Nitric oxide(NO)has emerged as an important signal molecule in plants,having myriad roles in plant *** addition,NO also orchestrates both biotic and abiotic stress responses,during which intensive cellular metabolic reprogramming *** to these responses is the location of NO biosynthetic and scavenging pathways in diverse cellular compartments,enabling plants to effectively organize signal transduction *** regulates plant metabolism and,in turn,metabolic pathways reciprocally regu-late NO accumulation and ***,these diverse cellular processes are inextricably *** re-view addresses the numerous redox pathways,located in the various subcellular compartments that pro-duce NO,in addition to the mechanisms underpinning NO *** focus on how this molecular dance is integrated into the metabolic state of the *** this context,a reciprocal relationship be-tween NO accumulation and metabolite production is often *** also showcase cellular pathways,including those associated with nitrate reduction,that provide evidence for this integration of NO function and ***,we discuss the potential importance of the biochemical reactions governing NO levels in determining plant responses to a changing environment.

PDX1.1-dependent biosynthesis of vitamin b6 protects roots from ammonium-induced oxidative stress

Despite serving as a major inorganic nitrogen source for plants,ammonium causes toxicity at elevated con-centrations,inhibiting root elongation early *** previous studies have shown that ammonium-inhibited root development relates to ammonium uptake and formation of reactive oxygen species(rOS)in roots,it remains unclear about the mechanisms underlying the repression of root growth and how plants cope with this inhibitory effect of *** this study,we demonstrate that ammonium-induced apo-plastic acidification co-localizes with Fe precipitation and hydrogen peroxide(H_(2)O_(2))accumulation along the stele of the elongation and differentiation zone in root tips,indicating Fe-dependent rOS *** screening ammonium sensitivity in T-DNA insertion lines of ammonium-responsive genes,we identified PDX1.1,which is upregulated by ammonium in the root stele and whose product catalyzes de novo biosyn-thesis of vitamin *** growth of pdx1.1 mutants is hypersensitive to ammonium,while chemical complementation or overexpression of PDX1.1 restores root *** salvage strategy requires non-phosphorylated forms of vitamin b6 that are able to quench rOS and rescue root growth from ammo-nium ***,these results suggest that PDX1.1-mediated synthesis of non-phosphorylated b6 vitamers acts as a primary strategy to protect roots from ammonium-dependent rOS formation.

Hepatic failure caused by plasma cell infiltration in multiple myeloma

Although plasma cell infiltration is not rare in autopsy of patients with multiple myeloma （MM）, it is very rarely detected in living patients. This is because MM rarely causes significant liver dysfunction that requires further evaluation. A 49-year-old man presented with acute renal failure and was diagnosed with kappa light chain MM stageⅡb. Thalidomide and dexamethasone were initiated. The patient developed a continuous increase in bilirubin that led to severe cholestasis. A liver biopsy revealed plasma cell infiltration. He then rapidly progressed to liver failure and died. Treatment options are limited in MM with significant liver dysfunction. Despite new drug therapies in MM, those patients with rapidly progressive liver failure appear to have a dismal outcome.

Occult hepatitis b virus(HbV)infection(ObI)is a challenging pathobiological and clinical issue that has been widely debated for several *** definition,ObI is characterized by the persistence of HbV DNA in the liver tissue(and in some cases also in the serum)in the absence of circulating HbV surface antigen(HbsAg).Many epidemiological and molecular studies have indicated that ObI is an important risk factor for hepatocellular carcinoma(HCC)*** may exert direct pro-oncogenic effects through the activation of the same oncogenic mechanisms that are activated in the course of an HbsAg-positive ***,in ObI as in HbV-positive infection,HbV DNA can persist in the hepatocytes both integrated into the host genome as well as free episome,and may maintain the capacity to produce proteins-mainly X protein and truncated preS-S protein-provided with potential transforming ***,ObI may indirectly favor HCC *** has been shown that the persistence of very low viral replicative activity during ObI may induce mild liver necro-inflammation continuing for life,and substantial clinical evidence indicates that ObI canaccelerate the progression of liver disease towards cirrhosis that is considered the most important risk factor for HCC development.

The estimated world prevalence of hepatitis b virus(HbV)infection is 316 *** infection was identified in 1963 and nowadays is a major cause of cirrhosis and hepatocellular carcinoma(HCC)despite universal vaccination programs,and effective antiviral ***-term administration of nucleos(t)ide analogues(NA)has been the treatment of choice for chronic hepatitis b during the last *** NA has shown a good safety profile and high efficacy in controlling viral replication,improving histology,and decreasing the HCC incidence,decompensation,and ***,the low probability of HbV surface antigen seroclearance made necessary an indefinite *** knowledge,in recent years,about the different phases of the viral cycle,and the new insights into the role of the immune system have yielded an increase in new therapeutic ***,several clinical trials evaluating combinations of new drugs with different mechanisms of action are ongoing with promising *** integrative literature review aims to assess the knowledge and major advances from the past of hepatitis b,the present of NA treatment and withdrawal,and the future perspectives with combined molecules to achieve a functional cure.

Methane from enteric fermentation is the gas with the greatest environmental impact emitted by ***(Lv)addition to feedstocks could be a strategy to mitigate rumen methane emissions via decreasing the population of methanogenic archaea(MA).Thus,this paper provides the first overview of the effects of Lv supplementation,focusing on the inhibition of methane production,rumen microbiota,and ruminal *** indicated that Lv treatment had a strong anti-methanogenic effect on pure strains of ***,there are uncertainties from in vitro rumen fermentation trials with complex substrates and rumen ***-state fermentation(SSF)has emerged as a cost-effective option to produce *** this way,SSF of agricultural residues as an Lv-carrier supplement in sheep and goats demonstrated a consistent decrease in ruminal methane *** experimental evidence for in vitro conditions showed that Lv did not affect the volatile fatty acids(VFA).However,in vivo experiments demonstrated that the production of VFA was *** did not negatively affect the digestibility of dry matter during in vitro and in vivo methods,and there is even evidence that it can induce an increase in *** the rumen microbiota,populations of MA were reduced,and no differences were detected in alpha and beta diversity associated with Lv ***,some changes in the relative abundance of the microbiota were *** studies are recommended on:(i)Lv biodegradation products and stability,as well as its adsorption onto the solid matter in the rumen,to gain more insight on the“available”or effective Lv concentration;and(ii)to determine whether the effect of Lv on ruminal fermentation also depends on the feed composition and different ruminants.
