Introduction:Alzheimer 's disease(AD) is a common neurodegenerative disorder and the primary cause of dementia. Considerable evidence supports the “amyloid hypothesis,” stating that the pathogenesis of AD is primarily caused by the deposition of amyloid-β(Aβ), which drives tau phosphorylation, neuroinflammation, and neurodegeneration in the brain. The amyloid hypothesis is strengthened by the significant and moderate benefit of lecanemab, a humanized antibody through an anti-amyloid mechanism,showing slowed clinical decline(van Dyck et al.,2023). The recent positive results of anti-amyloid trials have brought back focus on the amyloid hypothesis through biochemical, genetic, and pharmacological approaches(Zhang, 2023).

Effect of 5.25% Sodium Hypochlorite on Shear Bond Strength of Orthodontic Brackets Bonded with OBA-MCP

Introduction: Bracket debonding is a frequent issue that clinicians encounter, leading to increased chair time, lost revenue, and material usage. In addition to patient compliance with their diet recommendations, the preparation and conditioning of teeth for bonding significantly influence bond strength and consequently impact orthodontic treatment success and efficiency. Because of OBA-MCP’s (orthodontic bonding adhesive with modified calcium phosphate) decreased shear bond strength (SBS), the purpose of this study was to evaluate the effects of conditioning with 5.25% sodium hypochlorite (NaOCl) before etching in the bonding protocol. Materials and Methods: 90 extracted teeth were divided into 3 groups to be bonded with orthodontic brackets with different bonding protocols: 1) Transbond XT with regular bonding protocol (etch + prime + adhesive);2) OBA-MCP with regular bonding protocol;and 3) OBA-MCP with NaOCl prior to acid etching in the regular bonding protocol. SBS (in Newtons) were measured using an MTS universal testing machine with a custom jig to apply a vertical force onto the bracket and ARI (adhesive remnant index) scores were recorded for each sample after de-bond to rate the amount of adhesive remaining. Results: The addition of NaOCl to the bonding protocol statistically significantly increased the SBS of OBA-MCP to comparable levels to Transbond XT. The ARI scores showed that when NaOCl was added, more adhesive remained. Conclusion: The addition of NaOCl to the bonding protocol can increase the SBS of adhesives with historically weaker bond strengths. However, the increased amount of adhesive remaining and the increased time spent during bonding must be considered. Further testing can be done in vivo to demonstrate the practicality of this new procedure.

Co3O4possesses both direct and indirect oxidation effects and is considered as a promising catalyst for the oxidation of 5-hydroxymethylfurfural（HMF）.However,the enrichment and activation effects of Co3O4on OH-and HMF are weak,which limits its further *** defect engineering can regulate the electronic structure,optimize the adsorption of intermediates,and improve the catalytic activity by breaking the symmetry of the material,which is rarely involved in the upgrading of *** this work,we prepare Co3O4with metal defects and load the precious metal platinum at the defect sites（PtVco）.The results of in-situ characterizatio ns,electrochemical measurements,and theoretical calculations indicate that the reduction of Co-Co coordination number and the formation of Pt-Co bond induce the decrease of electron filling in the antibonding orbitals of Co *** resulting upward shift of the d-band center of Co combined with the characteristic adsorption of Pt species synergically enhances the enrichment and activation of organic molecules and OH species,thus exhibiting excellent HMF oxidation activity(including a lower onset potential（1.14 V） and 19 times higher current density than pure Co3O4at 1.35 V).In summary,this work explores the adsorption enhancement mechanism of metal defect sites modified by precious metal in detail,provides a new option for improving the HMF oxidation activity of cobalt-based materials,broadens the application field of metal defect based materials,and gives an innovative guidance for the functional utilization of metal defect sites in biomass conversion.

