Differential mucin phenotypes and their significance in a variation of colorectal carcinoma

AIM: To investigate mucin expression profiles in colorectal carcinoma (CRC) histological subtypes with regard to clinicopathologic variables and prognosis. METHODS: Mucin (MUC)2 and MUC5AC expressions were assessed by immunohistochemistry for a total of 250 CRC cases that underwent surgical resection. CRCs included 63 well-to-moderately differentiated adenocar-cinomas (WMDAs), 91 poorly differentiated adenocarcinomas (PDAs), 81 mucinous adenocarcinoma (MUAs), and 15 signet-ring cell carcinomas (SRCCs). MUC2 and MUC5AC were scored as positive when ≥ 25% and ≥ 1% of cancer cells were stained positive, respectively. The human mutL homolog 1 and human mutS homolog 2 expressions were assessed by immunohistochemistry in PDAs to investigate mismatch-repair (MMR) *** that did not express either of these two were considered MMR-deficient. Results were analyzed for associations with clinicopathologic variables and the prognosis in individual histological CRC subtypes. RESULTS: MUC2-positive and MUC5AC-positive WMDA percentages were 49.2% and 30.2%, respectively. In contrast, MUC2-positive and MUC5AC-positive PDA percentages were 9.5% and 51.6%, respectively. MUC2 levels tended to decrease and MUC5AC levels tended to increase from WMDA to PDA. In 21 tumors comprising both adenoma and adenocarcinoma components in a single tumor (4 WMDAs, 7 PDAs, and 10 MUAs), MUC2 was significantly downregulated in PDA and MUC5AC was downregulated in PDA and MUA in the adenoma-carcinoma sequence. These results suggested that MUC2 levels might be associated with malignant potential and that MUC5AC expression was an early event in tumorigenesis. Despite worse prognoses than WMDA, high MUC2 expression levels were maintained in MUA (95.1%) and SRCC (71.5%), which suggested a pathogenesis for these subtypes distinct from that of WMDA. 本文是利用崇祯年间刊行的《盟水斋存牍》来考察明末珠江三角洲社会的一环。明末的乡绅在包括广州城和老家的广大范围内,扩张以宗祠中心的宗族网络,参与了沙田经营,海外贸易等种种利权。聚集在乡绅周围的,不仅仅族人,受到乡绅权威和权利的吸引,奴仆、无赖之类猬集而至。乡绅的奴仆和无赖,在围绕沙田利权的纷争中也是实战部队。在明末的三角洲地带,宗族之间的竞争变得激烈。拥有乡绅的话,其宗族(宦族)就可以以宗祠为中心强化向心力,不仅仅是族人,也会吸引来奴仆、无赖等各种各样的人,多方面开展利权争夺。无法辈出乡绅,就意味着无法强固地维持宗族体制,于是在利权争夺的竞争中败北。产生宦族和弱小宗族之间差距的理由就在于此。

In the mammalian brain, four neurotrophins have been identified： nerve growth factor （NGF）, brain-derived neurotrophic factor （BDNF）, neurotrophin-3 （NT-3） and neurotrophin-4/5 （NT-4/5）. NGF exerts an important role in the development and functions of the central and peripheral nervous system. However, it has recently been documented that several types of immune cells, such as mast ceils, iymphocytes, basophils and eosinophils, produce, store and release NGF. Accumulating preclinical and clinical data indicate that dysfunctions of NGF and the other neurotrophins may contribute to impaired immune responses and concentration of NGF frequently correlates with disease severity. Thus, the aim of this study was to elucidate the potential signaling mechanisms of cytokineneurotrophins interactions contributing to increased NGF levels. Our data show that the transcription factor NF-κB plays a pivotal role in regulating B-cell-derived NGF expression.

Synthesis,structure and mechanical properties of Zr-Cu-based bulk metallic glass composites

The unusual glass-forming ability（GFA） of the Zr48Cu36Ag8Al8 alloy and the high ductility of the Zr48Cu36Ag8Al8 metallic glass-matrix composites containing Ta powder were *** bulk metallic glass rod with a diameter of 25 mm was successfully synthesized using copper mold casting for the Zr48Cu36Ag8Al8 *** GFA of this alloy was found to be related to a large supercooled liquid region and a quaternary eutectic point with low melting *** bulk metallic glass matrix composites were prepared by introducing extra Ta particles into the Zr48Cu36Ag8Al8 *** composites consist of Ta particles homogenously distributed in the Zr48Cu36Ag8Al8 metallic glass *** optimum content of Ta powder is 10at%for the composite with the highest plasticity,which shows a plastic strain of 31%.

Stress Dependence of Transformation Plastic Behavior

Transformation plasticity is known to play an important role in the course of heat treatment processes, and so affect the results of heat treatment simulations, which means that the transformation plasticity coefficient is necessary to be identifies. The authors developed a new method by use of four-point bending system of a beam to identify transformation plasticity coefficient taking advantage of its easiness and high accuracy compared with other conventional methods like tension test, etc., and identified the coefficient for four kinds of steels; plane carbon steel, Cr-steel, Cr-Mo steel and bearing steel. In this paper, further experimental data are presented for the steels if the coefficient Kp during pearlite transformation depend on the applied stress. Obtained results reveals that the stress dependence of Kp is rather trivial except for uncertain tendency detected in plane carbon steel. The relation between the Kp and carbon content included is discussed in comparison with other data referred from references.

