The nano era demands the synthesis of new nanostructured materials,if possible by simplified techniques,with remarkable properties and versatile ***,we demonstrate a new single-step reproducible melt-quench methodology to fabricate core-shell bimetallic(Au0 Ag0)nanoparticles(2889 nm)embedded glasses(dielectrics)by the use of a new reducing glass matrix,K_(2)O B_(2)O_(3) Sb_(2)O_(3)(KBS)without applying any external reducing agent or multiple processing *** surface plasmon resonance(SPR)band of these nanocomposites embedded in KBS glass is tunable in the range 554-681 *** remarkably,taking advantage of the selective reduction capability of Sb_(2)O_(3),this single-step methodology is used to fabricate inter-metallic:rare-earth ions co-embedded(AuAg:Sm^(3+))dielectric(glass)-based-dnanocomposites and study the effect of enhanced local fi eld on the red upconversion fl uorescence of Sm^(3+)ions at 636 *** enhancement is found to be about 2 *** single-step in-situ selective reduction approach can be used to fabricate a variety of hybrid-nanocomposite devices for laser based applications(see supplementary information).

Background： The Y chromosome in mammal is paternally inherited and harbors genes related to male fertility and spermatogenesis. The unique intra-chromosomal recombination pattern of Y chromosome and morphological difference of this chromosome between Bos taurus and Bos indicus make it an ideal model for studying structural variation, especially in crossbred （Bos taurus x Bos indicus） bulls. Copy Number Variation （CNV） is a type of genomic structural variation that gives information complementary to SNP data. The purpose of this study was to find out copy number differences of four Y chromosomal spermatogenesis-related candidate genes in genomic DNA of crossbred and purebred Indicine bulls. Result： Four Y chromosomal candidate genes of spermatogenesis namely, sex determining gene on Ychromosome （SRY）, DEAD box po/ypeptide 3-Y chromosome （DDX3 Y）, Ubiquidn specific peptidase 9, Y-linked （ usPgY）, testis-specific protein on Y chromosome （TSPY） were evaluated. Absolute copy numbers of Y chromosomal genes were determined by standard curve-based quantitative real time PCR. Copy numbers of SRYand TSPYgenes per unit amount of genomic DNA are higher in crossbred than Indicine bulls. However, no difference was observed in DDX3Yand usPgYgene copy numbers between two groups. Conclusion： The present study demonstrates that the structural organization of Y chromosomes differs between crossbred and Indicine bulls which are reproductively healthy as observed from analysis of semen attributes. The absolute copy numbers of SRY and TSPY genes in unit mass of genomic DNA of crossbred bulls are significantly higher than Indicine bulls. No alteration in absolute copies ofDDX3Yand usPgYgene was found between the genome of crossbred and Indicine bulls. This study suggests that the DDX3Yand USPgYare likely to be single copy genes in the genome of crossbred and Indicine bulls and variation in Y chromosome length between crossbred and Indicine bulls may be due to the copy number variation of

Effect of recombinant and native buffalo OVGP1 on sperm functions and in vitro embryo development: a comparative study

Background： An oviduct-specific glycoprotein, OVGP1, is synthesized and secreted by non-ciliated epithelial cells of the mammalian oviduct which provides an essential milieu for reproductive functions. The present study reports the effects of recombinant buffalo OVGP1 that lacks post-translational modifications, and native Buffalo OVGP1 isolated from oviductal tissue, on frozen-thawed sperm functions and in vitro embryo ***： The proportion of viable sperms was greater（P 〈 0.05） in the recombinant OVGP1-treated group compared to the native OVGP1-treated group at 2 h, 3 h, and 4 h of incubation. The proportion of motile sperms at3 h and 4 h of incubation; and membrane-intact sperms at 4 h was greater（P 〈 0.05） in the native OVGP1-treated group compared to the control and recombinant OVGP1-treated groups. The proportion of capacitated and acrosome-reacted sperms was greater（P 〈 0.05） in the native OVGP1-treated group compared to the recombinant OVGP1 group at 4 h. The rates of cleavage of embryos and their development to the blastocyst stage were greater（P 〈 0.05） in the presence of either native or recombinant OVGP1 in comparison to control at 10 μg/mL concentration as compared to 5 or 20 μg/***： The study suggests that both native and recombinant OVGP1 impart a positive effect on various sperm features and in vitro embryo development. However, native OVGP1 was found to have a more pronounced effect in comparison to recombinant non-glycosylated OVGP1 on various sperm functions except viability. Hence,our current findings infer that glycosylation of OVGP1 might be essential in sustaining the sperm functions but not the in vitro embryo development.