The present work aims to stabilize the room temperature allotropic transition of ammonium nitrate(AN)particles utilizing a microencapsulation technique,which involves solvent/non-solvent in which nitrocellulose(NC)has been employed as a coating *** SEM micrographs revealed distinct features of both pure AN and NC,contrasting with the irregular granular surface topography of the coated AN particles,demonstrating the adherence of NC on the AN *** analysis via infrared spectroscopy(IR)demonstrated a successful association of AN and NC,with slight shifts observed in IR bands indicating interfacial *** X-ray Diffraction(PXRD)analysis further elucidated the structural changes induced by the coating process,revealing that the NC coating altered the crystallization pattern of its pure *** analysis demonstrates distinct profiles for pure and coated AN,for which the coated sample exhibits a temperature increase and an enthalpy decrease of the room temperature allotropic transition by 6℃,and 36%,***,the presence of NC coating alters the intermolecular forces within the composite system,leading to a reduction in melting enthalpy of coated AN by~39%compared to pure *** thermal decomposition analysis shows a two-step thermolysis process for coated AN,with a significant increase in the released heat by about 78%accompanied by an increase in the activation barrier of NC and AN thermolysis,demonstrating a stabilized reactivity of the AN-NC *** findings highlight the synergistic effect of NC coating on AN particles,which contributed to a structural and reactive stabilization of both AN and NC,proving the potential application of NC-coated AN as a strategically advantageous oxidizer in composite solid propellant formulations.

Mannan-decorated STINg-activating vaccine carrier for spatial coordinative stimulating antigen-specific immune responses

In recent years, the use of nanotechnologies to improve immunotherapy efficiency has attracted increasing interest in preventive and therapeutic cancer vaccine design. However, current nanocarriers are restricted by difficulties in the systematic spatial coordinative transport of antigens, which greatly hampers the immune response efficacy of nanovaccines. Herein, we designed a mannan-decorated stimulator of the interferon genes (STINg)-activating vaccine carrier for spatial coordinative stimulation of antigen-specific immune responses and elicitation of robust antitumor immunity. Mannan-decoration as the shell could significantly enhance the lymph node draining ability of the nanovaccines, especially in CD8+ dendritic cells (DCs). Azole molecule end-capped polylactic acid-polyethylenimine (PLA-PEI-4BImi) with innate stimulating activity was applied as the inner core for coordinating antigen-presenting cell activation and antigen cross-presentation. In the in vivo therapy study, single usage of this nanovaccine could achieve a 93% tumor suppression rate in the B16-OVA tumor model, which is superior to the commercialized aluminum adjuvant. This study demonstrates that a rational design of vaccine carriers for solving spatial transmission issues could greatly improve cancer vaccine efficiencies. © 2023

Regulation of cytokine gene expression by orosomucoid in neonatal swine adipose tissue

Background： Porcine adipose tissue expresses orosomucoid（ORM1） mRNA, a protein with anti-inflammatory and immunomodulatory properties. Previous research has demonstrated that porcine ORM1 can reduce insulin stimulated glucose metabolism in porcine adipose tissue in vitro. The present study was designed to examine the preweaning ontogeny of ORM1 m RNA abundance in porcine subcutaneous adipose and to determine if ORM1 can regulate m RNA abundance of inflammatory cytokines that contribute to insulin resistance in primary cultures derived from neonatal porcine subcutaneous adipose tissue. Cultures were differentiated in vitro and subsequently the adipocyte containing cultures were incubated for 24 h with 0–5000 ng porcine ORM1/m L medium. Cultures were then harvested, total RNA extracted for use in reverse transcription and the m RNA abundance of cytokine m RNA quantified by real-time ***： ORM1 m RNA abundance within neonatal adipose tissue does not change from d 1 to d 21 of age and is a very small fraction relative to liver m RNA abundance. The ORM1 m RNA level in porcine adipocytes and stromalvascular cells are similar（P 〉 0.05）. Treatment with ORM1 did not affect TNFα（tumor necrosis factor α） m RNA level（P 〉 0.05）, while interleukin 6（IL6） m RNA abundance was reduced 32 % at 1,000 ng ORM1/m L（P 〈 0.01）. However,TNFα protein content in the cell culture media was reduced by ORM1 treatment（5,000 ng/m L, P 〈 0.05）, whereas ORM1 had no detectable effect on the media content of IL6（P 〉 0.05）. The reduction of macrophage migration inhibitory factor（MIF） m RNA abundance by ORM1 was dose dependent（P 〈 0.01）. Monocyte chemotactic protein（MCP） m RNA level was reduced 27 % by 1,000 ng ORM1/m L（P 〈 0.05）.Conclusions： The data suggest that ORM1 has limited effects TNFα, IL6, MIF or MCP expression at the concentrations tested. Laser cut hole matrices in novel armour plate steel for appliqué battlefield vehicle protection

