following 1 session of treadmill *** the exercise bout,we also recorded blood lactate,glucose,and ketone body levels and measured cardiac mitochondrial *** both male and female mice,moderate-and high-intensity exercise acutely increased blood lactate *** both sexes,low-and moderate-intensity exercise augmented circulating 3-hydroxybutryrate levels immediately after the exercise bout;however,only in female mice did high-intensity exercise increase 3-hydroxybutyrate levels,with significant increases occurring 1 h after the exercise *** metabolomics analyses of sedentary female and male hearts suggest considerable sex-dependent differences in basal cardiac metabolite levels,with female hearts characterized by higher levels of pantothenate,pyridoxamine,homoarginine,tryptophan,and several glycerophospholipid and sphingomyelin species and lower levels of numerous metabolites,including acetyl coenzyme A,glucuronate,gulonate,hydroxyproline,prolyl-hydroxyproline,carnosine,anserine,and carnitinylated and glycinated species,as compared with male *** after a bout of treadmill exercise,both male and female hearts had higher levels of corticosterone;however,female mice showed more extensive exercise-induced changes in the cardiac metabolome,characterized by significant,time-dependent changes in amino acids(e.g.,serine,alanine,tyrosine,tryptophan,branched-chain amino acids)and the ketone body *** from experiments using isolated cardiac mitochondria suggest that high-intensity treadmill exercise does not acutely affect respiration or mitochondrial coupling;however,female cardiac mitochondria demonstrate generally higher adenosi