Long-term prognostic impact of circulating tumour cells in gastric cancer patients

AIM To analyse the long-term prognostic impact of circulating tumour cells(CTCs) in gastric cancer patients who underwent surgery. METHODS A 7.5-m L peripheral vein blood sample was obtained from each patient with treatment-negative gastric adenocarcinoma before surgery. OBP-401, a telomerasespecific, replication-selective, oncolytic adenoviral agent carrying the green fluorescent protein gene, was used to label CTCs. Correlations between the number of CTCs and clinical end points were evaluated. RESULTS The median follow-up period of the surviving patients with gastric cancer was 60 mo. The CTC number tended to increase concomitantly with disease progression. The overall survival of patients with more than five CTCs in 7.5-m L of peripheral blood was lower than that of patients with five or less CTCs, although the difference was not significant(P = 0.183). A significant difference in relapse-free survival was found between patients with more than five and those with five or less CTCs(P = 0.034).CONCLUSION A lower number of CTCs was correlated with higher relapse-free survival rates in patients. Detection of CTCs using OBP-401 may be useful for predicting prognosis in gastric cancer.

Development of PC-based Electronic Teaching and Learning Materials for De- scriptive Geometry

Descriptive geometry is very important and recognized as a basic skill and knowledge for mechanical engineering student. In this study, PC-based electronic teaching/learning materials for descriptive geometry are created using Flash, which is a typical animation creator. Furthermore, several axonometric representations, created by 3D-CAD, SolidWorks, for 3D objects are auxiliary materials to promote understanding of descriptive geometry. The axonometric representations in 3D-CAD are also dynamic, in other words, a viewpoint can be moved free. The movement of 3D model in a PC monitor can be recorded using a normal function of SolidWorks and replayed by typical animation software. The developed materials are excellent at accuracy of drawing, repeatability of self-study and visual attraction in comparison to oral presentation using still image and inaccurate drawing on a textbook or blackboard in a classroom. Actually, questionnaire survey results present favorable impressions from student-users, although they point out the further improvement in the replaying speed. The replaying speed can be controlled easily by using a normal function of Flash. In addition, usual playback software for animation has functions of pause and replay on demand and, thus, it is not contro-versial.

A specific gene-expression signature quantifies the degree of hepatic fibrosis in patients with chronic liver disease

AIM： To study a more accurate quantification of hepatic fibrosis which would provide dinically useful information for monitoring the progression of chronic liver disease. METHODS： Using a cDNA microarray containing over 22000 clones, we analyzed the gene-expression profiles of non-cancerous liver in 74 patients who underwent hepatic resection. We calculated the ratio of azanstained： total area, and determined the morphologic fibrosis index （MFI）, as a mean of 9 section-images. We used the MFI as a reference standard to evaluate our method for assessing liver fibrosis. RESULTS： We identified 39 genes that collectively showed a good correlation （r 〉 0.50） between geneexpression and the severity of liver fibrosis. Many of the identified genes were involved in immune responses and cell signaling. To quantify the extent of liver fibrosis, we developed a new genetic fibrosis index （GFI） based on gene-expression profiling of 4 clones using a linear support vector regression analysis. This technique, based on a supervised learning analysis, correctly quantified the various degrees of fibrosis in both 74 training samples （r = 0.76, 2.2% vs 2.8%, P 〈 0.0001） and 12 independent additional test samples （r = 0.75, 9.8% vs 8.6%, P 〈 0.005）. It was far better in assessing liver fibrosis than blood markers such as prothrombin time （r = -0.53）, type IV collagen 7s （r = 0.48）, hyaluronic acid （r = 0.41）, and aspartate aminotransferase to platelets ratio index （APRI）（r = 0.38）. CONCLUSION： Our cDNA microarray-based strategy may help clinicians to precisely and objectively monitor the severity of liver fibrosis.

Potential significance of CX3CR1 dynamics in stress resilience against neuronal disorders

Recent findings have implicated inflammatory responses in the central nervous system in a variety of neuropsychiatric and neurodegenerative diseases,and the understanding and control of immunological responses could be a major factor of future therapeutic strategies for neurological ***,derived from myelogenous cells,respond to a number of stimuli and make immune responses,resulting in a prominent role as cells that act on inflammation in the central nervous ***(FKN or CX3CL1)signaling is an important factor that influences the inflammatory response of *** receptor for FKN,CX3CR1,is usually expressed in microglia in the brain,and therefore the inflammatory response of microglia is modified by ***,FKN often suppresses inflammatory responses in microglia and activation of its receptor may be effective in the treatment of inflammatory neurological ***,it has also been suggested that inflammatory responses facilitated by FKN signaling aggravate neurological ***,further studies are still required to resolve the conflicting interpretation of the protective or deleterious contribution of microglial FKN *** notably,regulation of FKN signaling has recently been shown to be beneficial in the treatment of human diseases,although not neurological *** addition,a CX3CR1 inhibitor has been developed and successfully tested in animal models,and it is expected to be in human clinical trials in the *** this review,I describe the potential therapeutic consideration of microglial CX3CR1 dynamics through altered FKN signaling.