Objective:To understand the level of progesterone (P4) in different quality of buffalo cumulus oocyte complexes (COCs) and further to evaluate the effect of exogenous P4 supplementation on maturation and subsequent developmental ability of poor quality brilliant cresyl blue (BCB-)***: Progesterone secreted by different quality of buffalo oocytes was estimated by enzyme linked immunosorbent assay and the concentration differences were translated into P4 doses to be incorporated in the maturation medium of BCB-ve COCs followed by expression analysis of genes involved in the cumulus expansion, extracellular matrix disintegration and progesterone receptor signalling. In addition, the study also evaluated the effect of exogenous P4 on sperm-cumulus interaction. Results: More than 10-fold up-regulated expression of progesterone receptor in P4 supplemented oocytes signified that P4 might be acting predominantly through this receptor. Also, exogenous P4 supplementation had significant effect on transcatheter arterial chemoembolization protease regulated by P4-progesterone receptor pathway which in turn had an important role in extracellular matrix disintegration. On the contrary, cumulus expansion genesHAS2,TNFAIP6,AREG were not altered upon P4 supplementation. Also, it was observed that P4 addition did facilitate passage of significantly more number of spermatozoa through P4 treated cumulus cells. Further, incorporation of different doses of P4 did not improve significantly the cleavage and blastocyst rates of BCB-ve ***: Different qualities of buffalo COCs secrete substantially diverse levels of P4, and its supplementation has a role in oocyte maturation via modulation of cumulus characteristics but perhaps not fertilization.

Community Based Seed Production-A Sustainable Seed Production Model for Subsistence of Bhutanese Maize Farmers

Bhutanese maize farmers grow different open-pollinated traditional varieties and improved high yielding varieties recommended by the national maize program. All most every maize farmer uses the recycled seeds from their farms for planting in the next season. Farmers traditional or informal seed system was found to be poorly organized and unscientific that has resulted in the deterioration and contamination of seed quality. Bhutanese maize farmers living in remote areas are highly constrained by the inconsistent supply and poor access to good quality maize seed. The formal public seed sectors lack adequate resources to produce and supply good quality seeds. This called for an urgency to identify and adapt an innovative and a sustainable seed production approach to service the subsistence needs of Bhutanese farmers in a cross-pollinated crop, like maize. The community based seed production （CBSP） approach was introduced and evaluated under the Bhutanese maize production system as an alternative farmer based seed production model. A total of seven farmers CBSP groups were formed and promoted as a smallholder seed enterprise. The technical skills of these CBSP group members on maize seed production were improved through practical demonstrations and trainings. The CBSP groups were supplied with high quality source seed of new maize varieties by the national maize program. For long term, the sustainability of CBSP groups were linked to the formal seed sector in the country for marketing the seeds. The CBSP approach has proven to be a successful small scale maize seed production model under highland maize production ecosystem that has improved the production and supply of good quality maize seeds. The Bhutanese experiences with CBSP in maize, its merits for small holder Bhutanese maize farmers and lessons learnt from the CBSP model are discussed in this paper.

Background: Diabetes Mellitus results in several serious complications and among them foot problem is one of the commonly ignored complications. This study is an attempt to find out the level of patient’s awareness and practice in Nepalese context. Objective: The aim of this study is to determine the knowledge and practice of foot care among patients with diabetes in Nepal. Methods: This is a six-month long cross-sectional study done in the General Practice Clinic of Tribhuvan University Teaching Hospital. Diabetic patients were given structure questionnaires on knowledge, foot care practices, and demographic information. Results: Illiteracy was found be significantly associated with poor knowledge and practice of foot care. Those who have no education are more likely to have poor knowledge and foot care practice. The odds of having poor knowledge is 6.414 (3.075, 13.379) in those who have no education/primary education compared to those who have education and the odds of poor foot care practice is 4.5180 (2.077, 8.411) in those who have no education/primary education compared to those who have education with significance of Conclusion: This study has highlighted the need of appropriate education intervention for the patients with diabetes who have no education background to prevent from foot complications. Health education materials such as graphics, photos are needed to make them comprehensible for the illiterate patients.