During this research, experimental rolled homogeneous armour steel was cast, annealed and laser cut to form an appliqué plate. This Martensitic–Bainitic microstructure steel grade was used to test a novel means of engineering lightweight armour. It was determined that a laser cutting speed of 1200 mm/min produced optimum hole formations with limited distortion. The array of holes acts as a double-edged solution, in that they provide weight saving of 45%, providing a protective advantage and increasing the surface area. Data collected were used to generate laser cut-edge hole projections in order to identify the optimum cutting speed, edge condition, cost and deformation performance. These parameters resulted in the generation of a surface, with less stress raising features. This can result in a distribution of stress across the wider surface. Provided that appropriate process parameters are used to generate laser cut edges, then the hardness properties of the surface can be controlled. This is due to compressive residual stresses produced in the near edge region as a result of metallurgical transformations. This way the traverse cutting speed parameter can be adjusted to alter critical surface characteristics and microstructural properties in close proximity to the cut-edge. A relationship was identified between the width of the laser HAZ and the hardness of the cut edge. It is the thickness of the HAZ that is affected by the laser process parameters which can be manipulated with adjusting the traverse cutting speed.

Expanding the phenotypic plasticity paradigm to broader views of trait space and ecological function

Foundational concepts of trait spaces, including phenotypic plasticity and function of traits, should be expanded and better integrated with ecological theory. This article addresses two areas where plasticity theory can become further integrated with ecological, evolutionary, and developmental thinking. First is the idea that not only trait means within environments and plasticity of trait means across environments is optimized by selection, but that the entire shape of phenotype distributions such as variance or skew should be optimized within and across environments. In order for trait distribution shape to evolve into adaptations, there must be a genetic basis for and selection upon variation in distribution shapes and their plasticities. I present published and new data demonstrating genetic control and selection for higher moments of phenotype distributions; though, plasticity in these values has not yet been tested. genetic control of phenotype distribution moments is shown for Neurospora crassa ascospore size and shape. Selection on trait distribution moments is shown for Eurosta solidaginis gall size. Second, there is a tradition in modeling plasti- city as an adaptive strategy that pits it as an alternative to ecological specialization or generaliza- tion. However, these strategies need not be considered alternatives. Rather, with environmental fluctuation within generations plasticity may produce additive or non-additive intermediate （gener- alist） phenotypes, or something new altogether. I present published and new data on the snail Physa virgata and fish gambusia affinis that show plasticity produces partly intermediate （general- ist） and partly unique phenotypic elements in mixed and fluctuating environments. Plasticity can thus be viewed in the context of a broader trait space and as having broader ecological roles than currently is conceived.

For much of the world＇s population, food is the major source of exposure to *** to this non-essential metalloid at relatively low levels may be linked to a wide range of adverse health effects. Thus, evaluating foods as sources of exposure to arsenic is important in assessing risk and developing strategies that protect public health. Although most emphasis has been placed on inorganic arsenic as human carcinogen and toxicant, an array of arsenic-containing species are found in plants and animals used as foods. Here,we 2evaluate the contribution of complex organic arsenicals（arsenosugars, arsenolipids,and trimethylarsonium compounds） that are found in foods and consider their origins,metabolism, and potential toxicity. Commonalities in the metabolism of arsenosugars and arsenolipids lead to the production of di-methylated arsenicals which are known to exert many toxic effects. Evaluating foods as sources of exposure to these complex organic arsenicals and understanding the formation of reactive metabolites may be critical in assessing their contribution to aggregate exposure to arsenic.
